本发明公开了一种人VEGF2多肽和编码该VEGF2多肽的DNA(RNA),以及一种通过重组技术生产该多肽的方法和针对该多肽的抗体和拮抗剂。本发明还公开了利用这种多肽刺激伤口愈合和血管组织修复的方法。本发明也公开了使用所述的拮抗剂抑制肿瘤生长、炎症以及治疗糖尿病性视网膜病、风湿性关节炎和银屑病的方法。本发明还公开了用于检测来自宿主的样品中VEGF2编码序列中的突变以及VEGF2蛋白浓度的改变的诊断方法。
Description
本发明是Rosen,C.等人于1994年3月8日在美国专利商标局申请的申请号为08/207,550的申请的继续部分申请。
本发明涉及新鉴定的多核苷酸,该多核苷酸编码的多肽,该多核苷酸和多肽的用途以及该多核苷酸和多肽的生产。本发明的多肽已鉴定为血管内皮生长因子家族的一个成员,更具体地,本发明的多肽为血管内皮生长因子2,在下文中有时称作“VEGF2”。本发明也涉及抑制该多肽的作用。
新血管的形成或称血管生成对于胚胎发育、后续的生长以及组织修复是非常重要的,然而血管生成也是某些病理状态如瘤形成(例如肿瘤和神经胶质瘤)的重要部分,异常血管生成与其它一些疾病相关,如炎症、风湿性关节炎、银屑病和糖尿病性视网膜病(Folkman,J.and Klagsbrun,M.,科学235:442-447(1987))。
酸性和碱性成纤维细胞生长因子分子均是内皮细胞和其它细胞的促分裂原。尽管血管趋向肽(angiotropin)和血管生成素的功能尚不清楚,但是它们可以诱导血管生成(Folkman,J.,1993,癌症药物,153-170页,Lea and Febiger出版社)。血管内皮细胞的一种高度选择性促分裂原是血管内皮生长因子或称VEGF(Ferrara,N.等,内分泌综述13:19-32(1992)),其也称为血管通透因子(VPF),血管内皮生长因子是分泌的血管生成促分裂原,其靶细胞特异性似乎限于血管内皮细胞。
已鉴别了小鼠VEGF基因并分析了其在胚胎发生中的表达模式。在邻近透明内皮的上皮细胞中,例如在脉络膜丛和肾小球中观察到VEGF的持续表达。该资料与VEGF是内皮细胞生长和分化的多功能调节物的作用一致(Breier,G.等,发育114:521-532(1992))。
VEGF在结构上与血小板衍生生长因子(PDGF)和胎盘生长因子(PLGF)的α和β链相关,PDGF是间质细胞的促分裂原,PLGF是内皮细胞的促分裂原。这三个蛋白属于同一家族并共享一保守基序,在这些蛋白中负责形成二硫键的8个半胱氨酸严格保守。已鉴别了VEGF、PLGF和PDGF的可变剪接mRNA,这些不同的剪接产物的生物学活性和受体结合特异性不同。VEGF和PDGF以同二聚体或异二聚体起作用并结合受体,这些受体在二聚化后激发内在的酪氨酸激酶活性。
由于可变剪接,VEGF具有4种不同形式,分别含121、165、189和206个氨基酸,VEGF121和VEGF165是可溶的并能促进血管生成,而VEGF189和VEGF206与细胞表面的含有肝素的蛋白聚糖结合。VEGF的时空表达已证明与血管的生理性增殖相关(Gajdusek,C.M.and Carbon,S.J.,细胞生理学139:570-579(1989);McNeil,P.L.,Muthukrishnan,L.,Warder,E.,D’Amore,P.A.,细胞生物学杂志109:811-822(1989))。其高亲和性结合位点只位于组织切片中的内皮细胞(Jakeman,LB.等,临床研究89:244-253(1989))。所述的因子可以从垂体细胞和几种肿瘤细胞系中分离并已用于某些人神经胶质瘤(Plate,K.H.,自然359:845-848(1992))。令人感兴趣的是VEGF121或VEGF165的表达赋予中国仓鼠卵巢细胞以在裸鼠中形成肿瘤的能力(Ferrara,N.等,临床研究杂志91:160-170(1993))。已证明用抗VEGF单克隆抗体对VEGF的抑制可以在免疫缺陷的小鼠中抑制肿瘤生长(Kim,K.J.,自然362:841-844(1993))。另外,VEGF受体的一种显性阴性突变体已证明能抑制小鼠中的成神经细胞瘤的生长。
已发现血管通透因子负责甚至在损伤停止后仍持续的对于细胞质蛋白的微血管高通透性,这是正常伤口愈合的一个特征,提示VPF是伤口愈合中的一个重要因子,Brown,L.F.等,实验医学杂志176:1375-9(1992)。
VEGF在血管化组织(例如肺、心脏、胎盘和实体肿瘤)中的表达很高并与血管生成在时间和空间上相关。还证明VEGF能在体内诱导血管生成。由于血管生成对于正常组织特别是血管组织的修复很重要,所以VEGF已被提议用于促进血管组织修复(例如在动脉粥样硬化中)。
美国专利5,073,492(1991年12月17日授予Chen等)公开了一种在合适环境中协同增强内皮细胞生长的方法,该方法包括向该环境中加入VEGF,效应子以及血清衍生因子。另外,还通过聚合酶链反应技术制备了血管内皮生长因子C亚基DNA,该DNA编码一种能以异二聚体或同二聚体存在的蛋白质,该蛋白质是哺乳动物血管内皮细胞促分裂原,因此能用于促进血管发育和修复,如欧洲专利申请92302750.2(1992年9月30日公开)中所公开的一样。
根据与人VEGF的氨基酸序列同源性,本发明的多肽被推断鉴定为一种新的血管内皮生长因子。
根据本发明的一个方面,提供了一种新的成熟多肽及其具有生物学活性并在诊断学或治疗学上有用的片段,类似物与衍生物。本发明的多肽是人源的。
根据本发明的另一个方面,提供了编码本发明的多肽的分离的核酸分子,该核酸分子包括mRNA,DNA,cDNA,基因组DNA,及其具有生物学活性并在诊断学或治疗学上有用的片段、类似物和衍生物。
根据本发明的另一个方面,提供了通过重组技术生产这种多肽的方法,该方法包括在促进所述蛋白表达及所述蛋白的随后回收的条件下培养含有编码本发明的多肽的核酸序列的重组原核和/或真核宿主细胞。
根据本发明的再一个方面,提供了一种利用该多肽或编码该多肽的多核苷酸用于治疗目的的方法,例如刺激血管生成、伤口愈合和促进血管组织修复。
根据本发明的又一个方面,提供了抗这种多肽的抗体。
根据本发明的另一个方面,提供了抗这种多肽的拮抗剂,其可用于抑制这种多肽的作用,例如抑制肿瘤生长,治疗糖尿病性视网膜病、炎症、风湿性关节炎和银屑病。
根据本发明的另一个方面,提供了含有足够长的核酸分子的核酸探针,该探针可以特异地杂交到本发明的核酸序列上。
根据本发明的又一个方面,提供了用于诊断与本发明的核酸序列中的突变以及由这种核酸序列编码的蛋白质有关的疾病或疾病易感性的方法。
根据本发明的又一个方面,提供了一种将这种多肽或编码这种多肽的多核苷酸用于与科学研究、DNA合成以及DNA载体的生产有关的体外目的的方法。
根据本文的教导,本发明的这些以及其它方面对于本领域的技术人员而言应该是显而易见的。
下面的附图仅仅意味着是对本发明的具体实施方案的说明,而并不意味着以任何的方式限制本发明。
图1示出了本发明的多肽的cDNA序列以及相应推导的氨基酸序列,使用了标准的单字母氨基酸缩写。测序用373自动DNA测序仪(Applied Biosystems,Inc.)进行,测序精度预计大于97%。
图2示出本发明的多肽和人PDGF/VEGF家族其它成员之间的氨基酸序列同源性。加框的区域指示保守的序列以及8个保守半胱氨酸的位置。

图7示出用RP-300柱(0.21×3cm,Applied Biosystems,Inc.)对纯化的VEGF2进行反相HPLC分析,柱用0.1%三氟乙酸(溶剂A)平衡并用0-60%溶剂B(由含0.07%TFA的乙腈组成)的7.5min梯度洗脱蛋白质。蛋白洗脱液用在215nm(红线)和280nm(蓝线)处的吸收值而监控。溶剂B的百分比由绿线示出。
图8示出与碱性成纤维细胞生长因子相比,部分纯化的VEGF2蛋白对血管内皮细胞的生长的作用。
图9示出纯化的VEGF2蛋白对血管内皮细胞的生长的作用。
术语“基因”是指参与产生一个多肽链的DNA区段;它包括在编码区之前与之后的区域(前导区与尾区)以及在各个编码区段(外显子)之间的间插序列(内含子)。
根据本发明的一个方面,提供了分离的核酸分子(多核苷酸),其编码具有图1的推导的氨基酸序列(SEQ ID NO:2)的成熟多肽,或者编码由1995年5月24日保藏的ATCC保藏号为97161的克隆的cDNA所编码的成熟多肽,或者编码具有比图1所示少的氨基酸残基的多肽。
编码本发明的多肽的多核苷酸可以从早期人胚胎(8周至9周)破骨细胞瘤、成人心脏或几种乳腺癌细胞系中获得,本发明的多核苷酸在衍生自9周的早期人胚胎衍生的cDNA文库中发现。它在结构上与VEGF/PDGF家族相关。VEGF2含有一个编码419个氨基酸残基的蛋白质的开放读框,其中大约前23个氨基酸残基是推断的前导序列,从而成熟蛋白包括396个氨基酸,该蛋白质显示与人血管内皮生长因子具有最高的氨基酸序列同源性,有30%相同性,依次是PDGFα(23%)和PDGFβ(22%)。
特别重要的是在这一家族的所有4个成员中所有8个半胱氨酸均是保守的(见图2的加框区域),另外PDGF/VEGF家族的特征,PXCVXXXRCXGCCN(SEQ ID NO:3)在VEGF2中是保守的(见图2)。
本发明的VEGF2多肽意在包括全长多肽和编码任何前导序列和全长多肽的活性片段的多核苷酸序列,活性片段意在包括任何具有比SEQ ID NO.2和图2所示的完整419个氨基酸的全长氨基酸序列短的氨基酸序列但是仍含有在图2中显示保守的8个半胱氨酸残基的全长氨基酸序列的部分,这种片段仍含有VEGF2活性。
在正常组织中至少存在两种可变剪接的VEGF2 mRNA序列,图3示出了分别对应于全长和截短译本的两种VEGF2 mRNA序列的大小,泳道5示出两条带,指示存在编码本发明的VEGF2多肽的可变剪接的mRNA。
本发明的多核苷酸可以是RNA形式或是DNA形式,其中DNA包括cDNA,基因组DNA和合成DNA,该DNA可以是双链或者是单链的,如果是单链则可以是编码链或非编码(反义)链。编码成熟多肽的编码序列可以与显示在图1中的编码序列或者保藏克隆的编码序列相同,或者该序列可以是一个不同的编码序列,但由于遗传密码的丰余性或简并性它编码与图1的DNA或保藏的cDNA编码相同的成熟多肽。
编码图1的成熟多肽或者编码由保藏的cDNA编码的成熟多肽的多核苷酸可以包括:仅编码成熟多肽的编码序列;成熟多肽的编码序列(以及任选的附加的编码序列)与非编码序列,如内含子或者成熟多肽编码序列的5’和/或3’非编码序列。
因此,术语“编码多肽的多核苷酸”包括:仅包括该多肽编码序列的多核苷酸以及包括附加的编码和/或非编码序列的多核苷酸。
本发明进一步涉及如上所述的多核苷酸的变异体,其编码具有图1的推导的氨基酸序列的多肽的片段、类似物与衍生物或者由保藏克隆的cDNA编码的多肽的片段、类似物与衍生物。该多核苷酸的变异体可以是该多核苷酸的一种天然发生的等位基因变异体,或者是该多核苷酸的一种非天然发生的变异体。
因此,本发明包括编码如图1所示的相同成熟多肽或由保藏克隆的cDNA编码的相同成熟多肽的多核苷酸及其变异体,其中该变异体编码图1的多肽或者由保藏克隆的cDNA编码的多肽的片段、衍生物或者类似物。该核苷酸变异体包括缺失变异体,取代变异体以及添加或插入变异体。
如上文所示,所述的多核苷酸可以具有是图1所示的编码序列或者保藏克隆的编码序列的天然发生的等位基因变异体的编码序列。正如在本领域中已知的,一种等位基因变异体是一个多核苷酸序列的可变形式,它可以具有一个或多个核苷酸的取代,缺失或添加,但它基本上不改变所编码多肽的功能。
本发明的多核苷酸也可具有在同一读框内与标记序列融合的编码序列,所述的标记序列使得能纯化本发明的多肽。在细菌宿主的情况下,标记序列可以是由pQE-9载体提供的六组氨酸标记物以使与标记融合的成熟多肽纯化,或者,例如当使用哺乳动物宿主如COS-7细胞时,标记序列可以是血凝素(HA)标记物,HA标记物对应于从流感血凝素蛋白衍生的一个表位(Wilson,I.等,细胞37:767(1984))。
术语“基因”是指参与产生一个多肽链的DNA区段;它包括在编码区之前与之后的区域(前导区与尾区)以及在各个编码区段(外显子)之间的间插序列(内含子)。
本发明的全长基因的片段可以用作一个cDNA文库的杂交探针,以便于分离全长cDNA并且分离与该基因具有高的序列相似性或相似的生物学活性的其它cDNA。这种类型的探针优选具有至少30个碱基并且可以含有例如50或更多个碱基。该探针也可以用于鉴定对应于一个全长转录物的cDNA克隆以及含有完整基因的一个基因组克隆或多个基因组克隆,其中完整基因包括调节区与启动子区,外显子和内含子。一个筛选的实例包括通过使用已知的DNA序列合成寡核苷酸探针以分离出基因的编码区。具有互补于本发明基因序列的序列的标记寡核苷酸可以用于筛选人cDNA、基因组DNA或mRNA的文库,从而测定与该探针杂交的文库成员。
本发明进一步涉及与上述序列杂交的多核苷酸,条件是序列间存在至少70%、优选至少90%且更优选至少95%的相同性。本发明特别涉及在严格条件下与上述多核苷酸杂交的多核苷酸。文中所使用的术语“严格条件”是指只有序列间存在至少95%且优选至少97%的相同性时才发生杂交。在一个优选的实施方案中,与上述多核苷酸杂交的多核苷酸编码的多肽基本上保留了与由图1的cDNAs(SEQID NO:1)或保藏的cDNA(s)所编码的成熟多肽所具有的同样的生物学功能或活性。
另一方面,多核苷酸可以含有至少20个碱基,优选30个碱基并且更优选至少50个碱基与本发明的多核苷酸杂交且与之具有如上所述相同性,并且可以保留或不保留活性。例如这样的多核苷酸可以用作SEQ ID NO:1的多核苷酸的探针,例如用于回收多核苷酸或用作诊断探针或PCR引物。
因此,本发明涉及与编码SEQ ID NO:2的多肽的多核苷酸及其片段具有至少70%的相同性、优选至少90%的相同性且更优选至少95%的相同性的多核苷酸,以及该多核苷酸所编码的多肽,其中所述的片段具有至少30个碱基并且优选至少50个碱基。
本文所涉及的保藏物将按照关于国际承认的用于专利程序的微生物保藏的布达佩斯条约进行保存。这些保藏物仅为了本领域技术人员的方便而提供,不是对按35 U.S.C.§112要求保藏物的许可。包含在保藏材料中的多核苷酸的序列及其所编码的多肽的氨基酸序列在本文中作为参考而附入,并且在与本文序列描述有冲突的任意的一种情况下起决定作用。制造、使用或销售该保藏材料需要许可证,并且此处不颁发这样的许可证。
本发明进一步涉及一种具有图1的推导的氨基酸序列或者具有由保藏的cDNA所编码的氨基酸序列的多肽以及该多肽的片段、类似物和衍生物。
当涉及图1的多肽或由保藏的cDNA编码的多肽时,术语“片段”、“衍生物”与“类似物”是指保留了图2所示的VEGF蛋白的保守基序并基本上保留了该多肽的同样的生物学功能或活性的多肽。
本发明的多肽可以是重组多肽,天然多肽或合成多肽,优选重组多肽。
图1的多肽或由保藏的cDNA编码的多肽的片段、衍生物或类似物可以是:(i)一种多肽,其中一个或多个氨基酸残基被一个保守的或非保守的氨基酸残基(优选一个保守的氨基酸残基)所取代并且被取代的氨基酸残基可以是或可以不是由该遗传密码所编码,或者(ii)一种多肽,其中一个或多个氨基酸残基包含一个取代基,或者(iii)一种多肽,其中成熟多肽与另一种化合物,诸如一种提高该多肽的半寿期的化合物(例如聚乙二醇)融合,或者(iv)一种多肽,其中成熟多肽与附加的氨基酸融合,或者(v)一种多肽,其包括比SEQ ID NO.2所示少的氨基酸残基并保留保守基序而且仍保持VEGF家族多肽的活性特征。从本文的教导出发,这样的片段、衍生物与类似物视为是在本领域的技术人员所熟知的范围内的。
本发明的多肽与多核苷酸优选以一种分离的形式而提供,并且优选将其纯化成均一物质。
术语“分离的”是指从其原始环境(例如如果该物质是天然存在的,即指天然环境)中分离出该物质。例如,一种存在于活体动物中的天然存在的多核苷酸或多肽是未分离的,但是从天然系统中一些或所有的共存物质里分离出的同样的多核苷酸或DNA或多肽是分离的。这样的多核苷酸可以是一个载体的一部分和/或这样的多核苷酸或多肽可以是一种组合物的一部分,并且如果该载体或组合物并非是其天然环境的一部分,则其仍然是分离的。
本发明的多肽包括SEQ ID NO:2的多肽(特别是成熟多肽)以及与SEQ ID NO:2的多肽具有至少70%的相似性(优选至少70%的相同性)的多肽、更优选与SEQ ID NO:2的多肽具有至少90%的相似性(更优选至少90%的相同性)的多肽、并且特别优选与SEQ ID NO:2的多肽具有至少95%的相似性(特别优选至少90%的相同性)的多肽,本发明的多肽也包括该多肽的部分,其中该多肽的这些部分通常含有至少30个氨基酸并且更优选至少50个氨基酸。
正如在本领域中已知的,两种多肽之间的“相似性”是通过把一个多肽的氨基酸序列及其保守氨基酸取代与第二个多肽的序列进行比较而确定的。
通过肽合成,本发明多肽的片段或部分可以用于生产相应的全长的多肽;因此,该片段可以用作生产全长多肽的中间体。本发明的多核苷酸的片段或部分可以用于合成本发明的全长的多核苷酸。
本发明也涉及含有本发明的多核苷酸的载体,用本发明的载体基因工程化的宿主细胞以及通过重组技术生产本发明的多肽。
宿主细胞可以是用本发明的载体进行基因工程化的(转导或转化或转染),其中载体例如可以是克隆载体或表达载体,载体例如可以呈质粒、病毒颗粒、噬菌体等的形式。基因工程化的宿主细胞可以在经改良以适于激活启动子、筛选转化体或扩增VEGF2基因的常规营养培养基上培养。培养条件(诸如温度、pH及其它)是为了表达而选择的宿主细胞以前所采用的条件,并且对本领域的普通技术人员来讲是显而易见的。
通过重组技术,本发明的多核苷酸可以用于生产多肽。因此,例如该多核苷酸序列可以包含在表达多肽的多种表达载体中的任意一种中,具体而言如载体或质粒中。这样的载体包括染色体、非染色体及合成DNA序列,例如:SV40衍生物;细菌质粒;噬菌体DNA;杆状病毒;酵母质粒;质粒与噬菌体DNA结合所得到的载体、病毒DNA(诸如痘苗病毒,腺病毒,禽痘病毒与假狂犬病)。但是,只要任意一种其它的载体或质粒在宿主中可以复制并且存活便可以使用它们。
可通过许多种方法把合适的DNA序列插入到载体中。一般而言,通过本领域已知的方法可以把DNA序列插入到一个合适的限制性内切酶位点上。该方法及其它方法被视为是在本领域的技术人员所熟知的范围之内。
表达载体中的DNA序列可操作地与一个合适的表达调控序列(启动子)连接以指导mRNA的合成。作为上述启动子有代表性的实例可以提到的有:LTR或SV40启动子,大肠杆菌lac或trp,噬菌体λPL启动子以及其它已知调控原核细胞或真核细胞或其病毒中基因表达的启动子。表达载体也含有一个用于翻译起始的核糖体结合位点与一个转录终止子。载体也可以包括用于扩增表达的合适序列。
此外,表达载体优选含有一种为转化宿主细胞的筛选而提供一种表型性状的基因,诸如用于真核细胞培养物的二氢叶酸还原酶或新霉素抗性,或者诸如在大肠杆菌中的四环素或氨苄青霉素抗性。
含有如上所述的合适的DNA序列以及一个合适的启动子或调控序列的载体可以用于转化一种合适的宿主从而允许该宿主表达蛋白。
作为合适宿主有代表性的实例可以提到的有:细菌细胞,诸如大肠杆菌,链霉菌属,鼠伤寒沙门氏杆菌;真菌细胞,如酵母;昆虫细胞,诸如果蝇S2与苜蓿银纹夜蛾Sf9;动物细胞,如CHO,COS或Bowes黑素瘤;腺病毒;植物细胞等等。根据本文的教导,选择合适的宿主被认为是在本领域的技术人员所熟知的范围之内。
更具体地讲,本发明也包括含有一个或多个如上广义所述的序列的重组构建体。构建体包括将本发明的序列以正向或逆向插入其中的载体,如质粒或病毒载体。在这一实施方案一个优选方面,构建体进一步包括可操作地与序列连接的调节序列,例如启动子。许多合适的载体与启动子是本领域的技术人员所熟知的并且可以通过商业途径得到。下面的载体以举例的方式提供:细菌载体:pQE70,pQE60,pQE-9(Qiagen),pBS,pD10,phagescript,psiX174,pBluescript SK,pBSKS,pNH8A,pNH16a,pNH18A,pNH46A(Stratagene);ptrc99a,pKK223-3,pKK233-3,pDR540,pRIT5(Pharmacia);真核载体:pWLNEO,pSV2CAT,pOG44,pXT 1,pSG(Stratagene),pSVK3,pBPV,pMSG,pSVL(Pharmacia)。但是,只要任意一种其它的质粒或载体在该宿主中可以复制并存活便可以使用它们。
采用具有选择标记的CAT(氯霉素转移酶)载体或其它载体,可以从任意一种所需要的基因中筛选出启动子区域。两个合适的载体为pKK232-8与pCM7。特别指出的细菌启动子包括lacI,lacZ,T3,T7,gpt,λPR,PL和trp。真核启动子包括CMV立即早期,HSV胸苷激酶,早期与晚期SV40,来自逆转录病毒的LTRs和小鼠金属硫蛋白-I。选择合适的载体与启动子在本领域普通的技术水平上是可以完成的。
在更进一步的实施方案中,本发明涉及含有上述构建体的宿主细胞。宿主细胞可以是高等真核细胞(如哺乳动物细胞),或是低等真核细胞(如酵母细胞),或者宿主细胞可以是原核细胞(如细菌细胞)。通过磷酸钙转染、DEAE一葡聚糖介导的转染或电穿孔可以实现该构建体向宿主细胞的引入(Davies,L.,Dibner,M.,Battey,I.,分子生物学基本方法,(1986))。
宿主细胞中的构建体可以以常规的方式使用从而生产由该重组序列所编码的基因产物。另一方面,本发明的多肽也可以通过常规的肽合成仪合成生产。
在合适的启动子的控制下,成熟蛋白可以在哺乳动物细胞、酵母、细菌或其它细胞中表达。采用从本发明的DNA构建体得到的RNAs,也可通过无细胞翻译系统生产该蛋白。用于原核与真核宿主的合适的克隆与表达载体在Sambrook等人的《分子克隆实验手册)》,第二版(纽约冷泉港,1989)中进行了描述,其公开内容此处引用为参考。
通过向载体中插入增强子序列可提高通过高等真核生物对编码本发明多肽的DNA的转录。增强子为DNA的顺式作用元件,一般大约从10至300bp,它作用于启动子以提高其转录。它的例子包含有位于复制起点晚期一侧的SV40增强子(bp100至270),巨细胞病毒早期启动子增强子,位于复制起点晚期一侧的多瘤增强子与腺病毒增强子。
一般而言,重组表达载体包括复制起点与允许宿主细胞转化的选择标记(例如大肠杆菌的氨苄青霉素抗性基因和啤酒酵母TRP1基因)以及从一个高度表达的基因中得到的指导下游结构序列的转录的启动子。这些启动子可以从编码如3-磷酸甘油酸激酶(PGK)这样的糖酵解酶、α因子、酸性磷酸酶、或热休克蛋白等的操纵子中得到。异源结构序列以合适的方式与翻译起始和终止序列、以及优选地一个能够指导被翻译的蛋白分泌至周质空间或胞外培养基中的前导序列进行装配。异源序列可以任选地编码一种融合蛋白,该蛋白含有一个N-末端鉴定肽,其中的鉴定肽赋予了表达的重组产物所需要的特征,例如稳定性或易于纯化的特性。
通过将编码一种所需蛋白的结构DNA序列与合适的翻译起始与终止信号以可操纵的阅读方式和一个功能启动子一起插入,可以构建对细菌的使用而言有用的表达载体。该载体将包括一个或多个表型选择标记和一个复制起点,以确保维持该载体并且如果需要的话可以保证在宿主中进行扩增。用于转化的合适的原核宿主包括大肠杆菌、枯草芽孢杆菌、鼠伤寒沙门氏杆菌以及属于假单胞杆菌属、链霉菌属和葡萄球菌属中的许多种,当然其它种的生物也可以采用。
作为代表性但非限制性的例子,对细菌的使用而言有用的表达载体可以包括选择标记和来自含有所熟知的克隆载体pBR322(ATCC37017)的遗传因子的商购质粒的细菌复制起点。这些商购载体包括如pKK223-3(Phamacia精细化学公司,Uppsala,瑞典)和GEM1(Promega Biotec,Madison,WI,美国)。这些pBR322“骨架”部分与一种合适的启动子和待表达的结构序列相结合。
进行合适的宿主株系的转化并且该宿主株系生长至合适的细胞密度后,通过合适的方法(例如温度变动或化学诱导)诱导所选择的启动子并且再将细胞培养一段时间。
细胞通常经离心收获,通过物理或化学方法进行破碎,并且保留得到的粗提物以便进一步的纯化。
在蛋白表达中所使用的微生物细胞可以通过任意一种方便的方法进行破碎,该方法包括冷冻-融化循环,超声处理,机械破碎,或者使用细胞裂解剂,这种方法是本领域熟练技术人员公知的。
各种哺乳动物细胞的培养系统也可以用于表达重组蛋白。哺乳动物表达系统的实例包括由Gluzman(细胞,23:175(1981))所述的猴肾成纤维细胞的COS-7细胞系以及可以表达相容载体的其它细胞系,例如C127,3T3,CHO,HeLa与BHK细胞系。哺乳动物表达载体包括复制起点、合适的启动子和增强子、以及任何必需的核糖体结合位点、聚腺苷酸化位点、剪接供体和受体位点、转录终止序列和5’侧翼非转录序列。从SV40病毒基因组中得到的DNA序列,例如SV40起点,早期启动子,增强子,剪接与聚腺苷酸化位点可以用于提供所需要的非转录遗传因子。
本发明的多肽可以通过迄今为止所使用的方法从重组的细胞培养物中回收并纯化,该方法包括硫酸铵或乙醇沉淀,酸提取,阴离子或阳离子交换层析,磷酸纤维素层析,疏水作用层析,亲和层析,羟基磷灰石层析与凝集素层析。如果需要的话,在完成该成熟蛋白的构型中可以采用蛋白重折叠步骤。最后,可以采用高效液相层析(HPLC)用于最终的纯化步骤。
本发明的多肽可以是一种天然纯化的产物,或者是一种化学合成方法的产物,或者是从原核或真核宿主(例如通过细菌,酵母,高等植物,昆虫和哺乳动物细胞培养)经重组技术制备的。根据在重组生产方法中所使用的宿主,本发明的多肽可以是糖基化或者可以是非糖基化的。该多肽也可以包含一个起始的甲硫氨酸残基。
如图8和9所示,SEQ ID NO.2的VEGF2多肽(减去开始的46个氨基酸)是血管内皮细胞潜在的促分裂原并且刺激其生长和增殖。针对编码这一多肽的VEGF2核酸序列进行的Northern印迹分析(在这一分析中用32P-VEGF2探查来自几种人组织的20μg RNA)的结果显示这一蛋白在心脏和肺中活跃表达,这是促分裂原活性的进一步的证据。
因此,VEGF2多肽可以用于促进血管生成,例如刺激进行冠状动脉搭桥手术的移植组织的生长,VEGF2也可以用于促进伤口愈合,特别是对于再血管化损害的组织,或在局部缺血过程中以及希望新的毛细血管生成的地方刺激旁路血流。VEGF2可以用来治疗深度伤口如真皮溃疡,包括压伤、静脉溃疡和糖尿病性溃疡。另外,当使用皮肤移植物或皮瓣来修复深度烧伤和损伤时,VEGF2可以用来治疗深度烧伤和损伤。VEGF2还可用于整形外科,例如用于修复外伤和手术切口的伤口。
除此之外,VEGF2可以用于诱导损伤骨骼、牙周或韧带组织的生长,VEGF2还可用于再生由于疾病和外伤而损害的牙齿支持组织如牙釉质和牙周韧带。
由于血管生成对于保持伤口清洁及不发生感染是重要的,因此VEGF2可以用于外科手术以及后来的切口修复中,其也可以用来治疗处于感染高危状态的腹膜伤口。
VEGF2可以在血管移植手术中用于促进内皮化,在使用移植的或合成的材料的血管移植物时,可以将VEGF2施加到移植物的表面或者连接处以促进血管内皮细胞的生长。VEGF2还可用于修复由于心肌梗塞所致的心肌组织损伤,VEGF2还可用于修复局部缺血后的心血管系统,VEGF2还可用于治疗冠状动脉疾病以及周围和CNS血管疾病导致的损伤的血管组织。
VEGF2还可用于涂覆待移植进体内的人工修补物或天然器官以最大程度地降低对移植材料的排斥并刺激移植材料的血管化。
VEGF2例如还可用于在动脉粥样硬化和随后的气囊血管移植中损伤的血管组织所需的血管组织修复中。
VEGF2核酸序列和VEGF2多肽还可以用于各种体外目的,如科学研究、DNA合成和DNA载体的制备,以及生产治疗人类疾病的诊断剂和治疗剂。例如,VEGF2可以用于体外培养血管内皮细胞,其是以10pg/ml-10ng/ml的浓度加入条件培养基中。
全长VEGF2基因的片段可以用作一cDNA文库的杂交探针以分离与该基因有高度序列相似性或相似生物学活性的其它基因。这种类型的探针通常至少为50bp,当然它们可以具有更多个碱基。探针还可用于鉴别对应于全长转录物的cDNA克隆以及含有完整的VEGF2基因(包括调节和启动子区、外显子和内含子)的基因组克隆。一个筛选例子包括用已知的DNA序列合成寡核苷酸探针以分离VEGF2基因的编码区。具有互补于本发明基因的序列的标记寡核苷酸可用于筛选人cDNA、基因组DNA或mRNA文库以确定探针与哪些文库成员杂交。
本发明提供了鉴定VEGF2受体的方法。编码受体的基因可以通过许多为本领域普通技术人员已知的方法进行鉴定,例如配体淘选与FACS分选(Coligan,等,免疫学通用方案,1(2),第五章,(1991))。优选采用表达克隆法,其中从对VEGF2有反应的细胞中制备聚腺苷酸化的RNA,并且把从该RNA产生的cDNA文库分割成多个集合体并且将其用于转染COS细胞或其它对VEGF2无反应的细胞。将生长于载玻片上的转染细胞与标记的VEGF2接触,VEGF2可通过许多方法,包括碘化作用或引入一个位点特异性蛋白激酶的识别位点来标记。固定与温育后,对玻片进行放射自显影分析。鉴定阳性集合体并且采用一种重复亚集合与再筛选方法来制备和再转染亚集合体,最终得到一种编码推定的受体的单一克隆。
作为一种用于受体鉴定的替代方法,标记的VEGF2可以和细胞膜或表达该受体分子的提取制剂光亲和连接。交联物质通过PAGE分析进行分辨并且在X-光胶片上曝光。可以切下含有VEGF2的标记复合物,分解成肽片段并且对之进行蛋白微量测序。从微量测序得到的氨基酸序列可以用来设计一套简并的用于筛选cDNA文库的寡核苷酸探针,从而鉴定编码该推定受体的基因。
本发明还设计一种筛选化合物以鉴定那些是VEGF2的激动剂或拮抗剂的物质的方法。这种方法的一个实例是利用VEGF2在comitogen Con A存在下能明显刺激人内皮细胞增殖的能力。取内皮细胞在96孔平底培养板(Costar,Cambridge,MA)中的补加Con-A(Calbiochem,La Jolla,CA)的反应混合物中培养,加入Con-A,本发明的多肽以及待筛选的化合物,在37℃温育后用1μCi3[H]胸苷(5Ci/mmol;1Ci=37BGq;NEN)脉冲培养物足够时间以掺入3[H]并收获至玻璃纤维滤膜(PhD;Cambridge Technology,Watertown,MA)上。用液闪计数仪(Beckman Instruments,Irvine,CA)确定三份重复培养物的平均3[H]胸苷掺入(cpm)。与不加化合物的对照测定相比显著的3[H]胸苷掺入指示对内皮细胞增殖的刺激。
为测定拮抗剂化合物,可以进行上述分析测定,在VEGF2存在下该化合物抑制3[H]胸苷掺入的能力指示该化合物是VEGF2的一种拮抗剂。或者,可以在适合竞争抑制分析的条件下将VEGF2和一潜在的拮抗剂化合物与膜结合VEGF2受体或重组受体合并来检测VEGF2拮抗剂。VEGF2可以标记如用放射性标记,从而与受体结合的VEGF2分子数可以用来确定潜在的拮抗剂的效果。
或者,可以在化合物存在或不存在下测定VEGF2和受体相互作用后已知的第二信使系统的反应并进行比较。这种第二信使系统包括但不限于cAMP鸟苷酸环化酶、离子通道或磷酸肌醇水解。在另一方法中,在化合物的存在下将表达哺乳动物细胞或表达VEGF2受体的膜制剂与标记的VEGF2保温,然后可以测定化合物增强或阻断这一相互作用的能力。
潜在的VEGF2拮抗剂包括抗体,或者在某些情况下为寡核苷酸,它们与多肽结合并有效地消除VEGF2功能。或者,一种潜在的拮抗剂可以是一种结合VEGF2受体的紧密相关的蛋白质,但是它们是多肽的失活形式并因此阻止VEGF2的作用。这些拮抗剂的例子包括VEGF2多肽的阴性显性突变体,例如异二聚体形式的VEGF2的一条链可以是显性的并可以经突变而不再保持生物学活性。阴性显性突变体的一个例子包括二聚体化VEGF2的截短译本,其能与另一二聚体相互作用形成野生型VEGF2,然而,得到的同二聚体是失活的,不能显示特征性VEGF活性。
另一种潜在的VEGF2拮抗剂为采用反义技术制备的反义构建体。通过三股螺旋形成或反义DNA或RNA,反义技术可以用于控制基因的表达,上述两种方法均是建立在一种多核苷酸结合到DNA或RNA的基础上的。例如,编码本发明的成熟多肽的多核苷酸序列的5’编码部分可以用于设计一种反义RNA寡核苷酸,其长度从大约10至40个碱基对。将一种DNA寡核苷酸设计成互补于涉及转录的基因区域(三股螺旋-参见Lee等,核酸研究,6:3073(1979);Cooney等,科学,241:456(1988);与Dervan等,科学,251:1360(1991)),从而阻止转录以及VEGF2的生产。反义RNA寡核苷酸在体内杂交到mRNA上并且阻止mRNA分子翻译成VEGF2多肽(反义-Okano,神经化学杂志,56:560(1991);作为基因表达的反义抑制剂的寡脱氧核苷酸,CRC出版社,Boca Raton,FL(1988))。还可以把上述寡核苷酸传递至细胞,从而该反义RNA或DNA可以在体内表达以抑制VEGF2的生产。
潜在的VEGF2拮抗剂还包括一些小分子,这些小分子可以结合并占据多肽的活性位点从而使催化位点接触不到底物以阻止正常生物学活性。小分子的实例包括但不限于小肽或类肽分子。
拮抗剂可以用于治疗固体肿瘤转移所需的有限血管生成。
发现编码VEGF2的mRNA在至少两种乳腺肿瘤细胞系中以中等水平表达,提示VEGF2多肽在恶性表型中的作用。神经胶质瘤也是瘤形成的一种类型,其可用本发明的拮抗剂治疗。
拮抗剂还可用于治疗由增加的血管通透性导致的炎症,除这些紊乱外,拮抗剂还可用于治疗糖尿病性视网膜病、风湿性关节炎和银屑病。
拮抗剂可以与一种如下所述的药物学可接受的载体一起用于组合物中。
VEGF2多肽和激动剂和拮抗剂可以结合一种合适的药物载体而使用,这种组合物包括治疗上有效量的多肽或激动剂或拮抗剂以及一种药用可接受的载体或赋形剂。这样的载体包括但不限于盐水,缓冲盐水,葡萄糖,水,甘油,乙醇及其混合物。配方应当适合于给药的方式。
本发明也提供了一种药物包装或试剂盒,它们包括一个或多个容器,该容器中装有一种或多种本发明的药物组合物的成分。与该容器相伴的可以是一则以管理药品或生物制品的生产、使用或销售的政府机构规定的形式存在的通知,该通知反映出生产、使用或销售机构对人体给药的批准。此外,该药物组合物可以与其它的治疗化合物结合使用。
该药物组合物可以按照常规的方式给药,如通过局部、静脉内、腹膜内、肌内、肿瘤内、皮下、鼻内或者真皮内的途径。该药物组合物以对治疗和/或预防具体症状有效的量进行给药。一般而言,该组合物以至少约10μg/kg体重的量进行给药,并且在大多数情况下该组合物的给药量将不超过约8mg/kg体重/天。在大多数情况下考虑到给药的途径、症状等,该剂量从每天约10μg/kg体重至约1mg/kg体重。
根据本发明,VEGF2多肽以及同为多肽的激动剂或拮抗剂也可以通过在体内表达所述多肽而使用,这通常称作“基因治疗”。
因此,例如可以采用编码多肽的多核苷酸(DNA或RNA)在体外工程化细胞如骨髓瘤细胞,随后将工程化的细胞提供给待采用该多肽治疗的患者。上述方法是本领域所熟知的。例如,可以通过本领域已知的方法、采用一种含有编码本发明多肽的RNA的逆转录病毒颗粒来工程化细胞。
类似地,为了在体内表达多肽,例如可以通过本领域已知的方法在体内工程化细胞。正如在本领域已知的,为了在体内工程化细胞并且在体内表达多肽,可以给予患者一种生产含有编码本发明多肽的RNA的逆转录病毒颗粒的生产细胞。从本发明的教导出发,通过上述方法给予本发明的一种多肽的这些或者其它的方法对本领域的普通技术人员是显而易见的。例如,除了逆转录病毒颗粒以外,用于工程化的细胞的表达载体还可以是如一种腺病毒,它可以在与一种合适的输送载体结合后用于在体内工程化细胞。
可以得到上述逆转录病毒质粒载体的逆转录病毒包括但不限于Moloney小鼠白血病病毒,脾坏死病毒,逆转录病毒诸如Rous肉瘤病毒,Harvey肉瘤病毒,禽白血病病毒,长臂猿白血病病毒,人免疫缺陷病毒,腺病毒,骨髓增生性肉瘤病毒与乳房肿瘤病毒。在一个具体的实施方案中,逆转录病毒质粒载体是从Moloney小鼠白血病病毒得到的。
载体包含一个或者多个启动子。可以采用的合适的启动子包括但不限于逆转录病毒LTR;SV40启动子;以及在Miller,等,生物技术,第7卷,第9期,第980-990页(1989)中所描述的人巨细胞病毒(CMV)启动子,或者任何一种其它的启动子(例如细胞启动子,如真核细胞启动子,包括但不限于组蛋白、pol III和β-肌动蛋白启动子)。其它可以采用的病毒启动子包括但不限于腺病毒启动子,胸苷激酶(TK)启动子及B19细小病毒启动子。根据本文中的教导,选择合适的启动子对于本领域普通技术人员来说是显而易见的。
编码本发明多肽的核酸序列在合适的启动子的控制下。可以采用的合适的启动子包括但不限于腺病毒启动子,如腺病毒主要晚期启动子;或者异源启动子,如巨细胞病毒(CMV)启动子;呼吸道合胞病毒(RSV)启动子;诱导型启动子,如MMT启动子、金属硫蛋白启动子;热休克启动子;白蛋白启动子;ApoAI启动子;人珠蛋白启动子;病毒的胸苷激酶启动子,如单纯疱疹胸苷激酶启动子;逆转录病毒LTRs(包括上述修饰的逆转录病毒LTRs);β-肌动蛋白启动子;以及人生长激素启动子。该启动子也可以是控制编码该多肽的基因的天然启动子。
采用逆转录病毒质粒载体来转导包装细胞系从而形成生产细胞系。可以被转导的包装细胞的实例包括但不限于在Miller,人类基因治疗,第1卷,第5-14页(1990)中所描述的PE501,PA317,φ-2,φ-AM,PA12,T19-14X,VT-19-17-H2,φCRE,φCRIP,GP+E-86,GP+envAm12以及DAN细胞系,其中该文献以其全文引入本文以供参考。载体可以通过本领域已知的任何一种方法来转导包装细胞,这些方法包括但不限于电穿孔,采用脂质体以及CaPO4沉淀。在另一种方法中,逆转录病毒质粒载体可以包埋在脂质体中,或者被偶联到脂类上,然后再将该载体引入宿主中。
生产细胞系产生含有编码该多肽的核酸序列的侵染性逆转录病毒载体颗粒,然后可以使用这些逆转录病毒载体颗粒或在体外或在体内转导真核细胞。被转导的真核细胞将表达编码该多肽的核酸序列。可以被转导的真核细胞包括但不限于胚干细胞,胚胎癌性细胞,以及造血干细胞,肝细胞,成纤维细胞,成肌细胞,角质形成细胞,内皮细胞和支气管上皮细胞。
本发明也涉及VEGF2基因作为一种诊断测定的一部分的用途从而检测与在VEGF2核酸序列中存在的突变相关的疾病或疾病易感性。
携带VEGF2基因中的突变的个体可以通过许多技术在DNA水平上进行检测。用于诊断的核酸可以从病人的细胞中得到,如来自血液、尿液、唾液、活组织检查与尸体剖检材料的细胞。基因组DNA可以直接用于检测或者在分析之前可以通过采用PCR(Saiki等,自然,324:163-166(1986))进行酶促扩增。RNA或cDNA也可以用于同样的目的。作为一个实例,互补于编码VEGF2的核酸的PCR引物可以用于测定和分析VEGF2突变。例如,与正常的基因型比较,通过扩增产物大小的变化可以检测缺失和插入。通过将扩增的DNA与放射性标记的VEGF2 RNA或者放射性标记的VEGF2反义DNA序列杂交便可以测定点突变。通过RNase A消化或通过解链温度的差异,完全配对序列可以同错配的双螺旋区别开来。
基于DNA序列差异的基因测试可以通过检测DNA片段在含有或者不含变性剂的凝胶中的电泳迁移率的变化来实现。小的序列缺失与插入可以通过高分辨率的凝胶电泳观测。不同序列的DNA片段可以在变性甲酰胺梯度凝胶上进行区分,其中根据DNA片段的特殊解链温度或部分解链温度、不同DNA片段的迁移率被阻滞在凝胶不同的位置上(参见例如,Myers等,科学,230:1242(1985))。
通过如RNase和S1保护的核酸酶保护分析或化学切割方法(例如,Cotton等,PANS,美国,85:4397-4401(1985))也可以揭示在特殊位置上的序列变化。
因此可以通过如杂交,RNase保护,化学切割,直接DNA测序或者使用限制性酶(例如,限制性片段长度多态性(RFLP))和基因组DNA的Southern印迹法这些方法来实现特异DNA序列的检测。
除了更常规的凝胶电泳和DNA测序外,突变也可以通过原位分析进行检测。
本发明也涉及一种用于检测在不同组织中VEGF2蛋白的变化水平的诊断分析,这是由于与正常对照组织样品相比该蛋白的过量表达可以检测疾病或疾病易感性的存在,例如异常细胞分化。用于检测从宿主得到的样品中的VEGF2蛋白含量的分析方法是本领域普通技术人员所熟知的,并且包括放射免疫测定、竞争结合测定、Western斑点印迹法、酶联免疫吸附测定和“夹心”测定。酶联免疫吸附测定(Coligan,等,免疫学通用方案,1(2),第六章,(1991))最初包括制备一种特异于VEGF2抗原的抗体,优选为一种单克隆抗体。此外制备一种抗该单克隆抗体的报道抗体,将该报道抗体结合到一个可检测的试剂上,如放射性、荧光性或在本实施例中为辣根过氧化物酶上。从宿主中取样并将其在一种结合样品中的蛋白的固相载体、如聚苯乙烯平皿上进行温育。然后通过用非特异性蛋白如牛血清白蛋白的温育来覆盖平皿上任意游离的蛋白结合位点。接下来,在平皿上温育该单克隆抗体,在此期间该单克隆抗体结合到已被附着在聚苯乙烯平皿上的任何VEGF2蛋白。用缓冲液洗去所有未结合的单克隆抗体。将连接到辣根过氧化物酶上的报道抗体放置在平皿上,使得该报道抗体结合到已和VEGF2结合的任何单克隆抗体上。然后洗去未被附着的报道抗体。随后把过氧化物酶的底物加入到平皿中,并且当与一条标准曲线进行对照时,在给定的时间段内形成的颜色量是对存在于给定体积的病人样品中的VEGF2蛋白的量的测量值。
可以采用一种竞争测定,其中将特异于VEGF2的抗体结合到固相支持物上,然后例如通过放射性标记本发明的多肽,将从宿主得到的样品通过该固相支持物,然后例如通过液闪计数检测的标记量可以与该样品中VEGF2量相互关联。
“夹心”测定类似于酶联免疫吸附测定。在“夹心”测定中,将VEGF2多肽通过固相支持物并结合到已附着在该固相支持物上的抗体上,然后将第二抗体结合到VEGF2上。随后将已标记并且特异于第二抗体的第三抗体引入固相支持物并结合到第二抗体上,之后可以定量测定其量。
本发明的序列对染色体鉴定也是有价值的。该序列特异地靶向位于单个的人染色体上的特定位置并能与之杂交。此外,现在需要鉴定染色体上的特定位点。目前,仅有少数几种以实际的序列数据(重复多态性)为基础的染色体标记试剂可以用于标记染色体的位置。本发明的DNA染色体作图是将这些序列和疾病相关基因相关联的重要的第一步。
简而言之,通过从cDNA制备PCR引物(优选15-25bp)便可以把序列定位到染色体上。采用3’未翻译区域的计算机分析可以快速地选择引物,其中引物不应跨越超过基因组DNA上的一个外显子,否则使得扩增方法复杂化。然后采用这些引物用于PCR筛选含有单个的人染色体的体细胞杂交体。只有那些含有与该引物对应的人基因的杂交体才会生产扩增片段。
体细胞杂交体的PCR作图是将一个特定的DNA定位于特定的染色体上的快速程序。根据本发明采用同样的寡核苷酸引物,用来自于特定染色体或者大基因组克隆集合体的一组片段、按照类似方式可以实现亚定位(sublocalization)。可以类似地用于对染色体作图的其它的作图策略包括原位杂交,用标记的经流式分选的染色体进行预筛选以及通过杂交进行预选,从而构建出染色体特异性的cDNA文库。
cDNA克隆与中期染色体涂片的荧光原位杂交(FISH)可以用来实现一步法准确染色体定位。该技术可以采用50或60个碱基长短的cDNA。关于该技术的综述参阅Verma等,人类染色体:基本技术手册,Pergamon出版社,纽约(1988)。
一旦一个序列已定位到一个准确的染色体位置,则染色体上该序列的物理位置可与遗传图谱数据相关联。这些数据例如可在V.McKusick,人类的孟德尔遗传中找到(可以通过Johns Hopkins大学Welch医学文库联机得到)。然后通过连锁分析(物理相邻基因的共遗传性)来鉴定基因与已定位到相同染色体区域上的疾病之间的关系。
接下来需要测定在受影响的和未受影响的个体之间cDNA或基因组序列中的差异。如果突变是在一些或所有的受影响个体中观察到的、但是又没有在任何一个正常个体中被观察到的话,那么该突变可能是疾病的病原体。
根据物理作图和遗传作图技术目前的分辨率,一个被准确定位到与疾病有关的染色体区域的cDNA可以是50-500个潜在的病原(causative)基因中的一种(其中假定有1兆碱基的作图分辨率且每20kb为一个基因)。
多肽、其片段或该多肽的其它衍生物或类似物、或者表达上述物质的细胞可以用作生产其抗体的免疫原。这些抗体可以是例如多克隆抗体或者单克隆抗体。本发明也包括嵌合,单链和人源化的抗体,以及Fab片段或Fab表达文库的产物。本领域已知的多种方法可以用于生产这些抗体和片段。
针对相应于本发明的一个序列的多肽而生成的抗体可以通过将该多肽直接注射入动物体内或者通过将该多肽向动物给药来得到,其中的动物优选非人类。然后,如此得到的抗体会结合到该多肽上。通过这种方式,即使是仅仅编码该多肽的一个片段的序列也可用于生成能结合整个天然多肽的抗体。然后,该抗体可以用于从表达该多肽的组织中分离这种多肽。为了制备单克隆抗体,可以采用任何一种提供了通过连续的细胞系培养生产抗体的技术。该实例包括杂交瘤技术(Kohler与Milstein,1975,自然,256:495-497),三体杂交瘤技术,人B-细胞杂交瘤技术(Kozbor等,1983,今日免疫学,4:72)以及生产人单克隆抗体的EBV-杂交瘤技术(Cole,等,1985,单克隆抗体与癌症治疗,Alan R.Liss,Inc.,pp.77-96)。
可以把所述的用于生产单链抗体的技术(美国专利4,946,778)进行修改从而生产出对本发明免疫原性的多肽产品有抗性的单链抗体。也可以使用转基因小鼠来表达对本发明免疫原性的多肽产品有抗性的人源化抗体。
本发明将参照下面的实施例进一步加以说明;但是,应当了解到本发明并不局限于这些实施例。除非另作声明的以外,所有的部分或数量均为重量单位。
为了有利于理解以下的实施例,现叙述一些经常出现的方法和/或术语。
“质粒”通过一个在前的小写p和/或跟随几个大写字母和/或数字加以命名。本文中的起始质粒或者可以通过商业途径得到或在不受限制的基础上可以通过公众得到,或者可以根据已公开的方法从可得到的质粒中构建出来。此外,对于与那些所述相当的质粒是本领域已知的并且对本领域普通技术人员是显而易见的。
DNA的“消化”是指用一种仅对DNA上的某些序列起作用的限制性酶催化裂解DNA。本文所采用的多种限制性酶可以通过商业途径得到,并且其反应条件、辅因子和其它的使用要求对本领域普通技术人员是已知的。为了分析的目的,通常把1μg的质粒或DNA片段与溶于约20μl缓冲溶液的约2个单位的酶一起使用。为了分离用于质粒构建的DNA片段,通常在一个更大的体积内用20至250个单位的酶消化5至50μg的DNA。针对特殊的限制性酶而言,合适的缓冲溶液和底物的量是由生产者规定的。通常采用在37℃下大约1小时的温育时间,但是该时间可以根据产品供应者的指示而变化。在消化后,该反应直接在聚丙烯酰胺凝胶上进行电泳以分离出所需的片段。采用由Goeddel,D.等,核酸研究,8:4057(1980)所描述的8%的聚丙烯酰胺凝胶进行被切片段的大小分离。
“寡核苷酸”或指一种单链的多脱氧核苷酸,或指可以通过化学合成的两条互补的多脱氧核苷酸链。这些合成的寡核苷酸不具有5’磷酸,因此当没有在一种激酶存在下以ATP添加一个磷酸时,该寡核苷酸将不会连接到另一个寡核苷酸上。合成的寡核苷酸将连接到未被去磷酸化的一个片段上。
“连接”是指在两个双链核酸片段之间形成磷酸二酯键的过程(Maniatis,T.,等,出处同上,p.146)。除非另行提供的以外,采用已知的缓冲液和条件、每0.5μg大约等摩尔量的待连接的DNA片段10个单位的T4DNA连接酶(“连接酶”)来实现连接。
除非另有说明,按Graham,F.和Van der Eb,A.,病毒学,52:456-457(1973)所述的方法进行转化。
实施例1
VEGF2在人组织和乳腺癌细胞系中的表达模式
进行Northern印迹分析以检测VEGF2基因在入组织和人乳腺癌细胞系中的表达水平。用RNAzolTMB系统(Biotecx Laboratories,Inc.)分离总细胞RNA样品。在1%琼脂糖凝胶上分离约10μg来自每种乳腺组织和特定的细胞系的总RNA,并印迹到尼龙滤膜上,(分子克隆,Sambrook Fritsch,and Maniatis,冷泉港出版社,1989)。根据Stratagene Cloning Systems,Inc.,Prime-It试剂盒用50ngDNA片段进行标记反应,标记的DNA用来自5Prime--3Prime,Inc.,Boulder,CO,USA的Select-G-50柱纯化。然后将滤膜与1,000,000cpm/ml的放射性标记的全长VEGF2基因在0.5MNaPO4和7%SDS中于65℃杂交过夜。用0.5×SSC,0.1%SDS在室温洗2次并且在60℃洗2次后将滤膜用增感屏在-70℃曝光过夜,在2个乳腺癌细胞系中观察到1.6Kd的信号。
实施例2
采用杆状病毒表达系统克隆和表达VEGF2
采用与基因的5’和3’序列对应的PCR寡核苷酸引物来扩增编码不带N-末端46个氨基酸的VEGF2蛋白的DNA序列,见ATCC#97161:
5’引物序列为5’TGT AAT ACG ACT CAC TAT AGG GAT CCCGCC ATG GAG GCC ACG GCT TAT GC 3’(SEQ ID NO:4),它含有一个BamHI限制性酶位点(黑体)和互补于VEGF25’序列(150-166位核苷酸)的17个核苷酸序列。
3’引物具有序列5’GATC TCT AGA TTA GCT CAT TTG TGGTCT 3’(SEQ ID NO:5),并且含有限制性内切核酸酶XbaI的切割位点以及互补于VEGF2的3’序列(包括终止密码子和终止密码子之前的15个核苷酸序列)的18个核苷酸。
采用一种可以通过商业途径得到的试剂盒(“Geneclean”,BIO101 Inc.,La Jolla,Ca.)从1%的琼脂糖凝胶中分离扩增序列。然后,该片段用内切核酸酶BamHI和XbaI进行消化并且在1%的琼脂糖凝胶上再次纯化。将这一片段在BamHI和XbaI位点与pAcGP67A杆状病毒转移载体(PHarmingen)连接,通过这一连接,VEGF2 cDNA克隆到杆状病毒gp67基因的信号序列的同一读框中,并位于载体中的信号序列的3’末端,这一载体命名为pAcGP67A-VEGF2。
为了将带有gp67基因的信号序列的VEGF2克隆到pRG1载体中用于表达,通过XhoI和XbaI限制性酶在位于VEGF2 cDNA上游的Xho限制性内切核酸酶位点和XbaI限制性内切核酸酶位点从pAcGP67A-VEGF2质粒上切下带有信号序列的VEGF2和一些上游序列,在1%琼脂糖凝胶上将这一片段从载体的其余部分分离出并用“Geneclean”试剂盒纯化,其被命名为F2。
通过杆状病毒表达系统(综述参阅Summers,M.D.与Smith,G.E.1987,杆状病毒载体与昆虫细胞培养步骤的方法手册,得克萨斯农业实验站会刊No.1555),利用载体PRG1(pVL941载体的改进)来表达VEGF2蛋白。该表达载体含有苜蓿银纹夜蛾核型多角体病毒(AcMNPV)的强多角体蛋白启动子,其后跟随了限制性内切核酸酶BamHI、SmaI、XbaI、BglII和Asp718的识别位点。限制性内切核酸酶XhoI的位点位于BamHI位点的上游,XhoI和BamHI之间的序列与PAcGp67A载体的对应部分相同。猴病毒(SV)40的聚腺苷酸化位点用于有效的聚腺苷酸化作用。为了容易地选择出重组病毒,将来自大肠杆菌的β-半乳糖苷酶基因以与多角体蛋白启动子同样的取向插入,其后面是多角体蛋白基因的聚腺苷酸化信号。为了实现共转染的野生型病毒DNA的细胞介导的同源重组,多角体蛋白序列两侧为病毒序列。许多其它的杆状病毒载体,如pAc373,pVL941和pAcIM1可以用于代替pRG1(Luckow,VA.与Summers,M.D.,病毒学,170:31-39)。
质粒以限制性酶XhoI和XbaI消化并且通过本领域已知的方法采用牛小肠磷酸酶使其去磷酸化,然后采用可以通过商业途径得到的试剂盒(“Geneclean”,BIO 101 Inc.,La Jolla,Ca.)从1%的琼脂糖凝胶中分离出DNA,该载体DNA命名为V2。
片段F2和去磷酸化的质粒V2通过T4 DNA连接酶进行连接。然后转化大肠杆菌HB101细胞并且采用限制性酶BamHI和XbaI来鉴定含有带VEGF2基因的质粒(pBac gp67-VEGF2)的细菌。克隆片段的序列通过DNA测序加以证实。
采用脂质体转染法(Felgner等,Proc.Natl.Acad.Sci.美国,84:7413-7417(1987))将5μg的质粒pBac gp67-VEGF2和1.0μg的一种可以通过商业途径得到的线性化杆状病毒(“BaculoGoldTM杆状病毒DNA”,Pharmingen,San Diego,CA.)进行共转染。
将1μg的BaculoGoldTM病毒DNA和5μg的质粒pBac gp67-VEGF2在一个含有50μl无血清Grace培养基(生命技术公司,Gaithersburg,MD)的无菌微量滴定板的孔中进行混合。之后加入10μl的Lipofectin和90μl的Grace培养基,混合并在室温下温育15分钟。然后将该转染混合物逐滴加入在35mm组织培养平板上接种的Sf9昆虫细胞(ATCC CRL 1711),其中平板上含有1毫升无血清的Grace培养基。来回摇动该平板以混合新添加的溶液。然后该平板于27℃下温育5小时。5小时后,从平板中除去转染溶液并且加入1ml补充有10%的胎牛血清的Grace昆虫培养基。将该平板放回至一个温育箱中并且于27℃下持续培养4天。
4天后收集上清液,并且与Summers和Smith所述(出处同上)类似地进行噬斑测定。作为一种改进,采用一种含有“Blue Gal”(生命技术公司,Gaithersburg,MD)的琼脂糖凝胶,它可以使染上蓝色的噬斑易于分离。(“噬斑测定”的详细描述也可以在用于昆虫细胞培养和杆状病毒学的使用者指南的第9-10页中找到,该指南由生命技术公司,Gaithersburg分发)。
在连续稀释4天后将病毒加入细胞中,并且用Eppendorf移液管的尖端挑取染上蓝色的噬斑。然后将含有该重组病毒的琼脂重新悬浮在一个含有200μl Grace培养基的Eppendorf管中。通过短时间的离心除去琼脂并且采用含有该重组杆状病毒的上清液去感染在35mm平皿上接种的Sf9细胞。4天后收集这些培养平皿中的上清液,然后于4℃下进行储藏。
将Sf9细胞在补充有10%热灭活的FBS的Grace培养基上进行培养。在感染复数(MOI)为1的条件下,采用重组杆状病毒V-gp67-VEGF2感染细胞。6小时后除去该培养基并且代之以不含蛋氨酸和半胱氨酸的SF900 II培养基(生命技术公司,Gaithersburg)。42小时后加入5μCi的35S-蛋氨酸和5μCi的35S半胱氨酸(Amersham)。在离心收获细胞前将细胞进一步温育16小时,并且通过SDS-PAGE和放射性自显影观察被标记的蛋白。
在还原和非还原条件下通过SDS-PAGE分析得自Sf9细胞的培养基和细胞质的蛋白质。见图4。将培养基对50mM MES,pH5.8透析,透析后获得沉淀物并重悬于100mM柠檬酸钠pH5.0中,用SDS-PAGE再次分析重悬的沉淀物并用考马斯亮蓝染色。见图5。
将培养基上清在50mM MES,pH5.8中以1∶10稀释并以1ml/分钟的流速加到SP-650M柱(1.0×6.6cm,Toyopearl)中。以200、300和500mM NaCl梯度洗脱蛋白质,用在500mMNaCl处的洗脱液获得VEGF2蛋白。在还原剂β-巯基乙醇的存在或不存在下通过SDS-PAGE分析洗脱液并用考马斯亮蓝染色。见图6。
实施例3
重组VEGF2在COS细胞中的表达
表达质粒VEGF2-HA衍生自载体pcDNAI/Amp(Initrogen),其含有:1)SV40复制起点,2)氨苄青霉素抗性基因,3)E.coli复制起点,4)CMV启动子,后接多接头区、SV40内含子和多腺苷酸化位点。编码完整VEGF2前体和在框内融合到其3’末端的HA标记的DNA片段被克隆至载体的多接头区,从而重组蛋白质在CMV启动子控制下表达。HA标记对应于衍生自前述流感血凝集素蛋白的表位(I.Wilson,H.Niman,R.Heighten,A Cherenson,M.Connolly,andR.Lerner,1984,Cell 37,767)。HA标记与靶蛋白质融合使得用识别HA表位的抗体很容易检测重组蛋白质。
下面阐述质粒构建策略:
使用两种引物由PCR构建编码VEGF2的DNA序列,ATTC#97161,5’引物(CGC GGA TCC ATG ACT GTA CTC TAC CCA)(SEQ ID NO:6)含有一BamHI位点,后接18个起自起始密码子的VEGF2编码序列的核苷酸;3’序列为(CGC TCT AGA TCAAGC GTA GTC TGG GAC GTC GTA TGG GTA CTC GAG GCT CATTTG TGG TCT 3’)(SEQ ID NO:7)含有互补于XbaI位点、HA标记、XhoI位点及VEGF2编码序列的最后15个核苷酸(不包括终止密码子)的序列。因此PCR产物含有一BamHI位点,编码序列,后接一XhoI限制性位点和框内融合的HA标记,HA标记后的转译终止密码子及XbaI位点。将PCR扩增的DNA片段和载体,pcDNAI/Amp,用BamHI和XabI限制性酶消化并连接。用连接混合物转化E.coli菌体SURE(得自Strategene Cloning Systems,11099North Torrey Pines Road,La Jolla,CA 92037),在氨苄青霉素培养平板上培养转化的培养物并筛选抗性克隆。从转化子中分离出质粒DNA并用限制性分析检测正确片段的存在。为表达重组VEGF2,将COS细胞通过DEAE-DEXTRAN方法用表达载体转染(J.Sambrook,E.Fritsch,T.Manictis,Molecular Cloning:A LaboratoryManual,Cold Spring Laboratory Press,(1987)。用放射标记和免疫沉淀法检测VEGF2-HA蛋白的表达(E.Harlw,D.Lane,Antibodies:ALaboratory Manual,Cold Spring Harbor Laboratory Press,(1988)。转染二天后,将细胞用35S-半胱氨酸标记8小时。然后收集培养物培养基,用去污剂(RIPA缓冲液(150mM氯化钠,1%NP-40,0.1%SDS,1%NP-40,0.5%DOC,50mMTris,pH7.5)。(Wilson,I.et al.,Id.37:767(1984))将细胞裂解,将细胞裂解物和培养基用HA特异性单克隆抗体沉淀。将沉淀的蛋白质在15%SDS-PAGE凝胶上分析。
实施例4
部分纯化的VEGF2蛋白对血管内皮细胞的生长的作用
在第1天,将人脐静脉内皮细胞(HUVEC)以2-5×104细胞/35mm培养皿的密度接种在含4%胎牛血清(FBS)、16单位/ml肝素和50单位/ml内皮细胞生长添加剂(ECGS,Biotechnique,Inc.)的M199培养基中。在第2天,用含10%FBS、8单位/ml肝素的M199替换前述培养基。以所示浓度加入SEQ ID NO.2的VEGF2蛋白(减去开始的45个氨基酸残基)(VEGF)和碱性FGF(bFGF)。在第4、6天替换培养基,在第8天用Coulter计数器测定细胞数(见图8)。
实施例5
纯化的VEGF2蛋白对血管内皮细胞的生长的作用
在第1天,将人脐静脉内皮细胞(HUVEC)以2-5×104细胞/35mm培养皿的密度接种在含4%胎牛血清(FBS)、16单位/ml肝素和50单位/ml内皮细胞生长添加剂(ECGS,Biotechnique,Inc.)的M199培养基中。在第2天,用含10%FBS、8单位/ml肝素的M199替换前述培养基。在此时向培养基中加入SEQ ID NO.2所示的纯化的VEGF2蛋白(减去开始的45个氨基酸残基)。在第4、6天用新鲜培养基和添加剂替换培养基,在第8天用Coulter计数器测定细胞数(见图9)。
实施例6
通过基因治疗的表达
成纤维细胞是通过皮肤活组织检查从一个研究对象中得到的。将得到的组织放置在组织培养基上并且分割成小块。将小的组织块放置在组织培养瓶的湿表面上,其中每个培养瓶中放置大约10块组织。将培养瓶倒置,合紧并于室温下保留过夜。在室温下过24小时后颠倒培养瓶,组织块仍固定在培养瓶底部,加入新鲜培养基(例如含有10%FBS,青霉素和链霉素的Ham’s F12培养基),然后于37℃下温育大约1周。这时加入新鲜培养基,随后每隔几天更换一次培养基。再培养2周后,出现了一单层成纤维细胞。该单层细胞经胰蛋白酶消化并放入更大的培养瓶中。
用EcoKI和Hind III消化侧翼有Moloney鼠肉瘤病毒长末端重复的pMV-7(Kirschmeier,P.T.等,DNA,7:219-25(1988)),接下来用牛小肠磷酸酶进行处理,将该线性载体在琼脂糖凝胶上分级分离并使用玻璃珠加以纯化。
采用分别与5’和3’末端序列对应的PCR引物扩增编码本发明多肽的cDNA。5’引物包含一个EcoRI位点,3’引物含有一个Hind III位点。在T4 DNA连接酶存在下,将等量的Moloney鼠肉瘤病毒的线性化骨架与EcoRI和Hind III片段加在一起,在适于两个片段连接的条件下维持所得到的混合物。将连接混合物用于转化细菌HB101,然后将该细菌涂布在含有卡那霉素的琼脂上以证实该载体具有插入正确的感兴趣的基因。
将兼嗜性的(amphotropic)pA3 17或GP+aml2包装细胞在组织培养物上进行培养,直至在含有10%小牛血清(CS)、青霉素和链霉素的Dulbecco’s改良Eagles培养基(DMEM)上达到汇合密度。然后将含有该基因的MSV载体加入培养基中并用该载体转导包装细胞,包装细胞随即产生含有该基因的具有感染性的病毒粒(现在包装细胞被称作生产细胞)。
向经转导的生产细胞中加入新鲜的培养基,接下来从一个铺满生产细胞的10cm平板中收集培养基。含有感染性病毒粒的旧培养基经一个微孔过滤器过滤以除去脱离(detached)的生产细胞,然后利用该培养基去感染成纤维细胞。从亚汇合的成纤维细胞的平板中除去培养基并迅速地代之以来自于生产细胞的培养基。除去该培养基并代之以新鲜的培养基。如果该病毒的滴度高的话,那么实质上所有的成纤维细胞均被感染并且无需选择。如果滴度非常低,那么就需要采用具有如neo或his这样的选择性标记的逆转录病毒载体。
然后将工程化的成纤维细胞注射入宿主,或者单独注射或者在一个cytodex 3微载体珠上已培养至汇合之后再注射。现在成纤维细胞生产蛋白产物。
根据以上的教导,本发明的许多改进和变化都是可能的,因此在附加的权利要求的范围内可以不同于所述的方式实施本发明。

Claims (20)
