Description
The present invention relates to a novel package having improved properties for lozenges containing ambroxol.
Hintergrund der ErfindungBackground of the invention
Ambroxol is also known as a locally acting in the pharynx drug form in the form of a lozenge.
In order to achieve optimum shelf life of the product ambroxol-containing lozenges, the packaging material is of particular importance. In addition to the protective properties of the packaging, it is also necessary that the product is also packed patient-friendly. Usually suitable as packaging blister packs by means of which individual tablets are packaged separately in individual blister cups, which in turn represent a blister strip or blister card. The blister packaging ensures the chemical and physical stability of the drug formulation. In addition, the blister pack is subject to the function of fulfilling further, product-specific requirements, so that this can also be regarded as patient-friendly. Specific requirements here are that the packaging material is child-resistant, senior-friendly, and that the packaging material is not changed by the product to be packaged in its properties during storage.
Stand der TechnikState of the art
Blister for medicines consist of a bottom foil or bottom plate, the more z. B. has circularly arranged cavities. The individual cavities each contain one tablet of the drug. Typically, a blister pack is understood to mean a cold-formed or thermoformed blister bottom portion containing cavities for receiving a drug, e.g. As the tablets contains, wherein the bottom part is sealed with a cover sheet.
The cover film and the bottom film can also be composed of one or more layers of different or the same materials. The cover sheet is z with the bottom sheet. B. connected by gluing, welding or sealing sealed. The cover film and / or the carrier film is usually formed as a metal and / or plastic and / or paper foil. These materials can be present in multiple layers. Typical metal foils include, for example, aluminum foils and aluminum composite foils made of aluminum and e.g. B. are made of a plastic. As the material for the plastic films may polyvinyl chloride (PVC) cycloolefin copolymer (COC), polychlorotrifluoroethylene (PCFE), polyethylene (PE), polypropylene (PP), polyethylene terephthalate (PET), polycarbonate (PC), polyester (UP), polyacrylate, polyamide (PA) or other plastics. Often a blister consists of a cover foil of aluminum and a bottom foil for receiving the pharmaceutical product in the cavities of the bottom foil.
This deep-drawn bottom foil may also be an aluminum foil. Likewise, the bottom foils of polyvinyl chloride (PVC), polyvinylidene chloride (PVDC), polypropylene (PP), polyethylene terephthalate (PET), polyethylene (PE) and composite films such as PVC / ACLAR ® (PCTFE), PVC / PVDC and COC or FORMPACK ® (Al -Al blisters) exist.
In a specific embodiment, blisters can be designed in the form of a push-through blister. Under a Durchdrückblister is to understand a blister in which the individual removal of drugs from the blister, such. As capsules, tablets and dragees by expressing the tablets, capsules or dragees containing wells (wells) can be removed.

The polyethylene glycols which can be used in the formulations of ambroxol-containing lozenges suitably have a molecular weight in an interval of 3,500 to 20,000. Such a polyethylene glycol is known under the name Macrogol 6000 from the prior art.
The preferred talcum used is generally understood as meaning a - widespread - hydrated magnesium silicate of the composition Mg 3 [(OH) 2 / Si 4 O 10 ] or 3 MgO 4 .SiO 2 . However, it is also possible to use another pharmaceutically suitable phyllosilicate.
The proportion of talc in one of the lozenge according to the invention with a total mass of 1.5 g is in a range of 5 to 100 mg, preferably 20 to 80 mg, more preferably 30 to 60 mg and most preferably 60 mg.
Conveniently, the proportion of active ingredient in a lozenge in a range of 1 to 50 mg, preferably 5 to 30 mg and particularly preferably in an interval of 10 to 25 mg of ambroxol in the form of its hydrochloride
Tablets according to formulations A and C are prepared by conventional direct compression technology.
According to the invention, it has been found that the packaging material for ambroxol-containing lozenges, in the form of a blister, fulfills the object according to the invention if the cover film is selected such that the push-through force for removal of the tablet ranges between 40 N and 160 N, particularly preferably between 50 N and 150 N, and most preferably between 60 N and 100 N. The test of the push-through property takes place mechanically. In this case, a semicircular punch is pressed onto the inside of the film clamped in a holder at a defined speed. The force required to push through is given in Newtons.

For the purposes of the present invention, therefore, child safety and senior citizen friendliness are understood to be met if the requirements of the test according to US 16 CFR § 1700.20 are met.
According to the invention, the requirements ("child-resistant packaging") are fulfilled if two tests with children are successfully carried out as part of the child restraint according to the guideline US 16 CFR § 1700.20. The first test includes the question of the extent to which children (sample: 100 children aged 42-51 months) can access the product from the package within 5 minutes before demonstrating the opening principle. The second test includes the question of the extent to which children (sample: 100 children aged 42-51 months), after demonstrating the opening principle, can access the product from the packaging within 5 minutes. The test evaluation is based on an F-classification (failure rate) of F = 9 in the present invention. An opening is defined as the withdrawal of nine tablets or more. The packaging according to the invention fulfills these requirements and has the following result: 0 (zero) openings before the demonstration (test 1) and 0 (zero) openings after the demonstration (test 2). According to the invention, the requirements ("elderly-friendly packaging material") are fulfilled if 100 subjects (adults, 25% between 50 and 54 years, 25% between 55 and 59 years and 50% between 60 and 50 years) are included in the seniors-friendly test 70 years) a maximum of 10% unsuccessful opening attempts are observed. The packaging according to the invention has an efficiency (openings) of 99% and thus meets the requirements of suitability for seniors.
