背景技术
生物反应器或者发酵器是这样一种容器,这些容器被用于发酵、酶反应、细胞培养、生物制品、化学试剂、生物制药、组织工程、微生物、植物代谢物、食品生产等等。从台式发酵罐到各种尺寸的独立单元,生物反应器在尺寸上是不同的。在一些生物反应器中用于无菌生产的严格无菌条件通常需要精密系统,以完成期望的产品体积。因此,在无菌生物反应器中产品的生产可能是昂贵的,这就形成了追求改善系统的动机。
传统的生物反应器经过单独的一种中空纤维来灌注培养基。这种生物反应器的各种缺陷包括不均匀细胞团块、难以取得典型细胞生长样本、由于无效的氧合作用和不能控制氧含量导致的性能差以及与细胞培养污染有关的问题。此外,例如pH值的微环境因素不能有效地控制,同时细胞的混合培养或者共培养也不可能。如在第5,622,857号美国专利中公开的那样,这种现有技术生物反应器的改进在于中空纤维反应器。此反应器包括反应容器,多孔中空纤维的中心股贯穿其中延伸,营养液经过该反应容器被抽吸。该中空纤维的中心股被多个中空纤维股同心围绕,气态培养基通过这些中空纤维股传送。这些股的中空纤维还以这样方式构成,即例如氧或者二氧化碳的气态培养基可至少部分地从这些股流出或者分别地进入这些股内。与现有技术装置相比,这类生物反应器可获得一定程度增强的培养基氧合作用。然而,细胞培养的偶然污染和不能对pH指标有效地控制一直是个问题。
由于需要的清洁、消毒和对标准生物反应器(即不锈钢或者玻璃反应器)的验证,从而增加了在无菌的生物反应器中生产细胞、生物制药、生物制品等等的费用。预先杀菌一次性生物反应器系统不需要终端用户清洁、杀菌或者验证,随着这种一次性生物反应器系统的发展,已经尝试解决这个问题。通过利用这种一次性生物反应器系统可明显节约成本。此外,塑料重量轻,便于运输以及比不锈钢或者玻璃反应器需要更小空间。在第6,245,555 B1号美国专利中提供了用于生物反应器中一次性元件的实例,其中该专利描述了带有支撑壳体的反应器腔室。该支撑罩的内腔镶有一次性衬里,并密封有附着于衬里上的顶板,以形成密封室。因为衬里在顶部敞开时,它必须用于垂直取向的生物反应器,以阻止顶板的污染。尽管此系统提供了一次性衬里,但顶板和内腔仍然需要清洁和灭菌。
另一种方案是开发柔软的、一次性的塑料容器,这种容器不需要清洁或者灭菌,且只需要最少的验证工作。例如,第5,523,228美国专利描述了一种柔软、一次性和可透气的细胞培养腔,其中该细胞培养腔水平地旋转。该细胞培养腔由熔凝在一起的两片塑料制成。另外,培养腔用透气材料制成,并安装在水平旋转盘驱动器上,其中该驱动器支撑柔软培养腔,而不阻碍在薄膜表面上方的气流。该腔处于细菌培养箱(incubator)内,同时根据袋子的通透系数通过控制在该细菌培养箱内气压来控制氧气输送。袋子的旋转帮助袋子中内容物的混合。然而,细胞培养腔会限于在被控的气体环境内使用。特别是,细胞培养腔没有支撑机构,因此被限制成小容积。此外,该培养腔没有提供在旋转期间用于把培养基从培养腔中恒定抽入和抽出的入口和出口。
一些公司已经开发出大量不需要清洁或者灭菌的预先无菌、一次性生物反应器(例如Wave Biotech,Bridgewater,N.J)。这种反应器由柔软不透气材料片制成,以形成袋子。该袋子局部填充有培养基,然后用连续穿过袋子顶部空间的空气膨胀。该培养基通过摇摆袋子而混合并充有气体,以增加空气液体接触面。然而,由于没有支撑袋子的固定壳体,从而由于这些袋子在尺寸上增加而变得笨重并难以操纵。此外,在摇摆袋子内部的波动作用可产生破坏性的扰动力。某些细胞培养物,特别是人类细胞培养物在更温和条件下会生长得更好。
因此,在这个领域一直存在着开发柔软、预先灭菌、一次性的生物反应器的需求,这种生物反应器容易操纵并几乎不需要培训就可以操作,但仍然能提供用于成功的细胞和组织培养而必要的气体输送和培养基组合。这种一次性生物反应器对于化学试剂、生物制药、生物制品、细胞、微生物、植物代谢物、食品等等的生产同样有用。
发明内容
本发明提供一种具有一次性元件的搅拌罐反应器系统,其中该一次性元件例如为带有固定轴承、轴和叶轮组件的柔软塑料袋子。本发明还涉及作为一次性生物反应器以及在具有一次性元件的套件中的该新颖搅拌罐反应器中的使用。本发明的优点是多方面的。具体地说,搅拌罐反应器系统可预先灭菌,而不需要在培养或者生产系统中从批次到批次或者从产物到产物变化地就地蒸发(SIP)或者就地清洁(CIP)环境。因而,该系统通过确保批次到批次的零污染而需要很少的调节控制,并因此可以相当低的成本来操作,同时在使用前准备很少或者根本不需要准备。另外,该系统为不同于其他一次性反应器系统的纯粹搅拌罐反应器系统。这提供了附加的优点,即本发明提供了这样的流体动力环境,其中可类似于传统的非一次性反应器系统那样按照比例变化各种尺寸。由于该系统一般不需要清洁或者灭菌,在细胞培养或者生产过程期间,它把柔软、易于使用、纯粹的搅拌罐反应器环境与零交叉污染结合起来。

本发明的搅拌罐反应器系统提供用于各种目的的柔软和一次性的袋子,其中各种目的包括培养细胞、微生物或者植物代谢物以及加工食品、化学试剂生物制药和生物制品。该一次性袋子可包括例如轴、叶轮和轴承的一次性元件,并设计成能嵌入到例如反应器壳体的固定壳内。该袋子可还包括一个或多个开口、小囊、端口等等。该搅拌罐反应器系统允许用户轻松并几乎不用培训就能操作细胞培养或者生产。特别是,该一次性系统不需要清洁或者灭菌。此外,该系统在生产过程之间不需要连续的验证。因此,它在生产过程中将零交叉污染与柔软、易于使用、纯粹的搅拌罐反应器环境结合起来。
参见附图,图1示出了具有至少一个开口的柔软袋子(4)和具有可连接叶轮(13)的搅动轴(12)。如在这里所示的那样,搅动轴(12)和相连接的叶轮(13)位于袋子(4)内。此外,该搅动轴(12)连接到轴承(5)上,其中该轴承可通过密封件或者O形圈封接到袋子上。包括密封件或者O形环(6)的该袋子(4)、搅动轴(12)、叶轮(13)和轴承(5)可选地为一次性的。该一次性袋子可以是柔软的塑料袋子。该袋子(4)可通过至少一个密封件或者O形圈固定到搅动轴(12)和轴承(5)上,从而在该柔软袋子内保持无菌。如图1所示,该密封件或者O形圈可进一步固定到该袋子上。此外,该一次性搅拌罐反应器系统可连接到容纳轴承(5)和电机(1)的支撑件或者一个或者多个支架(3)上。在一个实施例中(如图1所示),支撑件(3)为电机和轴承座(3),其中该搅动轴(12)的上端进一步连接到电机联轴节(2)上。该电机联轴节(2)连接到电机(1)上,而该电机驱动引起袋子(4)内部的流体动力环境的搅动轴(12)和叶轮(13)的搅拌运动。该袋子(4)设计成能嵌入到例如桶或者腔的壳体(11)内。该壳体可以是金属桶、塑料桶、木桶、玻璃桶或者由固体材料制成的任何其它桶或者腔。在本发明的一个实施例中,壳体还包括多个挡板,其中袋子围绕该挡板折叠。在另一个实施例中,该柔软袋子(4)还包括顶部端口(一个或者多个)(8)、底部端口(一个或者多个)(9)以及侧部端口(一个或者多个)(10),其中柔性管(7)可与这些端口中的一个或者多个连通。
该搅拌罐反应器系统可选择性地包括例如加热板、蒸汽加热套或者循环流体或者水加热器等加热器。在一个实施例中,该加热器位于袋子(4)和壳体(11)之间。在另一个实施例中,加热器容纳在壳体(11)内(例如容纳在反应器壳体和袋子之间的双壁内)。在又一个实施例中,该搅拌罐反应器系统位于培育器内。该加热器能进行特殊培养或者生产的加热或者取暖。这对于常常在37℃下生长的细胞培养物来说特别重要。
在本发明的一个实施例中,袋子(4)、轴承(5)、密封件或者O形圈(6)、管(7)、顶部端口(8)、底部端口(9)、侧部端口(10)、轴(12)和叶轮(13)是一次性的。电机(1)、电动机联轴节(2)、支架或电机和轴承座(3)以及壳体(11)是固定的。
C)装置和端口
搅拌罐反应器系统可还包括传感器及其他装置。在一个实施例中,袋子包括PH值传感器和溶解氧传感器,其中传感器结合到该袋子内。同样地,传感器可与袋子同样是一次性的。在另一个实施例中,传感器可连接到袋子上,并且是独立的单元。可选地,这种传感器在灭菌后能再利用。在另一个实施例中,该系统包括其中液流经过PH值传感器和溶解氧传感器的产物回路,其中这些传感器结合到反应器壳体内。该系统是柔性的,并提供供应各种备选设备(例如传感器、探头、装置、小囊、端口等等)的可选方法。该系统还可包括封接到袋子上的一个或者多个内部小囊。在一个优选实施例中,该小囊具有至少向袋子外部敞开的一端,以把探头插入到反应器(也就是该袋子)内,同时保持在袋子外部上。该探头例如可以是测温探头、pH值探头、溶解气体传感器、氧气传感器、二氧化碳传感器、细胞团块传感器、培养基传感器、渗透压力计或者任何其他能测试或者检验培养或者生产的探头。在另一个优选实施例中,该系统在袋子中包括至少一个端口,用于把装置连接到端口上。这种装置包括但是不局限于管、过滤器、连接件、探头和采样器。把各种端口结合到袋子内可使气体流入到袋子内以及从该袋子流出,同时使液体流入到袋子和从该袋子流出。这种端口还可以对在袋子内培养基或者培养物进行采样或者测试。通过利用任何理想的管道连接技术,管道、过滤器、连接件、探头采样器或者其他装置可连接到端口上。密封或者附着到袋子上的小囊和端口可与该袋子一起为一次性的。该袋子还可包括封接到袋子上可与该袋子一起为一次性的喷雾器(即反应器把空气喷射到培养基内的元件)。
具体地说,端口可结合到柔软袋子的任何位置上,以适应下面情况:

每个端口可具有连接到端口上的柔性管,培养基袋子、采样装置、过滤器、气体管路或者采收泵可通过无菌或者消毒连接件固定到该端口上。在一个实施例中,在袋子制造期间,该端口封接到柔软袋子上,并与该袋子组件一起灭菌。
可能用来与柔性管形成消毒连接的装置如下:
WAVE无菌管熔凝器
TERUMO无菌管焊接机
PALL KLEENPAK连接件
利用无菌技术在层流净化罩下进行的连接
利用金属管道和感应加热器进行的BAXTER Hayward专用的″热对热″连接
在另一个实施例中,固定到适当不锈钢阀门组件上的柔性管可单独灭菌(例如通过高压灭菌器),然后用作把一次性生物反应器连接到传统反应器或者加工管线上的路径。阀门组件用于把传统的现场蒸发(SIP)连接到传统反应器或者其他的过程中,而柔性管用于与在一次性反应器上的端口形成无菌或者消毒连接。
参见附图,图2示出了一种探头连接,其中该连接可用在本发明的搅拌罐反应器系统。在一个实施例中(如图2所示),探头(1)连接到柔软套筒(2)或者袋子上,该柔软套筒或袋子延伸到PALL连接件(3)的一半处。该PALL连接件(3)可连接到PALL连接件(5)的另一半,以在探头和搅拌罐反应器系统之间提供无菌连接。该PALL连接件(3)、(5)包括罩(4)和过滤器(7),以使连接部位无菌。无菌套管(6)从PALL连接件(5)的另一半伸出到搅拌罐反应器系统的反应器器皿(9)的反应器端口(8)。为了固定探头,该PALL连接通过去掉罩(4)、与连接件(3,5)配合、除去过滤器(7)以及把连接件的活动部分滑动到位而实现。随着柔软套筒或者袋子聚集或者压缩(10),探头传感器尖端(12)然后被推到反应器内。探头传感尖端(12)接着与反应器器皿(9)内部直接接触。夹子(11)围绕探头和套管放置,以把反应器内容物与PALL连接组件隔离密封。因此,当在PALL连接件(3,5)两个半份之间形成无菌连接时,柔软套筒(2)或者袋子被压缩(10),同时探头与培养物或者生产培养基接触。
在一个实施例中,探头单独地灭菌(例如通过高压灭菌器),然后通过无菌或者消毒连接固定于反应器上。例如,通过把探头(1)插入到PALL KLEENPAK连接件(3)的一半内,并利用如上所述以及图2所示的柔软套筒或者袋子(2)把该探头封接到连接件上,形成探头组件。该套筒从探头外端延伸到达PALL连接件的倒钩部。此组件被单独灭菌。PALL连接件(5)的另一半经由将容纳探头的柔性管(6)连接到反应器(9)上的端口(8)。此组件作为反应器的一部分灭菌。在第6,655,655号美国专利中详细描述了PALL连接件,在专利的内容在这里全部引入以参照。
D)培养物
搅拌罐反应器系统设计成能容纳例如生物流体、细胞培养基、微生物培养物、食品等等的流体培养基。当可流动培养基是细胞培养基时,该系统例如以批次模式、半批次模式、补料模式或者连续模式操作。一批培养物可以是大量细胞培养物,其中细胞培菌液在槽或者发酵器内以最大密度培养,并成批采收和处理。补料培养物可以是这样的批次培养物,其中配有新鲜营养素(例如生长限定基质)或者添加剂(例如产物母体)。连续培养物可以是这样的悬浮培养物,其中通过流入新鲜的培养基而连续地配有营养素,同时培养物体积通常是恒定的。同样地,连续发酵可参照这样的方法,其中在指数生长期(exponential growth phase),通过连续添加新鲜培养基,而这通过从生物反应器中去除细胞悬液而精确地平衡,细胞或者微生物保持在培养物内。此外,该搅拌罐反应器系统可用于悬浮物、灌注液或者微载体培养物。通常,该搅拌罐反应器系统可作为任何传统的搅拌罐反应器,其中带有任何种类的搅拌器,例如拉什顿(Rushton)、水翼艇、倾斜桨叶(pitched blade)或者舰船式(marine)。该搅动轴(12)可相对于壳体(11)以任何角度或者位置安装,例如在中心竖立、偏心竖立或者15度偏心。控制搅拌罐反应器可通过传统方式,而不需要就地蒸发(SIP)或者就地清洗(CIP)控制。实际上,本发明的系统不局限于无菌的生物反应器操作,而是可以用于任何操作中,其中清洁产物利用搅拌罐混合,例如不需要清洁空间的食品生产或者任何清洁空间的组合。
E)套件
本发明包括套件,其中该套件包括搅拌罐反应器系统和使用说明书。在一个实施例中,套件包括一次性搅拌罐反应器系统。因此,该套件包括至少一个例如袋子、轴、叶轮或者轴承的一次性元件。优选的是,整个套件是一次性的。柔软的、一次性袋子可通过至少一个密封件或者O形环固定到轴和轴承上,使得该袋子内部保持无菌。另外,该袋子包括pH值传感器和溶解氧传感器,其中传感器结合到袋子内并随袋子一起是一次性的。该套件还可包括一个或多个封接到袋子上的内部小囊。该小囊具有敞开到袋子外部的一端,从而探头可被插入反应器中。该探头例如可以是测温探头、pH值探头、溶解气体传感器、氧传感器、二氧化碳(CO2)传感器、细胞团块传感器、营养素传感器、渗透压力计等等。此外,该系统可在袋子内包括至少一个端口,用于使装置连接到该端口上,其中该装置包括但不局限于管道、过滤器、采样器、探头、连接件等等。端口可允许采样、滴定、化学稳定剂添加、喷射等等。此套件的优点在于,它可以有选择地全部是一次性的,并通过遵循附加的说明书而易于使用。此套件根据优选的培养物体积而具有不同的尺寸,并可与任何期望的反应腔或者桶一起使用。此套件预先灭菌,且不需要验证或者清洁。该套件能用于细胞培养物、微生物培养物、植物代谢物培养物、食品生产、化学试剂生产、生物制药等等。
在另一个实施例中,套件包括保持可丢弃袋子的壳体或者桶。这种壳体或者桶与袋子一起配备或者分别提供。
F)实例
以下具体的实例是用来说明发明,而不应该理解为限制权利要求的范围。
(i)一次性生物反应器
本发明的搅拌罐反应器系统的一个实例是一次性生物反应器。该生物反应器与带有嵌入搅动和可连接传感器(例如pH值传感器、温度传感器、溶解氧(dO2)传感器等等)的600升培养基袋子相似。该反应器通过传统的控制器操作。该搅拌器(例如搅动轴和叶轮)和轴承是一次性的,并可嵌入到袋子内。电机安装在容纳袋子的600升桶上的支撑件(例如电机和轴承座)或者支架上。在尺寸、形状和操作上,此生物反应器与带有无菌衬里的不锈钢反应器相似,然而,此发明的生物反应器与传统的不锈钢反应器相比具有许多优点。最重要的是,不需要清洁和蒸汽灭菌。该袋子通过辐射而预先灭菌,从而备用。实际上,在培养启动或者在培养进行之间不需要清洁、灭菌、验证或者测试。因此,该生物反应器在培养进行之间提供了零交叉污染的培养环境。在传统系统中,成本大部分与同步进行的清洁(CIP)和同步进行的蒸发(SIP)以及垫架(skid)的设计和监视这些功能的控制系统有关。这些成本在一次性生物反应器中削减掉了,同时多种产品可同时培养或者制造且这样的操作变得更容易了。
该一次性生物反应器可通过利用较大的培养袋子和较大的容纳该袋子的桶而很容易按比例增大。在不需要任何大量指导或者清洁情况下,可同时操作多个生物反应器。该生物反应器为具有良好混合特性的纯粹的搅拌罐。因而该生物反应器具有附加的优点是,它可按比例变化,同时如果需要的话,它的内容物可转移到不锈钢反应器中。值得注意的是,该生物反应器把容易使用与低成本和适应性结合起来,并因此提供了用于细胞培养的新技术平台。
(ii)细胞培养物
本发明的一次性生物反应器可用于一批培养物中,其中在该培养物中,细胞培植到新鲜的培养基中。当细胞生长时,它们消耗培养基中的营养素,同时废物积聚。对于分泌的产物,当培养按常规进行时,细胞通过过滤或者离心步骤从产物中分离。对于病毒体的生产,在培养物生长阶段期间,细胞感染病毒,使病毒体表达,然后采收。由于在生物反应器中为零交叉污染,因此对于成批培养物工作良好。
生物反应器还可以用于灌注培养,其中产物和/或废物培养基连续去除,同时去除的体积由新鲜培养基取代。当除去废物时,新鲜培养基的恒定添加为细胞提供了营养素,这些营养素要求获得较高的细胞浓度。不同于成批培养的经常变化条件,该灌注方法提供了获得和保持在平衡状态中培养的方式,其中细胞浓度和生产率可维持在稳定状态。这能够如同在任何传统不锈钢反应器中那样容易地在一次性袋子中实现。对于病毒体的生产,该灌注过程能使细胞浓度增加,从而使后感染病毒滴定度增加。对于分泌产物,通过只增加培养袋子的尺寸,在该生物反应器中的灌注为用户提供了增加生产率的机会。最重要的是,由于系统在生产过程中是零交叉污染,因此不需要灭菌、验证或者清洁。
在不脱离发明精神和范围情况下,本发明的各种修改和变形对本领域技术人员是显而易见的。尽管已经结合具体的优选实施例描述了发明,应当理解的是,要求保护的发明不应过分地限于这种具体的实施例。实际上,那些对本领域技术人员显而易见的用于实现发明的描述方式的各种修改,规定为在权利要求书的范围内。
Claims (42)
1.一种搅拌罐反应器系统,该系统包括:
(i)带有至少一个开口的柔软袋子,其中该袋子作为用于流体培养基的无菌容器;
(ii)设置在该袋子内的轴;
(iii)可连接到所述轴上的叶轮,其中所述叶轮用来搅动流体培养基,以提供流体动力环境;以及
(iv)连接到所述轴以及该袋子的所述开口上的轴承。
2.如权利要求1的搅拌罐系统,其中所述系统是一次性的。
3.如权利要求1的搅拌罐系统,其中所述系统是预先灭菌的。
4.如权利要求1的搅拌罐反应器系统,其中所述系统还包括在所述袋子外部的壳体,其中所述壳体包括至少一个容纳轴承和电机的支撑件,且其中所述袋子容纳在该壳体内。
5.如权利要求4的搅拌罐反应器系统,其中所述系统还包括从以下组中选择的加热器,该组包括加热板、蒸汽加热套和循环流体或者水加热器。
6.如权利要求5的搅拌罐反应器系统,其中所述加热器位于所述袋子和所述壳体之间。
7.如权利要求5的搅拌罐反应器系统,其中所述加热器结合到所述壳体内。
8.如权利要求4的搅拌罐反应器系统,其中所述壳体还包括多个挡板和围绕这些挡板折叠的袋子。
9.如权利要求4的搅拌罐反应器系统,其中所述系统还包括产物回路,其中液流经过pH值传感器和溶解氧传感器,其中所述传感器结合到所述壳体内。

13.如权利要求12的搅拌罐反应器系统,其中所述流体培养基是细胞培养基,且该系统以批次模式、半批次模式、补料模式或者连续模式操作。
14.如权利要求1的搅拌罐反应器系统,其中所述叶轮通过搅拌来搅动所述流体培养基。
15.如权利要求14的搅拌罐反应器系统,其中所述叶轮从以下组中选择,其中该组包括拉什顿、水翼艇、倾斜浆叶或者舰船式。
16.如权利要求1的搅拌罐反应器系统,其中所述袋子通过至少一个密封件或者O形环固定到所述轴和轴承上,使得该袋子内部保持无菌。
17.如权利要求16的搅拌罐反应器系统,其中所述密封件或者O形环固定到所述袋子上。
18.如权利要求16的搅拌罐反应器系统,其中所述袋子还包括pH值传感器和溶解氧传感器,其中所述传感器结合到该袋子内。
19.如权利要求16的搅拌罐反应器系统,其中所述系统还包括封接到所述袋子上的至少一个内部小囊,其中该小囊包括能够向该袋子外部敞开的一端,使得探头能够被插入到所述反应器中。
20.如权利要求19的搅拌罐反应器系统,其中所述探头从以下组中选择,该组包括测温探头、pH值探头、溶解气体传感器、氧传感器、二氧化碳传感器、细胞团块传感器、营养素传感器和渗透压力计。
21.如权利要求16的搅拌罐反应器系统,其中所述系统还包括在所述袋子中的至少一个端口,允许装置连接到该端口上,其中所述装置从以下组中选择,该组包括管、过滤器、采样器、探头和连接件。
22.如权利要求21的搅拌罐反应器系统,其中所述端口允许气体进入所述袋子内并从其中排出。
23.如权利要求21的搅拌罐反应器系统,其中所述端口允许培养基流入所述袋子内并从其中排出。
24.如权利要求21的搅拌罐反应器系统,其中所述端口允许采样、接种、滴定、添加化学稳定剂和喷射。
25.一种用于制备搅拌罐反应器系统的方法,该方法包括:
(i)提供带有至少一个开口的柔软袋子,其中该袋子作为用于流体培养基的无菌容器;
(ii)将其上连接有叶轮的轴插入到所述袋子内,其中所述叶轮用来搅动流体培养基,以提供流体动力环境;
(iii)将轴承连接到所述轴以及所述袋子的所述开口上;以及
(iv)将所述袋子封接到所述轴和轴承上,使得该袋子内部保持无菌。
26.如权利要求25的方法,其中所述搅拌罐反应器系统是预先灭菌的。
27.如权利要求25的方法,其中所述搅拌罐反应器系统是一次性的。
28.如权利要求25的方法,其中所述搅拌罐反应器系统包括至少一个从以下组中选择的一次性元件,该组包括所述袋子、轴、叶轮和轴承。
29.一种套件,包括:
(i)如权利要求16的搅拌罐反应器系统;以及
(ii)使用说明书。
30.如权利要求29的套件,其中所述搅拌罐反应器系统是一次性的。
31.如权利要求29的套件,其中所述搅拌罐反应器系统包括至少一个从以下组中选择的一次性元件,该组包括所述袋子、轴、叶轮和轴承。
