技术领域
本发明涉及含有以下述物质作为有效成分的用于预防或治疗Th1型免疫疾病的药物组合物,所述的物质可作用于在树突状细胞中表达的钠利尿肽(NP)的受体鸟苷酸环化酶A(GC-A),促进环单磷酸鸟苷(cGMP)产生。
背景技术
免疫系统原本作为用以识别和排除外来异物(微生物等)的防御机构而得到进化。所以,机体能将自己的细胞或组织与外来的异物(非己)区别开来,对自己不反应,或即使反应也持续保持不能发挥功能的状态(免疫宽容),使能够迅速并且有效地排除非己的后天免疫日益成熟。在后天免疫中承担中心作用的是T细胞。未分化的外周原始T细胞(Thp),在抗原刺激下开始增殖、分化。此时,Thp通过表面上的T细胞受体接受来自巨噬细胞或树突状细胞等抗原呈递细胞(antigen-presenting cell)的抗原呈递,同时在来自活化相关分子群的信号的作用下得到活化,分泌IL-2,进行增殖。然后分化成几乎能够产生所有细胞因子的Th0,根据抗原刺激的种类或强度、抗原呈递细胞产生的刺激信号等,确定最终的分化方向,成熟为Th1或Th2,各自行使功能,分别产生特异的细胞因子,进一步诱导细胞增殖、细胞毒活性等。即,白介素12(IL-12)诱导向与细胞性免疫相关的Th1细胞的分化,分化了的Th1细胞产生白介素2(IL-2)或干扰素-γ(IFN-γ)等细胞因子,另一方面,白介素4(IL-4)诱导向参与体液性免疫相关的Th2细胞的分化,产生白介素4、10(IL-4、10)等细胞因子。两种细胞所产生的细胞因子或者使细胞向Th1或Th2细胞分化或使两种细胞产生的细胞因子的活动相互负向调控,保持Th1/Th2的平衡。
近年来,认为这种Th1/Th2的失衡是免疫性疾病的原因。当倾向于以Th1为中心的免疫应答(Th1型免疫)时,细胞性免疫被增强的针对癌症和所感染疾病的免疫反应被促进,这成为引起对自身组织伤害的自身免疫疾病并发症的原因。组织的损伤和感染导致持续的炎症反应、而且进一步导致组织的纤维化、脏器的功能障碍。
此外,在治疗方面,期待着能够抑制即使是正常免疫应答的伴随脏器移植的排斥反应或伴随骨髓(造血干细胞)移植的针对移植物的宿主病。这些所诱导的免疫应答基本上是一致的,以Th1性免疫为主体。
作为Th1型免疫导致的免疫异常而引发的Th1型免疫疾病,例如包括:移植排斥反应、骨髓(造血干细胞)移植导致的移植物抗宿主疾病以及自身免疫性肝炎、类风湿性关节炎、胰岛素依赖性糖尿病、溃疡性结肠炎、克郎氏病(Crohn′s disease)、多发性硬化症、自身免疫性心肌炎、银屑病、皮肤硬化症、重症肌无力、多发性肌肉炎/皮肤肌肉炎、桥本病(Hashimoto′s disease)、自身免疫性血细胞减少症(纯红细胞再生障碍性贫血、再生不良性贫血等)、干燥综合征、血管炎综合症、全身性红斑狼疮等的自身免疫疾病,此外,可列举伴随这类疾病导致的组织损伤或感染而引发的炎症反应、纤维化、脏器功能障碍等。
在针对Th1型免疫导致的自身免疫疾病或移植免疫,以及与之相伴的慢性活动性的免疫学炎症反应的治疗中,希望选择性抑制Th1型免疫.在这种情况下,现在,希望通过控制T细胞活化系统的介导因子实现治疗,环孢素或FK506针对T细胞的强有力的免疫抑制效果、抗细胞因子疗法、抗粘附分子(活化相关分子)疗法、单克隆抗体疗法等引人注目.
可是,这些疗法也和以往的甾体制剂、作用于核酸合成体系的某种免疫抑制剂或干扰素制剂的疗法相同,不具有对Th1型免疫的选择性,而且存在感染症恶化、糖尿病、血栓、满月脸、肾病、发热等各种副作用等问题需要解决,因此需要开发安全性更好、有效性更优越的药物制剂。
自身免疫性疾病除了是全身性疾病之外,也是可以在大多数脏器中可以发现的疾病。而且以后,伴随着脏器移植手术或造血干细胞移植的日益发展,排斥反应或移植物抗宿主病(Graft-versus-hostdisease)的问题也将增加。这样,因为Th1型免疫起到支配性作用的疾病涉及面广泛,所以期待开发有效的治疗药物。实际上,作为具有Th1型免疫抑制作用的药物制剂,据报道有serotonin 1A受体拮抗剂(J Immunol 153:489-498,1994)、pentoxifyllin(J CardiovascPharmacol 25Suppl 2:S75-79,1995)、beta2-adrenergic受体拮抗剂(J Immunol 158:4200-4210,1997、J Clin Invest 100:1513-1519,1997)、腺苷酸环化酶活化剂的iloprost(J Autoimmun 10:519-529,1997)、P38 MAP激酶抑制剂的pyridinyl imidazole化合物或SB203580(EMBO J 17:2817-2829,1998、Int Immunol 12:253-261,2000)、lisofylline(J Immunol 163:6567-6574,1999)、adenosineA2a受体拮抗剂的CGS-21680(J Immunol 164:436-442,2000)、NO-aspirin(Gastroenterology 118:404-421,2000)以及1,25-dihydroxyvitamin D(3)(Eur J Immunol 30:498-508,2000)等。可是,尚未开发出可选择性抑制Th1型免疫并减轻副作用的可用于临床的医药制品。
近年已经阐明克郎氏病是Th1型疾病,该病作为IL-12和IL-10治疗的靶标引人注目(最新医学90:1076-1081,2004)。即,IL-12是由活化的巨噬细胞或树突状细胞分泌的,其在由原始T细胞向Th1细胞的分化中起到支配性作用。据报道在模型动物中,抗IL-12抗体显示出治疗效果,因此认为也可以将其用做治疗人克郎氏病的候选目标。此外,IL-10是能够抑制从Th1细胞生成细胞因子的分子,在IL-10基因敲除的小鼠中,Th1应答增强,自然引发肠炎(Clin Invest Med2001;24:250-257),因此期待IL-10也可以作为克郎氏病的治疗的候选目标。实际上,已经研究了基因重组人IL-10对克郎氏病的效果,发现具有改善效果,但是其效力不充分,此外,还发现产生头疼、发热、贫血等副作用(Gastroenterology,2000;119:1473-1482)。当血中持续维持高浓度的IL-10时,暗示表现出免疫活化作用,期待着仅在病灶局部能够生理性地促进产生IL-10的治疗方法。最近,对于中等程度到重度的克郎氏病,采用了TNFα抗体的infliximab,虽然确认了其有效性,但是据报道也有高频率的副作用发生,这限制了其使用。
此外,多发性硬化症也是自身免疫疾病中特别以Th1型免疫占主导地位的疾病,已有指出复发-缓解型也是在缓解时向Th2偏移等,期待能将Th2倾向作用用于疾病治疗。现在作为用于多发性硬化症的治疗药物,已知在急性期使用甾体制剂、在复发-缓解期使用干扰素β1b、在稳定期使用各种免疫抑制剂等。已有指出这些制剂具有各种副作用,此外有效性也不充分,所以不能充分满足使病情确切好转、防止恶化、减轻副作用的需要。
所以,期待着开发出针对两种疾病能更有选择性地抑制Th1型免疫、减轻副作用的医药制品,现在尚无可以用于临床的药物。
已知树突状细胞是能在淋巴器官T区最强有力地活化原始T细胞的唯一的抗原呈递细胞,并在维持生物体防御的稳定性方面担当着重要的作用(Banchereau,J.等,Nature,Vol.392,p245,1998).树突状细胞在普通组织中以T细胞的活化能低下的未成熟状态存在,但在从病原体或损伤了的组织中释放出来的炎症性介质的作用下,受到成熟、活化的刺激,将来自抗原的肽与MHC复合体结合,呈递给原始T细胞.同时,使共刺激分子的表达增强,使之产生各种细胞因子,募集T细胞,活化抗原特异性的细胞,诱导免疫应答(伊豫田智典、Inaba Kayo,蛋白质、核酸和酶,Vol.47,p2133,2002)。此外,人树突状细胞中至少存在2中前体细胞。单核系前体细胞在GM-CSF和IL-4的刺激下而分化,此外,通过CD40L的刺激产生IL-12,通过IL-12的作用使原始T细胞分化成Th1细胞。转化细胞系前体细胞在病毒或CpG寡核苷酸、以及IL-3及CD40L的刺激下,分化成几乎不产生IL-12的树突状细胞,使原始T细胞分化成Th2细胞(樗木俊聪,医学のあゆみ、Vol.205,p57,2003)。即,树突状细胞是在免疫中承担控制Th1/Th2平衡的重要作用的细胞。所以,如果能够开发出特异性作用于树突状细胞,调节其活性或细胞因子的表达,抑制原始T细胞向Th1的分化增殖,使之倾向Th2型的药物,就有望可以根治或预防Th1型免疫导致的免疫异常(特别是克郎氏病或多发性硬化症)。可是,至今为止,尚没有具有这种作用的有效药物。
另一方面,作为能够作用于GC-A促进产生第二信使cGMP的物质,可列举肽性物质,特别是钠利尿肽,该肽有ANP(心房钠利尿肽)、BNP(脑钠利尿肽)以及CNP(C型钠利尿肽)3种,作为与该肽相对应的NP受体,有GC-A、GC-B(鸟苷酸环化酶B)以及NPR-C(NP受体-C)3种。已经发现:GC-A和GC-B具有膜结合型鸟苷酸环化酶结构;ANP以及BNP是GC-A的特异性配体;CNP是GC-B的特异性配体;以及这些物质分别与各自的受体结合后,能通过使细胞内的cGMP上升而发挥利尿作用以及血管扩张作用等生理作用。此外,据称NPR-C与cGMP的不产生共轭(couple),与这些的激素代谢、清除有关(Suzuki,T.等,Cardiovasc.Res.Vol.51,p489,2001)。
ANP是由心脏分泌的、对水电解质代谢以及血压调节具有重要作用的肽激素。已知在人以及模型动物中,伴随着心肌肥大以及心不全的加重,血中的APN浓度上升,认为其起到了对心不全病情的代偿性作用。实际上,在心不全的患者中,发现通过给予ANP发挥其血管扩张作用以及利尿作用,使心脏的前负荷、后负荷减轻,产生了改善血液流动状态的效果(Suzuki,T.等,Cardiovasc.Res.Vol.51,p489,2001)。
据报道ANP受体的GC-A不只在心血管系统中,也在白细胞中有表达,暗示ANP可能对血细胞承担有生理功能。即,ANP促进嗜中性细胞的释放(Izumi,T.等,J.Clin.Invest.Vol.108,p203,2001)、抑制大鼠胸腺细胞的增殖(Vollmar,A.M.,K.N.,等,Endocrinology.Vol.137,1706,1996)、增强人自然杀伤(NK)细胞的细胞杀伤性(Moss,R.B.,and M.G.Golightly,Peptides,Vol.2,p851,1991)、抑制从小鼠巨噬细胞中产生NO或TNFα(Kiemer,A.K.and A.M.,Vollmar.J.Biol.Chem.Vol.273,p13444,1998)等。
可是,据报道与作用于来自小鼠单核细胞的巨噬细胞的情况不同,人单核细胞不表达ANP受体,ANP在单核细胞中不表达产生cGMP等生理活性(Sprenger H.,等,Immunobiol.Vol.183,p94,1991)。完全没有关于作用于来自与免疫系统相关的人单核细胞的树突状细胞上的GC-A以促进cGMP产生物质的生理功能或病理生理性意义、或免疫调节作用的报道。
发明内容
因此,本发明鉴于上述现状以提供下述抑制剂为课题:该抑制剂针对Th1型免疫疾病的自身免疫疾病、脏器移植的排斥反应、骨髓(造血干细胞)移植导致的移植物抗宿主病、以及伴随着起因于自身免疫疾病或类似疾病的组织损伤或感染发生的炎症反应,还有组织纤维化和脏器的功能障碍,是不具有以抑制Th1型细胞因子的产生、抑制Th1细胞的增殖/功能抑制作为作用原理的副作用,可能适于临床使用并且具有Th1型免疫选择性的抑制剂。具体而言,提供了能够用于预防或治疗Th1型免疫疾病(特别是克郎氏病或多发性硬化症)的药物组合物,该组合物以作用于在树突状细胞表达的NP受体GC-A,促进cGMP的产生、调节树突状细胞的细胞因子的产生、能将T细胞分化成Th2的物质作为有效成分。
作为可以用作本发明所涉及的药物组合物的有效成分的物质,只要具有通过NP受体GC-A促进cGMP产生的特性的物质即可,优选肽类物质,因为没有特定的限定,也可以使用肽类物质以外的能够作用于NP受体GC-A促进cGMP的化合物。
作为肽类物质,优选钠利尿肽(natriuretic peptide),例如:特别是心房钠利尿肽(以下称为ANP)、脑钠利尿肽(以下称为BNP)等。
作为ANP,可以使用由28个氨基酸组成的来自人的α-hANP(序列号:1)或来自大鼠的α-rANP(序列号:2),作为本发明的有效成分的肽,只有具有ANP的环结构(基于Cys形成的二硫键)以及连在环结构上的C末端部分的肽即可。作为该肽,可列举具有α-hANP的7-28位的氨基酸残基的肽(序列号:3)。作为ANP,特别优选是来自人的α-hANP。
作为BNP,例如:由32个氨基酸组成的人BNP(序列号:4)等。
此外,作为本发明中涉及的具有通过NP受体GC-A促进cGMP产生的特性的物质,可以是从天然中单纯地分离、纯化出的物质,或是通过化学合成法或基因重组法制备得到的物质,例如,基于上述物质(α-hANP等)涉及的氨基酸序列,如果是本领域的技术人员,可以通过适当的公知方法,通过实施缺失、置换、添加、插入等修饰该序列中的氨基酸残基而得到。无论通过何种方法得到的物质,只要是能通过作用于NP受体GC-A促进cGMP产生的物质均可使用。作为该物质,除了上述物质之外,还可列举:蛙ANP(序列号:5)、猪BNP(序列号:6)、大鼠BNP(序列号:7)、鸡(序列号:8)等。
作为可以用作本发明的药物组合物的有效成分的物质,可以使用无机酸,例如:盐酸、硫酸、磷酸,或有机酸,例如:甲酸、醋酸、丁酸、琥珀酸、柠檬酸等酸的加成盐使用。也可以是钠、钾、锂、钙等的金属盐,有机碱的盐形式。此外,本发明的医药组合物可以作为有效成分物质的游离形式,也可以是其在医药上可被允许的盐。
可以用作本发明的药物组合物的有效成分的物质或其在药理学上允许的盐,优选和公知的药理学上允许的载体、赋形剂、稀释剂等混合,通过在医药中常规使用的给药方法,即经口给药方法,或静脉内给药、肌肉内给药或皮下给药等非经口给药方法进行给药。
有效成分为肽类物质时,可以作为在消化道内难以分解的制剂,例如:作为将活性成分的肽包容在脂质体中的微囊制剂,经口给药。此外,可以使用从直肠、鼻内、舌下等消化道以外的可使之从粘膜吸收的给药方法。在这种情况下,可以作为栓剂、鼻喷雾、舌下片的形态来给药。
可用作本发明医药组合物的有效成分的物质的给药量根据疾病的种类、患者的年龄、体重、症状的程度以及给药途径等有所不同,通常在0.1μg/kg~100mg/kg的范围内给药,优选以0.5μg/kg~5mg/此给药.
根据本发明可知:以作用于钠利尿肽受体GC-A促进cGMP产生的物质作为有效成分的组合物可以特异性地作用于树突状细胞,调节其产生细胞因子,因为其通过诱导原始T细胞的Th2倾向性,可抑制Th1型免疫反应,所以对Th1型免疫疾病有效。特别是,将ANP作为有效成分使用的时候,单独的ANP不能对从树突状细胞产生细胞因子或T细胞的增殖反应产生影响,仅表现出调节LPS(脂多糖)刺激时的反应的作用,不对正常时的免疫功能产生大的影响,仅抑制刺激时的过剩反应,即,表现出副作用小,可以安全使用,是有用的。
此外,LPS是革兰氏阴性菌的外膜的主要构成成分,能被对病原体固有的构成成分的识别极其重要的树突状细胞所表达的称为Toll样受体(TLR)家族的膜蛋白受体群中的TLR4所识别,是刺激树突状细胞的成熟、活化,并诱导细胞因子以及CD40等辅助功能分子表达的物质。
根据本发明,通过使用来自人的树突状细胞以及原始T细胞的一系列实验,阐明了在人树突状细胞中的GC-A的表达、可作用于NP受体GC-A促进cGMP产生的物质对树突状细胞的细胞因子产生的调节作用以及使T细胞倾向Th2的作用,这在临床上是有用的。
由此可知,可作用于NP受体GC-A促进cGMP产生的物质表现出作用于在树突状细胞中表达的GC-A,使原始T细胞倾向于Th2型的作用,因为能调节在免疫系统中的T细胞所涉及的Th1/Th2平衡,所以给予该物质,能够改善Th1型免疫疾病(特别是克郎氏病或多发性硬化症),以及即使在患有Th1型免疫疾病以前的状态下,根据常规方法测定Th1和Th2的比例,当Th 1的比例高时,给予该物质,通过调节免疫系统中的Th1/Th2的平衡,可以预防Th1型免疫疾病。
综上所述,本发明包含下述内容:
(1)用于预防或治疗Th1型免疫疾病的药物组合物,其中,以可作用于钠利尿肽受体鸟苷酸环化酶A促进产生环单磷酸鸟苷的物质为有效成分。
(2)上述(1)所述的药物组合物,其中,Th1型免疫疾病选自起因于伴随移植的排斥反应的疾病、骨髓(造血干细胞)移植导致的移植物抗宿主病以及自身免疫疾病。

发明效果
根据本发明,可作用于钠利尿肽受体鸟苷酸环化酶A促进产生环单磷酸鸟苷的物质,因为具有通过作用于树突状细胞,诱导Th2倾向性,通过使T细胞分化成Th2型细胞,抑制IL-12以及TNFα的产生,使IL-10的产生增强,由此抑制Th1型免疫反应的作用,所以以该物质为有效成分的药物组合物通过调节免疫系统中的Th1/Th2的平衡,作为用于预防或治疗Th1型免疫疾病(特别是克郎氏病或多发性硬化症)的药物组合物是非常有用的.

[图3]图3表示ANP对由经过LPS刺激的树突状细胞作用的同种原始T细胞的增殖反应的效果。树突状细胞在LPS(1μg/mL)、ANP(10×-7M)、LPS+ANP存在或不存在的情况下,培养24小时,经过照射后,和原始T细胞共同培养6天。可以用[3H]-胸苷掺入能力对此时的原始T细胞的细胞增殖能力进行评价。白色圆形:无处理,黑色圆形:ANP 10-7M处理,白色四方形:1μg/mL的LPS处理,黑色四方形:ANP+LPS处理。各个值用5例的平均值±标准误差表示。*以p<0.05表示与其他组间存在有意义差,用Student的t检验检测有意义差。
[图4]图4表示ANP以及CNP对经LPS刺激后的树突状细胞产生细胞因子的影响。表示的是将1×105个细胞/管的树突状细胞在ANP(10-8~10-6M)或CNP(10-6M)的存在或不存在的情况下,与LPS(1μg/mL)共同温育24小时后,检测培养基中的IL-12、TNF-α、IL-10的免疫活性。
[图5]图5表示的是将用LPS或LPS+ANP预处理过的树突状细胞与原始T细胞共同培养后,再在含有IL-2的培养液中增殖该T细胞,用流式细胞术解析该T细胞的细胞内的IFN-γ、I L-4的产生的结果。图中的数字表示在各个组分中的白细胞的百分比。1号样品、2号样品分别表示使用来自不同供体的树突状细胞的结果。
具体实施方式
本发明人对从人的外周血中分离、培养的树突状细胞中的ANP受体(GC-A)表达、和ANP促进cGMP产生的活性进行了研究,此外,为了阐明了ANP在树突状细胞中的生理功能,针对其在树突状细胞的分化、淋巴细胞的增殖作用、细胞因子的表达、Th1/Th2倾向性的作用进行研究,结果发现可作用于GC-A促进cGMP产生的物质,对于Th1型免疫疾病的预防或治疗有用。
A.人树突状细胞的分离、以及NP受体表达、促进cGMP产生作用的实验方法
1.来自人外周血的树突状细胞的分离和培养
在实验中,使用从正常人志愿者得到的外周血(京都府红十字中心提供)的白细胞层(buffy coats).用Ficoll-Paque密度梯度离心分离外周血单核细胞级分后,通过使用了MACS CD14的磁珠柱子,或在细胞培养瓶中于37℃下培养1小时,通过选择出粘附细胞,进行单核细胞的分离。将2×105个细胞/ml的单核细胞在10%胎牛血清、50ng/mL人GM-CSF、20ng/mL人IL-4存在的情况下,在37℃培养7天获得未成熟的树突状细胞。
2.NP受体的RT-PCR
使用RNA分离试剂盒,从单核细胞和树突状细胞中提取总RNA后,用1μg的总RNA和oligo(dT)引物,通过来自骨髓祖细胞病毒的逆转录酶合成一条cDNA。使用下述引物。
针对NP受体进行35个循环的PCR,针对β-actin进行25个循环的PCR。用1.5~2%的琼脂糖分离PCR产物,用溴化乙锭染色后,以紫外投射装置检测。
3.测定促进cGMP产生的活性
将1×105个细胞/样品的细胞加入到500μL的培养液中,在存在有10mM HEPES、0.5mM 3-异丁基-1-甲基黄嘌呤、1μM phospholamidon的情况下,于37℃下温育10分钟后,添加ANP或CNP使终浓度为10-12~10-6M,此外再温育15分钟。洗涤细胞、破碎后,用ELISA法测定细胞内的cGMP浓度。
B.通过树突状细胞使原始T细胞发生增殖反应、产生细胞因子、原始T细胞分化的实验方法。
1.通过活化的树突状细胞使T细胞增殖的反应
原始T细胞(原始CD4+T细胞)是采用用于分离MACS CD4+T细胞的磁珠从来自新生儿脐带血的单核细胞级分中分离得到的。在ANP(10-7M)存在或不存在的条件下,将树突状细胞与脂多糖(LPS、1μg/mL)培养24小时,使之激活。洗涤细胞,经放射线照射(30Gy)使之失去增殖能力后,和原始T细胞(1×105个细胞/孔)共同培养6天。通过使之掺入0.5μCi/孔的(甲基-3H)-胸苷8个小时,对T细胞的增殖能力进行评价。
2.测定从树突状细胞中产生的细胞因子
在ANP(10-8M~10-6M)或CNP(10-6M)存在或不存在的情况下,将树突状细胞与LPS(1μg/mL)温育24小时,用ELISA法测定培养上清中的IL-12、IL-10、TNFα的量。
3.原始T细胞的细胞内细胞因子的表达分析(研究树突状细胞的Th1/Th2的倾向性)
将树突状细胞预先在ANP(10-7M)存在或不存在的条件下与LPS(1μg/mL)共同温育24小时,使之活化后,通过放射线照射(30Gy)使之失去增殖能力。将该树突状细胞(1×105个细胞/孔)和从脐带血分离的原始T细胞(1×106个细胞/孔)共同培养6天。在IL-2(50U/mL)存在的情况下,再培养T细胞8天使之增殖后,回收T细胞,用50ng/mL的佛波醇酯(PMA)和500ng/mL的Ionomycin刺激4个小时。在温育结束的2小时之前加入BrefeldinA(10μg/mL)。用2%的福尔马林固定细胞后,用含有2%FBS、0.5%皂苷的溶剂破坏细胞膜,用FITC标记了的抗IFN-γ单克隆抗体以及PE标记了的抗IL-4单克隆抗体染色细胞内细胞因子,用流式细胞术进行分析。IFN-γ阳性细胞为Th1型细胞,IL-4阳性细胞为Th2型细胞。
实施例
下面通过实施例对本发明进一步详细说明。
实施例1在未成熟的树突状细胞上表达GC-A基因以及通过ANP促进cGMP产生的活性
本发明人等,首先针对3种NP受体基因在树突状细胞、单核细胞中的表达进行了研究。从获自正常人外周血的单核细胞以及未成熟树突状细胞中制备RNA,以RT-PCR法对NP受体GC-A、GC-B、NPR-C的基因表达进行讨论,结果如图1所示。
经确认作为阳性对照的来自胎盘的RNA中表达了全部3种NP受体基因。明确确认相对于在单核细胞中未表达任何受体基因,在未成熟的树突状细胞中仅表达GC-A基因。所以可以确定在单核细胞中不表达NP受体基因,在来自单核细胞的树突状细胞中特异性地表达GC-A基因。
然后,为了阐明在树突状细胞中表达的GC-A是否和生理功能相关联,分别使单核细胞、树突状细胞对ANP的cGMP产生促进活性进行了研究。作为对照,使用GC-B特异性的配体CNP。结果如图2所示。
ANP通过以10-12M这种低浓度呈现浓度依赖性地使细胞内的cGMP上升,但在单核细胞中以任何浓度都几乎不能使cGMP上升。此外,CNP在单核细胞、树突状细胞中的任何一个中都对细胞内的cGMP浓度没有影响。该结果支持图1中所示的GC-A基因的表达结果,也很强烈地显示可能是树突状细胞特异性地表达GC-A,并且该受体与ANP反应担负着生理活性。
实施例2有关ANP对树突状细胞表达细胞因子的作用以及Th2倾向性的研究
从实施例1的结果可知,ANP作用于树突状细胞,能够调节免疫反应。
树突状细胞的重要功能是引发原始T细胞的抗原特异性免疫反应。因此首先,在已知能使树突状细胞成熟、活化的LPS的存在下以及不存在的条件下,将ANP添加到树突状细胞中,对其涉及针对原始T细胞的活化、增殖反应的影响进行研究。结果如图3所示。
ANP单独不对原始T细胞的增殖反应造成影响,一旦向树突状细胞中添加LPS,就会引发显著的T细胞增殖反应。
另一方面,已知一旦同时添加ANP和LPS,就能够几乎完全地抑制LPS处理过的树突状细胞所诱导的T细胞增殖。
所以,ANP表现出能影响由LPS导致的树突状细胞的活化信号或影响树突状细胞的功能.
已知树突状细胞产生的细胞因子对树突状细胞与T细胞的相互作用表达具有非常重要的作用。所以,下面关于ANP对从树突状细胞表达细胞因子作用进行了研究。结果如图4所示。
LPS促进从树突状细胞中产生IL-12、TNF-α、IL-10,与此相对,ANP呈现浓度依赖性地使IL-12和TNFα降低,另一方面,使IL-10的产生增强。另外,在LPS未刺激时,从树突状细胞中产生IL-12、TNF-α、IL-10的能力低,单独使用ANP对此没有影响。
此外,因为GC-B配体CNP对细胞因子的表达没有影响,由此确认在LPS刺激时的ANP作用是通过树突状细胞所表达的GC-A所介导的。
由树突状细胞产生的细胞因子中IL-12是具有代表性的Th1型细胞因子,另一方面,已知IL-10对由Th1细胞、活化的单核细胞以及NK细胞所产生的包括IL-12在内的细胞因子的活性具有阻断作用。ANP抑制LPS所诱导的由树突状细胞中产生IL-12,但增强IL-10的产生,据此表现出ANP有可能诱导树突状细胞倾向于Th2细胞。
所以,针对ANP是否能够诱导树突状细胞倾向于Th2,用LPS刺激树突状细胞时,使之与ANP共存,分析原始T细胞究竟向Th1、Th2的那一种类型分化、增殖。已知LPS作用于树突状细胞的Toll样受体4,促进IL-12的表达,使原始T细胞倾向于具有高的IL-12产生能力的Th1型细胞(Akira S等,Nat.Immunol.Vol.2,p675,2001)。
单独的LPS、或LPS与ANP共存下刺激的树突状细胞和原始T细胞相互作用后,将T细胞在含有IL-2的培养液中增殖,使用流式细胞术分析Th1型细胞表达的细胞因子IFN-γ、Th2型细胞表达因子IL-4。图5表示了2例T细胞样品的分析结果。在图5中表示了IFN-γ阳性、IL-4阴性细胞向Th1型细胞分化、增殖,IL-4阳性、IFN-γ阴性细胞向Th2型的辅助T细胞分化、增殖。
在1号样品和2号样品的任何一个中,与LPS单独处理组相比,在ANP共处理组中,IL-4产生细胞明显增加,IFN-γ产生细胞明显减少。此外,对另外的3份样品进行了研究,得到了相同的结果。
通过本实施例,可知ANP促进树突状细胞产生IL-10,使IL-12的产生降低,据此拮抗LPS的作用,将树突状细胞向Th2倾向性方面诱导,使T细胞向Th2型辅助T细胞分化,据此抑制Th1型免疫反应。

Claims (9)
1.可作用于钠利尿肽受体鸟苷酸环化酶A促进产生环单磷酸鸟苷的物质在用于制造预防或治疗Th1型免疫疾病的药物组合物中的应用。
2.权利要求1所述的应用,其中,Th1型免疫疾病选自伴随移植的排斥反应所导致的疾病、造血干细胞移植导致的移植物抗宿主病以及自身免疫疾病。
3.权利要求2所述的应用,其中,免疫疾病选自自身免疫性肝炎、类风湿性关节炎、胰岛素依赖性糖尿病、溃疡性结肠炎、克郎氏病、多发性硬化症、自身免疫性心肌炎、银屑病、皮肤硬化症、重症肌无力症、多发性肌肉炎/皮肤肌肉炎、桥本病、自身免疫血细胞减少症、干燥综合征、血管炎综合症以及全身性红斑狼疮。
4.权利要求3所述的应用,其中,自身免疫血细胞减少症为纯红细胞再生障碍性贫血。
5.权利要求3所述的应用,其中,自身免疫血细胞减少症为再生不良性贫血。
6.权利要求3所述的应用,其中,自身免疫疾病为克郎氏病或多发性硬化症。
7.权利要求1所述的应用,其中,可作用于钠利尿肽受体鸟苷酸环化酶A促进产生环单磷酸鸟苷的物质为钠利尿肽。
8.权利要求7所述的应用,其中,钠利尿肽为心房钠利尿肽或脑性利尿肽。
9.权利要求8所述的应用,其中,心房钠利尿肽来自人。
