Abstract
Description
Reaktive Sauerstoffspezies (ROS), z.B. Reactive oxygen species (ROS), e.g. superoxide (O 2 .– ), hydrogen peroxide (H 2 O 2 ), hydroxyl radicals (OH . ), And reactive nitrogen species (RNS), especially nitrogen oxide (NO) and peroxynitrite (ONOO - ), are known as causes or as accompanying factors of a variety of diseases and age-related disorders, which can be directly or indirectly attributed to cell stress. Typical examples are primarily degenerative diseases of the brain, for example Parkinson's disease and Alzheimer's disease, or subsequent conditions after a stroke, after brain trauma or during neurosurgical interventions, but also damage in connection with heart attack, skin burns, reperfusion after transient ischemia and the like. Like. (1, 3, 10). In the conditions described, the free radical nitrogen oxide (NO) and other radicals such as superoxide (O 2 .– ) play a special role (7, 28, 31). NO is a very reactive, gaseous radical produced by the cells themselves, which can trigger pathological changes in overproduction. It has been shown to play a causal role in the CNS diseases mentioned above (30, 18, 4). NO, like many other potentially damaging biomolecules, is produced in the nervous system by both neurons and glial cells, especially the microglial cells. Microglial cells are macrophage-related cells that are activated in the nervous system, for example, by injury, infection or inflammation, and are used there for immune defense and to break down dead cells. After induction, NO is generated outside the nervous system by a wide variety of cell types, including endothelial cells and macrophages.
NO has a wide range of physiological and pathophysiological effects. There is extensive evidence for the involvement of NO and peroxynitrite, the adduct of superoxide (O 2 .– ) and NO, in processes that lead to apoptosis as a nitrosative stress factor in a wide variety of cell types (27, 33). NO production and mitochondrial dysfunction are causally related (4, 13), although the individual mechanisms of action have so far only been partially elucidated.
(6, 12, 30).A range of substances such as vitamin C, vitamin E, Trolox ® (±) -6-hydroxy-2,5,7,8-tetramethyl-chroman-2-carboxylic acid), melatonin, resveratrol (traps-3,4 ' , 5-trihydroxystilben), as well as other natural substances with antioxidative properties have already been tested in such a context, although their use has remained essentially limited to health care products or has been rather exceptional (e.g. (6, 12, 30).
Has received a lot of attention recently in connection with the so-called "French Paradox", the antioxidant Resveratrol excited (19-22, 35, 40).
Ferner weisen in vivo-Studien aus, dass Resveratrol eine protektive Wirkung auf das kardiovasculäre Sytem hat und die Risiken der Arteriosklerose verringert (29).Resveratrol is a natural polyphenolic stilbene derivative, which is used in red wine, occurs in pigeon cores and plant extracts. epidemiological and in vitro studies have shown that moderate consumption of red wine is natural Protection against coronary artery disease and cancer offers just that so-called "French paradox" (20, 35, 40). The biological properties of resveratrol have been extensively studied in recent years to use it as a pharmaceutical (11). Resveratrol works z. B. anti-inflammatory by inhibiting the arachidonic acid cascade (24, 26, 36) and by inhibiting NFkB (23, 38). There are Evidence suggests that resveratrol is neuroprotective both in vitro and in vivo Features (2, 18, 37, 39). Also show in vivo studies from that resveratrol has a protective effect on the cardiovascular system and reduces the risk of arteriosclerosis (29).
Unfortunately, resveratrol is relatively poor solubility in aqueous or protic solvents (e.g. aqueous alcohol, Propylene glycol e.c.), leading to prophylactic use of the substance or therapeutic purposes. There are also hints the literature suggests that resveratrol has a relatively high proliferative cytotoxicity in vitro Cells shows (8, 9, 17). Through our own investigations on a microglia cell line we confirm this finding.
Problemproblem
One way to get through ROS and RNA caused cellular damage, like you for Nerve tissue, heart muscle, kidney and many other tissue and Organ systems have been described to limit it is a targeted one pharmacological intervention using antioxidants.
The problem was a substance or To find substance class that the oxidative / nitrosative cell stress preferably comprehensively reduced. The substance should be as natural as possible Raw materials are isolated and should be easily and inexpensively accessible. additionally the substance / substance class in question should have good solubility properties have a stable active ingredient formulation and easy application to ensure.
- 1.) as a radical scavenger (narrow. Scavenger), which minimizes the overproduction of reactive oxygen or nitrogen species (ROS and RNS) and thus the oxidative / nitrosative cell stress.
- 2.) as an inhibitor of metabolic processes that lead to the formation of ROS / RNS, for example inhibition of the inducible nitrogen oxide synthase activity (iNOS) and thus leads to a reduction in NO production.
- 3.) as a blocker of the expression of enzymes relevant for point 2. on the DNA transcription / translation level.
- 4.) with specific cytotoxicity towards the body's own defense cells, which are essentially the production sites for ROS / RNS (eg for the CNS, these are the microglial cells). The general toxicity must be as low as possible.
The parameters of Medical indications could surprisingly be found with Oxyresveratrol a representative of the class of tetrahydroxystilbene can be improved. In the present invention, for the class of tetrahydroxystilbene derivatives, a new application using the example of oxyresveratrol (structural formula I) described, namely use as a neuroprotective, especially in treatment and prophylaxis of brain dysfunction and stroke. Under Neuroprotective means a substance that has the ability possesses nerve cells and other brain and peripheral cells Nervous system from damage to protect a wide variety of genesis or mortality. For drugs with neuroprotective Effect there is a great demand worldwide for therapy and prophylaxis of diseases associated with nerve cell death go along in the brain.
We have the naturally occurring for our investigations Stilbene oxyresveratrol (trans-2,3 ', 4,5'-tetrahydroxystilbene) the wood of the mulberry tree (Morus alba L.) isolated. The wood of the Mulberry tree in which oxyresveratrol is contained to about 2%, is easily accessible as a raw material. Oxyresveratrol is also important in others for natural medicine Contain plants of different genera, e.g. in Maclura, Artocarpus and Chlorophora (5, 14, 15, 25, 34). Oxyresveratrol showed in very good solubility properties in our applications in watery and ethanolic media and can therefore also be formulated in high concentration and applied, with what in the present inventive application the disadvantages of resveratrol are avoided. Is oxyresveratrol an independent Product of secondary plant metabolism, which differs fundamentally from the secondary products of resveratrol oxidation. We were also able to show that, surprisingly, oxyresveratrol - as a single component used - excellent has antioxidant properties that exceed that of resveratrol, whereas the cytotoxicity this connection is significantly lower.
Darstellung der Erfindungpresentation the invention
Object of the present invention is the use of oxyresveratrol (formula I) or extracts from Mulberry wood (e.g. Morus alba) in formulations as a protective agent against damage that are mediated by oxidative / nitrosative stress, whereby we have demonstrated the effectiveness experimentally using various damage models of the nervous system. The neuroprotective formulations promise a success not previously achieved by other substances the prophylaxis or treatment of brain trauma, cerebrovascular ischemia or Hypoxia, as well as primary neurodegenerative diseases and many other neuronal disorders, for this these states It is generally accepted that they are oxidative or nitrosative stress as causal Factor. Use preoperatively is also promising in neurosurgical interventions, but also in other organs or organ systems in which oxidative or nitrosative cell stress is the cause or secondary to disturbances or pathological conditions leads.

Oxyresveratrol (Mp. = 199–200°C, Mp.Lit. = 201°C (24)) wurde dünnschichtchromatographisch (stationäre Phase: Kieselgel 60; mobile Phase: CHCl3/MeOH /Ameisensäure = 75/16.5/8.5; Detektion: Anisaldehyd-H2SO4-Reagenz, Rf = 0.205) bzw. durch HPLC geprüft. Die 1H- und 13C-NMR-Daten des gewonnenen Oxyresveratrol entsprachen denen der Literatur (16, 32).500 g of mulberry wood shavings (Morus alba L.) were extracted with 6.0 liters of ethanol for two days at room temperature. The chips were then filtered off and the solvent was distilled off under reduced pressure. The syrupy residue obtained (25.2 g) was dissolved in 50 ml of methanol and then pre-cleaned by vacuum liquid chromatography (VLC, 70 g silica gel 60, TLC grade, MERCK). The chromatography column was eluted with chloroform / methanol with an increasing gradient (70/30 to 50/50). 430 ml of the eluate were collected and the solvent was distilled off. The residue was then refluxed with 800 ml of boiling chloroform (with 2.5% ethanol) and the extract was left in the refrigerator overnight. The crystallized oxyresveratrol was filtered off, washed with a little chloroform and dried in vacuo. Yield: 1.71 g (0.34% calculated on the amount of drug used). The purity of Oxyresveratrol (Mp. = 199-200 ° C, Mp. 5. Neuroprotektion im Hirninfarktmodell5. Neuroprotection in the brain infarction model
Nevertheless, the results of the Cell culture studies give no direct conclusions as to whether oxyresveratrol too is effective in vivo. A logical step in these results was therefore testing the protective effect of oxyresveratrol in one in vivo damage model of the central nervous system. For this, a common brain infarction model, the occlusion of the cerebral artery (“middle cerebral artery occlusion model ") followed by reperfusion on the rat, the neuroprotective effect of the substance by infarct size and by the effect on behavioral deficits was measured after the insult.

The surprisingly strong neuroprotective Effect of oxyresveratrol in this stroke model is an example for various neurodegenerative diseases because of the causative cell damage free radicals is common.
