Abstract
A mixing bag (10) for use in bioprocessing in which a fluid (F) is received and agitated using an internal fluid-agitating element (18) driven by an external motive device (24). The bag (10) may include an integral sparger (100) and sensor receiver (200).
Claims (35)
1. A bioprocessing vessel intended for receiving a fluid, comprising:
a bag capable of receiving and holding the fluid, the bag having a rigid, integral sparger including means for forming bubbles from a gas supplied to the fluid in the bag.
a bag capable of receiving and holding the fluid, the bag having a rigid, integral sparger including means for forming bubbles from a gas supplied to the fluid in the bag.
2. The vessel according to claim 1, wherein the rigid, integral sparger is welded to the bag to form a fluid-impervious seal.
3. The vessel according to claim 2, wherein the means for forming bubbles comprises a perforated film attached to the rigid material.
4. The vessel according to claim 2, wherein the means for forming bubbles comprises a gas permeable film attached to the rigid material.
5. The vessel according to claim 2, wherein the means for forming bubbles comprises a perforated piece of material attached to a face of the sparger along the interior of the bag.
6. The vessel according to claims 1, further including a magnetic fluid-agitating element.
7. The vessel according to claim 6, wherein the rigid material further includes a first receiver for receiving and holding the magnetic fluid-agitating element at a home location when positioned in the vessel.
8. The vessel according to claim 6, wherein the receiver is an upstanding post.
9. The vessel according to claim 1, wherein the sparger comprises a rigid material, and the means for forming bubbles comprises an outer wall of a post connected to the rigid material, wherein the post provides a centering function for an annular magnetic fluid-agitating element positioned in the bag.
10. The vessel according to claim 9, wherein the post includes an oversized head for retaining the fluid-agitating element.
11. The vessel according to claim 9, wherein the rigid material includes a recess for receiving a motive device for the fluid-agitating element.
12. A bioprocessing vessel intended for receiving a fluid, comprising:
a bag capable of receiving and holding the fluid, the bag having a rigid receiver facing an interior; and an at least translucent tube having a closed end for insertion through the receiver and in communication with the interior.

13. The vessel of claim 12, further including a magnetic fluid-agitating element.
14. The vessel of claim 12, wherein the rigid receiver is welded to the bag to form a fluid-impervious seal.
15. The vessel of claim 12, further including a bushing for holding the receiver in the tube.
16. The vessel of claim 12, further including means for sealing the tube in the receiver.
17. The vessel of claim 12, further including a fluorescent sensor element externally attached to the closed end of the tube.
18. The vessel of claim 17, in combination with a fiber optic cable for transmitting light to the sensor element and receiving the light reflected therefrom.
19. A bioprocessing vessel intended for receiving a fluid, comprising:
a bag capable of receiving and holding the fluid, the bag having an integral sparger including a perforated or gas permeable material for forming bubbles from a gas supplied to the fluid; and a magnetic element for agitating the fluid.

20. The vessel of claim 19, further including a rigid receiver for receiving the magnetic element in the bag.
21 21. The vessel of claim 20, wherein the receiver is a post and the material comprises an outer wall of the post.
22. The vessel of claim 20, wherein the sparger comprises a rigid portion welded to the bag and the material comprises a porous or permeable film attached to the face of the portion.
23. A bioprocessing vessel intended for receiving a fluid, comprising:
a bag capable of receiving and holding the fluid, the bag having a rigid receiver facing an interior;
an at least translucent tube having a closed end for insertion through the receiver and in communication with the interior; and a magnetic fluid-agitating element.

24. The vessel of claim 23, further including a rigid receiver for receiving the magnetic element in the bag.
25. A method of manufacturing a bioreactor, comprising:
providing a bag capable of receiving and holding a fluid; and attaching to the bag an integral sparger including means for forming bubbles from a gas supplied to the fluid in the bag.
providing a bag capable of receiving and holding a fluid; and attaching to the bag an integral sparger including means for forming bubbles from a gas supplied to the fluid in the bag.
26. The method of claim 25, wherein the sparger comprises a rigid, disk-shaped piece of material, and the attaching step comprises welding the bag to the periphery of the rigid material to form a fluid-impervious seal.
27. The method of claim 25, further including the step of placing a magnetic impeller in the bag.
28. A method of bioprocessing a fluid, comprising:
providing a bag capable of receiving and holding a fluid;
attaching to the bag a rigid, integral sparger including means for forming bubbles in the fluid when present;
supplying gas to the sparger; and disposing of the bag, including the sparger.
providing a bag capable of receiving and holding a fluid;
attaching to the bag a rigid, integral sparger including means for forming bubbles in the fluid when present;
supplying gas to the sparger; and disposing of the bag, including the sparger.
29. The method of claim 28, further including the step of recovering the fluid from the bag.
30. The method of claim 28, wherein the step of attaching comprises welding the bag to the rigid material to create a fluid-impervious seal.
31. The method of claim 28, further including the step of agitating the fluid using an externally driven magnetic fluid-agitating element.
32. A method of arranging a sensor for a fluid bioprocessing operation, comprising:
providing a bag capable of receiving and holding a fluid;
attaching to the bag a rigid receiver; and inserting a transparent tube having a closed end in the receiver, whereby the sensor may be positioned on or in the tube for interfacing with the fluid.

35. The method of claim 32, further comprising externally attaching a fluorescent sensor element to a closed end of the tube.
