- TECHNISCHES GEBIET DER ERFINDUNG
- Die Erfindung betrifft schnell lösliche, oral konsumierbare Filme. Die Filme werden zur Freisetzung Atem erfrischender Wirkstoffe, antimikrobieller Wirkstoffe und die Speichelsekretion anregender Wirkstoffe in der Mundhöhle verwendet. Die Filme können auch zur Freisetzung von pharmazeutischer Wirkstoffe verwendet werden.
- HINTERGRUND DER ERFINDUNG
- Unter idealen Umständen würden sich die Menschen im Rahmen ihrer gewohnten Mundhygiene nach jeder Mahlzeit den Mund gründlich reinigen. Leider verhindern verschiedene Faktoren, dass dieses grundlegende Erfordernis einer guten Mundhygiene auch weitestgehend erfüllt wird.
- Manchmal kann die Reinigung des Mundes schwierig oder unbequem sein, je nach Art der Reinigung und in Abhängigkeit von der Situation, in der die Reinigung erfolgen muss. Bürsten, Reinigung der Zähne mit Zahnseide, Reinigung der Zunge und Gurgeln unter Verwendung verschiedener Hilfsmittel und Pflegemittel sind die üblichen, für die häuslichen Umgebung geeigneten Mundpflegepraktiken. Jedoch sind die Hilfsmittel und Pflegemittel zur Mundreinigung ausserhalb des Hauses, wo eventuell keine entsprechenden oder nur unzureichende sanitäre Gegebenheiten vorliegen, weniger bequem anzuwenden.
- Da Bürsten, Reinigung der Zähne mit Zahnseide, Reinigung der Zunge und Gurgeln in der Öffentlichkeit in vielen, wenn nicht in allen Kulturen nicht als sozial akzeptables Verhalten angesehen werden, ist eine Reihe von Produkten zur weniger auffälligen Mundreinigung entwickelt worden. Dazu gehören Atem erfrischende Kaugummis und Pastillen. Wenn auch die Kaugummis und Pastillen so formuliert sind, dass verschiedene positive Wirkungen erzielt werden, sind sie doch nicht immer sozial akzeptabel.
- Beispielsweise ist Kaugummi in bestimmten Institutionen wie z.B. Schulen, und in bestimmten Ländern, wie z.B. Singapur, ausdrücklich verboten. Kaugummi und Pfefferminzpastillen werden über längere Zeit verwendet und erfordern Lutsch- und Kauaktivität von Seiten des Verbrauchers, die ablenkend, langweilig und unerwünscht sein kann.
- Ein weiteres tragbares Produkt zur Mundreinigung ist das Mundspray. Wie die Mundwäsche kann ein Mundspray dem Anwender rasch eine starke Atem erfrischende Wirkung liefern, die bei Produkten mit längerer Anwendung wie Kaugummis oder Pastillen ausserordentlich intensiv sein kann. Andererseits sind Mundsprays auffällig. Das Sprühen eines Mundsprays ist normalerweise mit einem Geräusch verbunden, das in unerwünschter Weise die Aufmerksamkeit der Umgebung auf den Anwender lenkt. Ausserdem sind Mundsprays im Allgemeinen in relativ teuren und komplizierten Metallbehältern verpackt, die beim Gebrauch verstopfen können und nicht umweltfreundlich sind. Weiter verschwendet versehentliches Sprühen in falscher Richtung nicht nur das Produkt, sondern kann auch zu Augenreizungen, einem klebrigen Gesicht und/oder verschmutzter Kleidung führen.
- Es wurde vorgeschlagen, zur unauffälligen Verabreichung Atem erfrischender Wirkstoffe essbare Filme einzusetzen, siehe JP 5-236885. Diese japanische Patentanmeldung lehrt jedoch nicht die Einbeziehung antimikrobieller Wirkstoffe in den Film, die Verwendung des Films zur Reduzierung der Menge unerwünschter Bakterien in der Mundhöhle oder seine Verwendung zur Förderung der Speichelsekretion. Weiter offenbart diese Patentanmeldung nicht die Verwendung des Films für andere Zwecke als zur Atemerfrischung oder an anderen Orten als im Mund.
- Das US-Patent Nr. 5,518,902 von Ozaki et al. (Hayashibara) offenbart Produkte mit hohem Pullulangehalt wie essbare Filme, Zahnpasten und pharmazeutische Produkte (Spalte 3, Zeilen 44–56 und Beispiel B-8). Die Produkte können neben Pullulan noch verschiedene weitere Bestandteile enthalten, beispielsweise weitere Polysaccharide, mehrwertige Alkohole, Antiseptika und Geschmacksstoffe (Spalte 4, Zeile 58 bis Spalte 5, Zeile 11). Keines der etherischen Öle wie Thymol, Eucalyptol, Methylsalicylat oder Menthol ist als geeigneter Bestandteil genannt.
- Das US-Patent Nr. 5,411,945 von Ozaki et al. (Hayashibara) offenbart Pullulan als Bindemittel und damit hergestellte Produte, u.a. essbare Filme (Beispiel B-2). Die Produkte können neben Pullulan noch verschiedene weitere Bestandteile enthalten, beispielsweise weitere Polysaccharide, antibakterielle Wirkstoffe, Geschmacksstoffe und pharamazeutische Wirkstoffe (Spalte 4, Zeilen 5–15). Keines der etherischen Öle ist als geeigneter Bestandteil genannt.
- Das US-Patent Nr. 4,851,394 von Kubodera offenbart essbare Filme aus Glucomannan und mehrwertigem Alkohol, die Pullulan enthalten könen (Spalte 3, Zeile 59 bis Spalte 4, Zeile 21). Die Filme werden mit bekannten Filmen auf Basis von Pullulan verglichen, die nicht wasserbeständig sein sollen (Spalte 1, Zeilen 40–44). Kein etherisches Öl ist als geeigneter Bestandteil genannt.
- Das US-Patent Nr. 3,784,390 von Hijiya et al. offenbart Filme aus Pullulan und deren Verwendung in Beschichtungs- und Verpackungsmaterialien für Lebensmittel, Arzneimittel und andere sauerstoffempfindliche Materialien. Zu allen Beispielen dieses Patents wird Pullulan in heißem Wasser gemischt.
- Das US-Patent Nr. 4,623,394 von Nakamura et al. offenbart einen allmählich abbaubaren Formkörper, der einen mit Pullulan hergestellten Film darstellen kann. Die Formkörper enthalten ein bestimmtes Heteromannan, das Johannisbrotkernmehl sein kann.
- Das US-Patent Nr. 4,562,020 von Hijiya et al. offenbart ein Verfahren zur Herstellung eines selbsttragenden Films aus einem Glucan, das Pullulan darstellen kann.
- Das japanische Patentdokument JP 5-1198 offenbart aus Polyvinylalkohol und wenigstens einem aus Carrageenan, wasserlöslicher Cellulose-α-Stärke und wasserlöslichen Polysacchariden ausgewählten Bestandteil hergestellte Filme.
- Die WO 99/17753 offenbart schnell lösliche Filme zur Verabreichung von Arzneimitteln, die im Verdauungstrakt adsorbiert werden sollen.
- Die WO 98/26780 offenbart ein flaches folien-, papier- oder waffelartiges Produkt zur Anwendung und Freisetzung von Wirkstoffen in der Mundhöhle. Der in der WO 98/26780 konkret genannte Wirkstoff ist Buprenorphin.
- Die WO 98/20862 offenbart einen Film zur Verwendung in der Mundhöhle, der einen kosmetischen oder pharmazeutischen Wirkstoff enthalten kann.
- Die WO 98/26763 offenbart ein flaches folien-, papier- oder waffelartiges Produkt zur Freisetzung von Wirkstoffen in der Mundhöhle. Der konkret genannte Wirkstoff ist Apomorphin.
- Obwohl schnell lösliche oral konsumierbare Filme vorbekannt sind, so sind diese Filme und die Verfahren zu deren Herstellung doch noch zu verbessern.
- Auf alle in diesem Dokument genannten Literaturstellen wird hiermit vollinhaltlich Bezug genommen.
- KURZBESCHREIBUNG DER ERFINDUNG
- Die Erfindung betrifft einen physiologisch akzeptablen Film, der besonders gut geeignet ist, im Mund der ihn einnehmenden Person zu haften und sich darin schnell aufzulösen. Der Film besteht aus einer einzigen Schicht, die wenigstens ein wasserlösliches Polymer und eine antimikrobiell wirksame Kombination wenigstens zweier etherischer Öle aus der Gruppe (i) 0,01–4 Gew.% Thymol, (ii) 0,01–4 Gew.% Methylsalicylat, (iii) 0,01–4 Gew.% Eucalyptol und (iv) 0,01–15 Gew.% Menthol enthält. Gemäß einer ersten Ausführungsform der Erfindung dient der Film zur Verabreichung wenigstens eines Mundpflegemittels wie z.B. eines antimikrobiellen Wirkstoffs oder eines Mittels zur Anregung der Speichelsekretion. Die antimikrobiellen Wirkstoffe wirken gegen Keime, die Halitosis, Zahnbelag und Zahnfleischentzündungen verursachen. Die die Speichelsekretion anregenden Wirkstoffe wirken gegen den als Xerostomie oder Mundtrockenheit bekannten Zustand. Außerdem sind die Mundpflegefilme Atem erfrischend und wirken gegen Mundgeruch. Der filmbildende Stoff, der zur Herstellung der erfindungsgemäßen Filme verwendet wird, hält die mundpflegenden Wirkstoffe in der Mundhöhle und gewährleistet Langzeitwirkung.
- Gemäß einer zweiten Ausbildungsform dient der schnell lösliche Film als Träger zur oralen Verabreichung eines pharmazeutischen Wirkstoffs durch eine Schleimhaut oder eine offene Wunde eines Patienten.
- Die Erfindung betrifft auch ein Verfahren zur Herstellung eines geschmeidigen, nicht an sich selbst haftenden Films, der insbesondere zur oralen Verabreichung geeignet ist. Das Verfahren umfasst das Mischen eines filmbildenden Mittels und wenigstens eines Stabilisators unter Erhalt einer filmbildenden Mischung, Auflösen wasserlöslicher Bestandteile in Wasser unter Erhalt einer wässrigen Lösung, Vereinigen der filmbildenden Mischung und der wässrigen Lösung unter Erhalt eines hydrierten Polymergels, Mischen der Öle unter Erhalt einer Ölmischung, Zugabe der Ölmischung zum hydrierten Polymergel und Mischen unter Erhalt eines einheitlich emulgierten Gels, Verstreichen des einheitlichen Gels auf ein Substrat und Trocknen des verstrichenen Gels unter Erhalt eines Films.

- Beschreibung der Zusammensetzungen für Mundpflegefilme
- Die erste Ausbildungsform der Erfindung besteht in einem physiologlisch akzeptablen Film, der besonders gut geeignet ist, im Mund der ihn einnehmenden Person zu haften und sich darin aufzulösen zur Verabreichung eines antimikrobiellen Wirkstoffs, der Halitosis, Zahnbelag und Zahnfleischentzündungen verursachende Keime abtötet. Somit kann der Film ein wirksames Mittel zur Prävention und Behandlung von Halitosis, der Entstehung von Zahnbelag, der Entstehung von Zahnstein und Zahnfleischentzündungen darstellen. Der Film enthält bevorzugt Pullulan, Thymol, Methylsalicylat, Eucalyptol und Menthol.
- Die Mundwäsche LISTERINE® ist vielleicht das bekannteste Beispiel einer antiseptischen oralen Zusammensetzung, die nachweislich Mikroben in der Mundhöhle abtötet, die für Zahnbelag, Zahnfleischentzündungen und schlechten Atem verantwortlich sind. Die Mundwäsche LISTERINE® erzielt ihre antimikrobielle Wirkung durch eine Kombination etherischer Öle, die in die Mikroorganismen eindringen und sie abtöten. Diese etherischen Öle umfassen genau abgestimmte Mengen Thymol, Methylsalicylat, Menthol und Eucalyptol (im Folgenden "die etherischen Öle") in einer hydroalkoholischen Lösung. Viele Mundgeruch verursachende Bakterien leben in Vertiefungen und Furchen auf der Zungenoberfläche. Listerine® Antiseptische Mundwäsche reduziert Mundgeruch auf Grund des hohen Gehalts an antimikrobiellen Wirkstoffen in einem flüssigen Medium, das leicht in diese Vertiefungen und Furchen eindringen kann. Dies wäre mit einer festen Darreichungsform, die geringe Mengen dieser antimikrobiellen Wirkstoffe enthält, nicht möglich. Jedoch erreicht der erfindungsgemäße konsumierbare Film in seiner bevorzugten Ausführungsform einen Großteil der hygienischen Vorteile und der Wirkung beim Verbraucher wie LISTERINE® Mundwäsche, bei einer leichter mitzuführenden und unauffälliger anzuwendenden Form.
- Ein bedeutendes Problem bestand darin, die gemeinsame Wirkung der etherischen Öle und den relativ hohen Ölgehalt von LISTERINE® Mundwäsche in einem Film beizubehalten. Die Erfinder haben dieses Problem jedoch mit dem erfindungsgemässen Film gelöst.
- Ein weiteres Merkmal der Erfindung besteht darin, dass, während in LISTERINE® die Menge an etherischen Ölen für die Einarbeitung in einen Film relativ groß ist, der erfindungsgemäße Film doch eine geringere Gesamtmenge an etherischen Ölen pro Dosierungseinheit enthält im Vergleich zur Menge der LISTERINE® Mundwäsche bereitstellt. Dennoch weist der Film in überraschender Weise in der Mundhöhle eine antimikrobielle Wirkung auf. Die Erfinder gehen davon aus, dass der bevorzugte filmbildende Bestandteil Pullulan auf den Oberflächen im Mund eine dünne Schicht bildet, die die geringe Menge an etherischen Ölen, die in die Vertiefungen und Furchen der Mundhöhle eindringen können, festhält und so eine lang anhaltende antimikrobielle Wirkung gewährleistet.
- Wenn auch nach Kenntnis der Erfinder zur Zeit kein anderer Atem erfrischender konsumierbarer Film bekannt ist, der antimikrobielle Wirkung aufweist, so gibt es doch einen konsumierbaren Film, offenbart in JP 5-236885, der angeblich Atem erfrischende Wirkung aufweist, wobei jedoch keine Bestandteile mit nennenswerter antimikrobieller Wirkung erwähnt sind. Außerdem lehrt die JP 5-236885, dass der Film Geschmacksstoffe und Extrakte in einer Menge von 5 bis 7 Gew.% enthält, wobei der Geschmacksstoff als Öl beigegeben wird (die etherischen Öle sind nicht offenbart), wohingegen der erfindungsgemäße Film bevorzugt einen Ölgehalt von wenigstens 10 Gew.%, bevorzugt 15 Gew.% bis 30 Gew.%, besonders bevorzugt 15 Gew.% bis 25 Gew.%, aufweist. Sofern in den Beispielen nicht anders angegeben, sind die Mengen der Öle und anderen Bestandteile des Films in Gew.% angegeben, bezogen auf das Gewicht nach dem Trocknen der Filmformulierung zur Bildung des Films.
- Die Mengen der einzelnen in den Filmzusammensetzungen verwendeten etherischen Öle sind variabel, solange sie für eine antimikrobielle Wirkung ausreichend sind. Allgemein beträgt die Menge an Thymol, Methylsalicylat und Eucalyptol zwischen 0,01 und 4 Gew.% der Filmzusammensetzung, bevorzugt 0,50 bis 3,0 Gew.%, besonders bevorzugt 0,70 bis 2,0 Gew.% des Films. Menthol kann in in einer Menge von 0,01 bis 15 Gew.% der Zusammensetzung, bevorzugt 2,0 bis 10 Gew.% und besonders bevorzugt 3 bis 9 Gew.% des Films zugegeben werden. Die zu verwendenden Mengen können durch den Fachmann leicht bestimmt werden und können diese Mengen übersteigen, so lange der Gesamtgehalt an Öl keine Probleme bezüglich Klebrigkeit und Verarbeitbarkeit hervorruft. Gemäß bestimmten Ausführungsformen liegen die etherischen Öle in Mengen mit synergistischer Wirksamkeit vor, die die Plaque bildenden Keime, die Zahnbelag, Zahnfleischentzündungen und Mundgeruch hervorrufen, abtöten.
- Ein Hauptproblem bei der Formulierung eines Films mit einem solch relativ hohen Ölgehalt besteht darin, dass bei einer einfachen Erhöhung der Ölmenge im Film ohne Bestimmung der genauen Verhältnisse der zahlreichen weiteren Bestandteile normalerweise ein Film erhalten wird, der zu feucht und daher schwer handhabbar oder verarbeitbar ist. Die Erfinder haben entdeckt, wie ein Film mit hohem Ölgehalt hergestellt werden kann, der feucht genug ist, um nicht spröde zu sein, aber nicht so feucht ist, dass er sich in unerwünschter Weise schleimig anfühlt oder stark an benachbarte Filme anhaftet. Somit kann der erfindungsgemäße nicht selbstanhaftende Film in Kontakt mit einem weiteren solchen Film gelagert werden (beispielsweise in einem Stapel) oder aufgewickelt werden (beispielsweise auf einer Spule), ohne dass ein das Anhaften verhinderndes Mittel (z.B. ein Plastikfilm, Papier oder ein anderer Träger) zwischen benachbarte Filmteile gelegt werden muss.
- Der in den erfindungsgemäßen Filmen verwendete filmbildende Bestandteil kann ausgewählt sein aus Pullulan, Hydroxypropylmethylcellulose, Hydroxyethylcellulose, Hydroxypropylcellulose, Polyvinylpyrrolidon, Carboxymethylcellulose, Polyvinylalkohol, Natriumalginat, Polyethylenglycol, Xanthangummi, Traganth, Guargummi, Akariengummi, Gummi arabicum, Polyacrylsäure, Methylmethacrylat-Copolymer, Carboxyvinyl-Polymer, Amylose, stark amylosehaltiger Stärke, hydroxypropylierter stark amylosehaltiger Stärke, Dextrin, Pectin, Chitin, Chitosan, Levan, Elsinan, Collagen, Gelatine, Zein, Gluten, Sojaproteinauszug, Molkeproteinauszug, Casein und deren Mischungen. Ein bevorzugter filmbildender Stoff ist Pullulan in einer Menge von 0,01 bis 99 Gew.%, bevorzugt 30–80 Gew.%, besonders bevorzugt 45–70 Gew.%, ganz besonders bevorzugt 60–65 Gew.% des Films.
- Der erfindungsgemäße Film enthält bevorzugt Pullulan als filmbildenden Bestandteil und die etherischen Öle als antimikrobielle Stoffe und Geschmacksstoffe und kann weiter Wasser, weitere antimikrobielle Stoffe, weitere filmbildende Stoffe, Weichmacher, weitere Geschmacksstoffe, Schwefel ausfällende Stoffe, die Speichelsekretion anregende Stoffe, Kühlmittel, Tenside, Stabilisatoren, Emulgatoren, Verdickungsmittel, Bindemittel, Farbstoffe, Süßstoffe, Duftstoffe u.ä. enthalten.
- Auf Grund des relativ hohen Ölgehalts des Mundpflegefilms werden bevorzugt wesentliche Mengen an Feuchthaltemittel im Film vermieden (besonders bevorzugt werden im Film keine Feuchthaltemittel verwendet), um einen übermäßig feuchten, selbstanhaftenden Film zu vermeiden. Insbesondere wird der Film bevorzugt mit einem Weichmacher formuliert, der von Glycerin, das auch ein Feuchthaltemittel darstellt, verschieden ist, sowie mit einem Süßstoff, der von Sorbit, einem schwachen Feuchthaltemittel, verschieden ist.
- Schwefel ausfällende Stoffe, die Mundgeruch verringern, können ebenfalls den erfindungsgemäßen Mundpflegefilmen beigegeben werden. Diese Stoffe binden und desaktivieren die flüchtigen Schwefelverbindungen, die einen Großteil des Mundgeruchs verursachen. In der Erfindung verwendbare Schwefel ausfällende Stoffe sind u.a. Metallsalze wie Kupfersalze und Zinksalze. Als bevorzugte Salze sind Kupfergluconat, Zinkcitrat und Zinkgluconat zu nennen. Die Menge an Schwefel ausfällenden Stoffen beträgt 0,01 bis 2 Gew.%, bevorzugt 0,15 bis 1,5 Gew.%, besonders bevorzugt 0,25 bis 1,0 Gew.% des Films.
- Den erfindungsgemäßen Mundpflegefilmen können weiter die Speichelsekretion anregende Stoffe zugegeben werden. Verwendbare die Speichelsekretion anregende Stoffe sind offenbart im US-Patent Nr. 4,820,506, auf das hiermit vollinhaltlich Bezug genommen wird. Dazu gehören für Lebensmittel verwendbare Säuren wie Zitronensäure, Milchsäure, Apfelsäure, Bernsteinsäure, Ascorbinsäure, Adipinsäure, Fumarsäure und Weinsteinsäure. Bevorzugte Säuren sind die Zitronensäure, Apfelsäure und Ascorbinsäure. Die Menge der die Speichelsekretion anregenden Stoffe im Film beträgt zwischen 0,01 und 12 Gew.%, bevorzugt 1 bis 10 Gew.%, besonders bevorzugt 2,5 bis 6 Gew.%.
- Ein bevorzugter Weichmacher ist u.a. Triacetin in einer Menge von 0 bis 20 Gew.%, bevorzugt 0 bis 2 Gew.%. Weitere geeignete Weichmacher sind Monoacetin und Diacetin.
- Ein bevorzugtes Kühlmittel ist Monomenthylsuccinat in einer Menge von 0,001 bis 2,0 Gew.%, bevorzugt 0,2 bis 0,4 Gew.%. Ein Monomenthylsuccinat enthaltendes Kühlmittel ist zu beziehen von Mane, Inc. Weitere geeignete Kühlmittel sind WS3, WS23, Ultracool II u.ä.
- Bevorzugte Tenside sind Fettsäuremono- und -diglyceride und Polyoxyethylensorbitolester wie z.B. Atmos 300 und Polysorbat 80. Die Tenside können in Mengen von 0,5 bis 15 Gew.%, bevorzugt 1 bis 5 Gew.% des Films zugegeben werden. Weitere geeignete Tenside sind Pluronsäure, Natriumlaurylsulfat u.ä.
- Bevorzugte Stabilisatoren sind Xanthangummi, Johannisbrotkernmehl und Carrageenan in Mengen von 0 bis 10 Gew%, bevorzugt 0,1 bis 2 Gew.% des Films. Weitere geeignete Stabilisatoren sind Guargummi u.ä.
- Bevorzugte Emulgatoren sind Triethanolaminstearat, quarternäre Ammoniumverbindungen, Kariengummi, arabicum, Gelatine, Lecithin, Bentonit, Veegum u.ä. in Mengen von 0 bis 5 Gew.%, bevorzugt von 0,01 bis 0,7 Gew.% des Films.
- Bevorzugte Verdickungsmittel sind Methylcellulose, Carboxylmethylcellulose u.ä. in Mengen von 0 bis 20 Gew.%, bevorzugt von 0,01 bis 5 Gew.%.
- Ein bevorzugtes Bindemittel ist Stärke in einer Menge von 0 bis 10 Gew.%, bevorzugt 0,01 bis 2 Gew.% des Films.
- Geeignete Süßstoffe, die beigegeben werden können, sind die bekannten Süßstoffe, sowohl natürliche als auch künstliche. Als geeignete Süßstoffe sind z.B. zu nennen:
- A. Wasserlösliche Süßstoffe wie Monosaccharide, Disaccharide und Polysaccharide wie Xylose, Ribose, Glucose (Dextrose), Mannose, Galactose, Fructose (Levulose), Saccharose (Zucker), Maltose, Invertzucker (eine Mischung aus Fructose und von Saccharose abgeleiteter Glucose), teilhydrolysierte Stärke, die festen Bestandteile von Maissirup, Dihydrochalcone, Monellin, Stevioside und Glycyrrhizin;
- B. wasserlösliche künstliche Süßstoffe wie die löslichen Saccharinsalze, d.h. Natrium- oder Calciumsaccharinsalze, Cyclamatsalze, das Natrium-, Ammonium- oder Calciumsalz von 3,4-Dihydro-6-methyl-1,2,3-oxathiazin-4-on-2,2-dioxid, das Kaliumsalz von 3,4-Dihydro-6-methyl-1,2,3-oxathiazin-4-on-2,2-dioxid (Acesulfam-K), die freie Säureform von Saccharin u.ä.;
- C. auf Dipeptiden basierende Süßstoffe wie von L-Aspartinsäure abgeleitete Süßstoffe, z.B. L-Aspartyl-L-phenylalaninmethylester (Aspartam) und im US-Patent Nr. 3,492,131 beschriebene Stoffe, L-alpha-Aspartyl-N-(2,2,4,4-tetramethyl-3-thietanyl)-D-alaninamidhydrat, Methylester von L-Aspartyl-L-phenylglycerin und L-Aspartyl-L-2,5-dihydrophenyl-glycin, L-Aspartyl-2,5-dihydro-L-phenylalanin, L-Aspartyl-L-(1-cyclohexen)-alanin u.ä.;
- D. von natürlich vorkommenden wasserlöslichen Süßstoffen abgeleitete wasserlösliche Süßstoffe wie z.B. ein chloriertes Derivat von normalem Zucker (Saccharose), bekannt z.B. unter der Produktbeschreibung von Sucralose; und
- E. auf Proteinen basierende Süßstoffe wie Thaumatoccus danielli (Thaumatin I und II).
- Im Allgemeinen wird eine wirksame Menge an Süßstoff als Hilfsstoff eingesetzt, um die für eine bestimmte Zusammensetzung gewünschte Süße zu erhalten, wobei die Menge je nach dem gewählten Süßstoff variiert. Die Menge liegt normalerweise zwischen 0,01 und 10 Gew.% der Zusammensetzung, wenn ein leicht extrahierbarer Süßstoff verwendet wird. Die oben in Kategorie A genannten wasserlöslichen Süßstoffe werden gewöhnlich in Mengen von 0,01 bis 10 Gew.%, bevorzugt in Mengen von 2 bis 5 Gew.% eingesetzt. Manche Süßstoffe der Kategorie A (z.B. Glycyrrhizin) können auf Grund ihrer bekannten Süßkraft in den im Folgenden für die Kategorien B bis E genannten Mengen verwendet werden. Im Gegensatz dazu werden die in den Kategorien B bis E genannten Süßstoffe im Allgemeinen in Mengen von 0,01 bis 10 Gew.%, bevorzugt von 2 bis 8 Gew.%, besonders bevorzugt von 3 bis 6 Gew.% verwendet. Diese Mengen können unabhängig vom durch gegebenenfalls verwendete Geschmacksöle erzielten Geschmack zum Erhalt des gewünschten Süßegrades eingesetzt werden. Selbstverständlich müssen Filmen, die nicht für die orale Verabreichung vorgesehen sind, keine Süßstoffe beigefügt werden.
- Die verwendbaren Geschmacksstoffe umfassen die dem Fachmann bekannten Stoffe wie natürliche und künstliche Geschmacksstoffe. Diese Geschmacksstoffe können ausgewählt sein aus synthetischen Geschmacksölen und geschmacksvermittelnden Aromen und/oder Ölen, Extraktkonzentraten von Gewürzen und aus Pflanzen, Blättern, Blüten, Früchten usw. gewonnenen Extrakten sowie deren Kombinationen. Beispiele für Geschmacksöle sind das Öl der grünen Minze, Zimtöl, Pfefferminzöl, Nelkenöl, Lorbeeröl, Thymianöl, Zedernblattöl, Muskatöl, Salbeiöl und Bittermandelöl. Weiter geeignet sind künstliche, natürliche oder synthetische Fruchtaromen wie Vanille, Schokolade, Kaffee, Kakao und Zitrusöl wie Zitrone, Orange, Traube, Limone und Grapefruit, sowie Fruchtessenzen wie Apfel, Birne, Pfirsich, Erdbeer, Himbeer, Kirsche, Pflaume, Ananas, Aprikose usw. Diese Aromen können einzeln oder in Mischung verwendet werden. Verbreitet verwendete Geschmacksstoffe sind Minzen wie Pfefferminze, künstliche Vanille, Zimtderivate und verschiedene Fruchtaromen, die einzeln oder in Mischung verwendet werden. Geschmacksstoffe wie Aldehyde und Ester, so z.B. Cinnamylacetat, Cinnamaldehyd, Citral, Diethylacetal, Dihydrocarvylacetat, Eugenylformiat, p-Methylanisol usw. können ebenfalls eingesetzt werden. Im Allgemeinen können alle Geschmacksstoffe oder Nahrungsergänzungsmittel verwendet werden, die in "Chemicals Used in Food Processing", Veröffentlichung 1274 der National Academy of Sciences, S. 63–258 genannt sind. Weitere Beispiele für Aldehyde als Geschmacksstoffe sind unter anderem Acetaldehyd (Apfel), Benzaldehyd (Kirsche, Mandel), Zimtsäurealdehyd (Zimt), Citral, d.h. alpha-Citral (Zitrone, Limone), Neral, d.h. beta-Citral (Zitrone, Limone), Decanal (Orange, Zitrone), Ethylvanillin (Vanille, Sahne), Heliotropin, d.h. Piperonal (Vanille, Sahne), Vanillin (Vanille, Sahne), alpha-Amylcinnamaldehyd (würzige und fruchtige Geschmacksrichtungen), Butyraldehyd (Butter, Käse), Valeraldehyd (Butte, Käse), Citronellal (modifiziert, viele Richtungen), Decanal (Citrusfrüchte), Aldehyd C-8 (Citrusfrüchte), Aldehyd C-9 (Citrusfrüchte), Aldehyd C-12 (Citrusfrüchte), 2-Ethylbutyraldehyd (Beeren), Hexenal, d.h. Trans-2 (Beeren), Tolylaldehyd (Kirsche, Mandel), Veratraldehyd (Vanille), 2,6-Dimethyl-5-heptenal, d.h. Melonal (Melone), 2,6-Dimethyloctanal (grünes Obst) und 2-Dodecenal (Citrus, Mandarine), Kirsch, Traube, deren Mischungen u.ä.
- Die verwendete Menge an Geschmacksstoffen ist normalerweise Sache des Geschmacks und ist abhängig von Faktoren wie der gewünschten Art des Geschmacks und der gewünschten Geschmacksintensität. So kann die Menge zum Erhalt des gewünschten Ergebnisses im Endprodukt variiert werden. Der Fachmann ist in der Lage, ohne unnötiges Experimentieren die Mengenveränderungen durchzuführen. Im Allgemeinen sind Mengen von 0,1 bis 30 Gew.% einsetzbar, bevorzugt 2 bis 25 Gew.%, besonders bevorzugt 8 bis 10 Gew.%.
- Die erfindungsgemäßen Zusammensetzungen können auch Farbstoffe oder Färbemittel enthalten. Die Farbstoffe werden in einer wirksamen Menge verwendet, die ausreicht, die gewünschte Farbe zu erhalten. Die im Rahmen der Erfindung verwendbaren Farbstoffe umfassen Pigmente wie Titandioxid, das in Mengen von bis zu 5 Gew.%, bevorzugt weniger als 1 Gew.%, zugegeben werden kann. Färbemittel können auch natürliche Nahrungsmittelfarben und für Nahrungsmittel, Arzneimittel und Kosmetika geeignete Färbemittel sein. Diese Färbemittel sind bekannt als FD&C (Food, Drug & Cosmetics) Färbemittel und Lacke. Die für das vorstehend angeführte Verwendungsspektrum geeigneten Stoffe sind bevorzugt wasserlöslich und umfassen u.a. FD&C Blau Nr. 2, das das Dinatriumsalz der 5,5-Indigotindisulfonsäure darstellt. In ähnlicher Weise stellt das unter der Bezeichnung Grün Nr. 3 bekannte Färbemittel einen Triphenylmethanfarbstoff dar und ist das Mononatriumsalz von 4-[4-N-Ethyl-p-sulfobenzylamino)diphenyl-methylen]-[1-N-ethyl-N-p-sulfoniumbenzyl)-2,5-cyclohexadienimin]. Eine vollständige Auflistung aller FD&C- und D&C-Färbemittel und ihrer entsprechenden chemischen Strukturen ist zu finden in der Encyclopedia of Chemical Technology von Kirk-Othmer, Band 5, S. 857–884, auf welche Textstelle hiermit Bezug genommen wird.
- Antimikrobielle Wirkung der Mundpflegefilme
- Die bevorzugte Ausbildungsform der erfindungsgemäßen Mundpflegefilm-Zusammensetzung enthält die etherischen Öle, die in der Listerine® Mundwäsche zur Erzielung einer antimikrobiellen Wirkung eingesetzt werden. Die Filme werden in einer der Verwendung in der Mundhöhle entsprechenden Form und Größe hergestellt. Der Film haftet an einer Oberfläche im Mund, normalerweise am Dach der Mundhöhle oder an der Zunge, und löst sich schnell auf. Die Menge an etherischen Ölen in einem einzelnen Film von zur Verwendung in der Mundhöhle bevorzugter Größe ist wesentlich geringer als in der empfohlenen Menge, 20 ml, der Listerine® Mundwäsche.
- In einer bevorzugten erfindungsgemäßen Formulierung beträgt die Menge an Thymol und Eucalyptol im Film etwa 70 Mal weniger als in der Mundwäsche. Die Menge an Methylsalicylat ist im Film etwa 46 Mal geringer als in der Mundwäsche. Die Menge an Menthol ist im Film etwa 2,8 Mal geringer als in der Mundwäsche. Diese Zahlen basieren auf dem Vergleich einer 20 ml-Dosis flüssiger Mundwäsche mit einem Film von 0,0358 g.
- Die Erfinder haben unerwarteter Weise gefunden, dass der Film bei solch geringen Mengen an Ölen eine lang anhaltende antimikrobielle Wirkung bietet. Die Erfinder sind der Meinung, dass die Wirkung der etherischen Öle durch die Bildung einer Pullulanschicht in der Mundhöhle, die die etherischen Öle festhält, gefördert wird. Dies war nicht zu erwarten, da Pullulan wasserlöslich ist und der Film sich sehr schnell auflöst.
- Die verlängerte antimikrobielle Wirkung wird durch die folgenden Versuche nachgewiesen.
- Das Ziel dieser Versuche bestand darin, die antibakterielle Wirkung einer Anwendung eines den Atem verbessernden Films auf Mundgeruch verursachende Mikroorganismen auf der Zunge 30, 60 oder 90 Minuten nach der Verwendung zu ermitteln. Im 30-Minuten-Versuch wurde auch die Wirksamkeit der Verwendung von zwei Filmen untersucht. Die wiederholbar gewinnbaren Basisdaten der Mundgeruch verursachenden Mikroben der Testperson wurden durch Auszählen eines auf ein selektives Agarmedium gegebenen Zungenabstrichs gewonnen. Das Testprodukt wurde angewendet, und die Testpersonen ließen ein oder zwei Atem erfrischende Filme auf der Zunge zergehen. Die Testpersonen verblieben in den Räumlichkeiten der Versuchsdurchführung, und 30, 60 oder 90 Minuten nach Anwendung des Testprodukts auf der Zunge wurde ein zweiter Abstrich gemacht. 48 Stunden später, nachdem das Testprodukt abgewaschen war (wash-out-time), wurde von denselben Testpersonen ein Kontrollabstrich zur Ermittlung der Daten ohne Behandlung vorgenommen.
- Die Testpersonen, die einen Film konsumiert hatten, zeigten nach 30 Minuten im Vergleich zur Kontrollgruppe eine Verringerung der Mean-Log-Zählwerte der Mundgeruch verursachenden Mikroben. Die Daten besaßen statistische Borderline-Signifikanz (p = 0,052). Die Differenz zwischen der Gruppe, die einen Film konsumiert hatte, und der Kontrollgruppe ohne Behandlung ergab bei der Auszählung der Kolonien der Mundgeruch verursachenden Mikroben eine Verringerung von 42,7 %.
- Eine statistisch signifikante Verringerung der Mundgeruch verursachenden Mikroben wurde auch bei der Gruppe, die zwei Filme konsumiert hatte, festgestellt. Die Aus zählung der Kolonien der Mundgeruch verursachenden Mikroben ergab eine Verringerung von 79,6 % (p < 0,001).
- Auch 60 Minuten nach Verwendung des Films wurde bei der Gruppe, die einen Atem erfrischenden Film konsumiert hatte, eine statistisch signifikante Verringerung der Mundgeruch verursachenden Mikroben festgestellt. Die Auszählung der Kolonien der Mundgeruch verursachenden Mikroben ergab eine Verringerung von 69,8 % (p = 0,002).
- Ebenso wurde 90 Minuten nach Verwendung eines einzigen Atem erfrischenden Films eine signifikante Verringerung des Mundgeruchs festgestellt. Die Auszählung der Kolonien der Mundgeruch verursachenden Mikroben ergab eine Verringerung von 69,1 % (p = 0,006).
- Die in diesem Versuch gewonnenen Daten lassen folgende Schlüsse zu: (1) Auf Pullulanpolymer basierende Atem erfrischende Filme, die etherische Öle enthalten, sind wirksame antibakterielle Zusammensetzungen gegen Mundgeruch verursachende Bakterien und (2) in vivo wurde eine wesentliche Verringerung der Bakterien 30, 60 und 90 Minuten nach Verwendung erreicht.
- Versuchsverfahren
- Die antimikrobiellen Versuche wurden wie folgt durchgeführt. Die Testpersonen wurden angewiesen, ab Mitternacht vor dem Versuch und bis Beendigung des Versuchs am jeweiligen Tag jegliche Mundhygiene (z.B. Zähneputzen, Mundspülen) zu unterlassen und kein Essen, kein Trinken und keine Süßigkeiten zu sich zu nehmen. Ab dem Morgen vor den Geruchsbewertungen unterließen die Testpersonen das Rauchen. Testgläser mit 10 ml sterilem 0,01 %-igem Pepton Sterile Abstrichwerkzeuge OOPS III Agar (B.-F. Turng, G. E. Minah und W. A. DF0792-17-3) 44 g Destilliertes Wasser 1 l Bleiacetata (Sigma L3396) 0,2 g Hemin-Lösungb (Sigma H-1652) 2 ml Glutathionc (Sigma G4251) 1,2 g - 44 g Columbia Blut Agar-Basis wurden in 1 l destilliertem Wasser gelöst und zur völligen Lösung aufgekocht. Das Medium wurde 15 Min. bei 121–124°C sterilisiert.
- a 0,2 g Bleiacetat wurden in 1 ml destilliertem H2O gelöst und durch Filtration sterilisiert. Sie wurden nach Autoklavieren des Basismediums zugegeben.
- b 50 mg Hemin wurden in 1 ml 1 N NaOH gelöst, mit destilliertem H2O auf 100 ml aufgefüllt und durch Filtration sterilisiert. Nach Autoklavieren des Basismediums wurden 2 ml pro Liter OOPS III zugegeben.
- c 1,2 g Glutathion wurden in 10 ml destilliertem H2O gelöst und durch Filtration sterilisiert. Sie wurden nach Autoklavieren des Basismediums zugegeben.
- 2. Verfahren
-
- a. Alle Medien wurden über Nacht in einer anaeroben Kammer vorreduziert. Die Platten wurden zum Schutz vor übermäßigem Austrocknen lose in Plastikbeutel gegeben.
- b. Die Testpersonen verzichteten ab Mitternacht vor Beginn des Assays und bis Abschluss des Assays auf Mundhygiene, Essen und Trinken. Zwölf Versuchspersonen wurden für die 60- und 90-Minuten-Versuche eingeteilt, und 18 Versuchspersonen wurden für den 30-Minuten-Versuch eingeteilt.
- c. Die Testpersonen nahmen jeweils von der rechten Zungenhälfte einen Abstrich, wobei sie das Abstrichwerkzeug in der Mitte der Zunge ansetzten und nach vorn zur Spitze hin strichen. Der Abstrich wurde in ein Glas mit Pepton gegeben.
- d. Die Testpersonen wurden entweder mit einem Film oder mit zwei Filmen behandelt. Die Testpersonen platzierten den Atem erfrischenden Film auf der linken Seite der Zunge, so dass er die Zunge von der Mitte bis zur Spitze bedeckte, und ließen den Film zergehen, wobei sie dreißig Sekunden lang den Mund leicht geöffnet hielten, um ein Anhaften des Films am Gaumen zu vermeiden.
- e. Nach 30 bzw. 60 Minuten nahmen die Testpersonen von der linken Zungenhälfte einen Abstrich, wobei sie das Abstrichwerkzeug in der Mitte der Zunge ansetzten und nach vorn zur Spitze hin strichen. Der Abstrich wurde in ein Glas mit Pepton gegeben.
- f. Die Peptongläser wurden 10 Sek. stark verquirlt, und danach wurden Serienverdünnungen hergestellt. Die 10–4-Verdünnung wurde je zweifach auf OOPS III-Agar-Platten gegeben, wobei ein Spiral Biotech Autoplate 4000 (Bethesda, MD, USA) verwendet wurde. Alle Platten wurden mit den Initialen der Testperson, dem Datum des Assays, dem Zeitpunkt der Probennahme und der Replikationszahl gekennzeichnet.
- g. Die Platten wurden in einer anaeroben Kammer bei 35–37°C 7 Tage inkubiert, um eine volle Entwicklung der Kulturen ohne übermäßiges Wachstum zu erreichen.
- h. Nach einer wash-out-Zeit von 48 Stunden wurden die Testpersonen einem Kontrollversuch ohne Behandlung unterzogen. Es wurde kein Film angewendet, und die Schritte (c) bis (g) wurden wie oben beschrieben durchgeführt.
- i. Nach einer wash-out-Zeit von 48 Stunden wurde an den Testpersonen des 60-Minuten-Versuchs nochmals ein Film angewendet. Die Schritte (a) bis (h) wurden durchgeführt, mit dem Unterschied, dass in Schritt (e) die Testpersonen den Abstrich nach 90 Minuten nahmen.
- j. Die dunkel pigmentierten Kolonien (H2S produzierende Organismen) wurden in Ganzplattenzählung von Hand bei geeigneter Vergrößerung oder in Segmentzählung unter Verwendung einer Spiral Biotech-Zählschablone gezählt. Der entsprechende Code wurde zur Interpretation der Zählungen auf dem Datenblatt eingetragen. Die gezählten koloniebildenden Einheiten (colony-forming units, CFU) wurden durch Division durch die entsprechende in Tabelle A aufgeführte exponentielle Volumenkonstante und Multiplikation mit 1000 in CFU/ml umgerechnet. Dieser Wert wurde anschließend mit dem Verdünnungsfaktor der Platte (104) multipliziert.
- Für die in vivo-Keimabtötungsversuche wurde der Film gemäß Beispiel 19, wie in Tabelle 2 beschrieben, verwendet. Die im Versuch verwendeten Filme waren etwa 22 mm × 32 mm groß, etwa 0,0013 bis 0,0015 Inches dick und hatten ein Gewicht zwischen etwa 35 mg und etwa 37 mg.
- Die erhöhte Wirksamkeit des etherische Öle enthaltenden Pullulanfilms geht auch aus den
1 und2 hervor.1 stellt eine Fotografie einer Agarplatte mit Streptococcus mutans, ATCC Nr. 25175, dar, der ein Stück eines erfindungsgemäßen etherische Öle enthaltenden Pullulanfilms zugegeben wurde. Das Stück Film gab etwa 0,391 mg etherische Öle ab, wobei der Film gemäß dem unten angeführten Beispiel 15 verwendet wurde. -
2 stellt eine Fotografie einer Agar-Platte mit Streptococcus mutans, ATCC Nr. 25175, dar, der Tropfen etherischer Öle zugefügt wurden. Die Tropfen hatten ein Volumen von 148 μl und enthielten 0,391 mg etherische Öle. Der Anteil der etherischen Öl im Tropfen betrug 2,200 % Menthol, 0,186 % Eucalyptol, 0,186 % Methylsalicylat und 0,1300 % Thymol in hydroalkoholischer Lösung. - Die Inhibitionszone um den Film ist gemäß
1 viel größer als die Abmessungen des Films selbst. Dies ist auf das Vorhandensein von Pullulan zurückzuführen, da die Öle im Pullulanfilm durch das Pullulan ausgebreitet und nach außen diffundiert wurden und nach wiederholtem Ausspülen nicht ausgewaschen wurden. Im Gegensatz dazu diffundierten die etherischen Öle gemäß2 nicht aus dem Tropfen heraus, verblieben kreisförmig und ließen sich durch ein- bis zweimaliges Ausspülen leicht auswaschen. Dies zeigt, dass die antimikrobielle Wirkung der etherischen Öle durch das Vorhandensein von Pullulan gesteigert wird. - Verfahren zur Herstellung der etherische Öle enthaltenden Filme Verfahren zur Herstellung der erfindungsgemäßen Filme ermöglichen die Verkapselung der Öle in der filmbildenden Matrix und die Aufrechterhaltung der unbeschädigten Filme auch bei einem Ölgehalt im Film von 10 Gew.% oder darüber.
- Gemäß bestimmten Verfahren zur Herstellung der erfindungsgemäßen Filme werden die filmbildenden Bestandteile getrennt von den wasserlöslichen Bestandteilen vermischt und mit Wasser hydriert, und die wasserlöslichen Bestandteile werden getrennt von den organischen Bestandteilen und Tensiden in wässriger Lösung gemischt. Bei diesen Verfahren wird die endgültige Formulierung bevorzugt durch Mischen der filmbildenden Phase mit der wässrigen Phase und anschliessendes Beimischen der organischen, auch Tenside wie Polysorbat 80 und Atmos 300 umfassenden Phase erhalten. Die Masse wird bis zur Emulsion gemischt. Gemäß anderen Ausführungsformen werden die wässrige und die filmbildende Phase durch Lösen der wasserlöslichen Bestandteile im Wasser und anschließende Zugabe der Gummen zur Hydrierung vereinigt. Die organische Phase wird danach dieser einzigen wässrigen Phase zugegeben.
- Die erhaltene Formulierung wird auf einem geeigneten Substrat ausgebreitet und unter Bildung eines Films getrocknet. Bevorzugt wird der Film an der Luft oder mit Hilfe von Warmluft getrocknet und in gewünschter Größe geschnitten, verpackt und gelagert. Der Film kann 0,1 % bis 10 Gew.%, bevorzugt 3 % bis 8 Gew.%, besonders bevorzugt 4 bis 7 Gew.% Feuchtigkeit enthalten.
- Die filmbildende Phase kann Pullulan und Stabilisatoren wie Xanthangummi, Johannisbrotkernmehl und Carrageenan enthalten. Diese Bestandteile werden vermischt und dann etwa 30 bis etwa 48 Stunden zur Bildung eines Gels in Wasser hydriert. Zur Förderung der Hydrierung wird das Wasser bevorzugt auf eine Temperatur von etwa 25 bis etwa 45°C erwärmt. Die Wassermenge beträgt 40 bis 80 % des Gels.
- Das erhaltene hydrierte Gel wird dann im Verlauf von etwa 1 h bis etwa 48 h auf eine Temperatur von etwa 20 bis etw 30°C gekühlt. Bevorzugt ist das Wasser entionisiert.
- Die wässrige Phase kann Bestandteile wie einen oder mehrere Farbstoff(e), Kupfergluconat und Süßstoffe enthalten. Das Wasser ist bevorzugt entionisiert, und die verwendete Wassermenge liegt zwischen 5 und 80 Gew.% der fertigen Gelmischung.
- Wenn sowohl Natriumsaccharin als auch Kupfergluconat Bestandteile der Formulierung sind, so werden sie bevorzugt separat gelöst, um ein Ausfällen zu vermeiden.
- Gemäß einer bevorzugten Ausführungsform der Herstellung der erfindungsgemässen etherische Öle enthaltenden Filme können die filmbildenden Bestandteile hydriert und alle Bestandteile ohne Erwärmen vereinigt werden. Das bevorzugte Herstellungsverfahren der Filme umfasst Lösen der wasserlöslichen Bestandteile in Wasser unter Erhalt einer wässrigen Mischung; Mischen der filmbildenden Bestandteile in Pulverform unter Erhalt einer Pulvermischung; Zugabe der Pulvermischung zur wässrigen Mischung unter Erhalt eines hydrierten Polymergels; Mischen des hydrierten Polymers bei Raumtemperatur im Verlauf von etwa 30 Minuten bis etwa 48 Stunden; Mischen von Kühlmittel, Thymol und Menthol im Geschmacksöl unter Erhalt einer Ölmischung; Zugabe von Methylsalicylat, Eucalyptol und Tensiden zur Ölmischung; Zugabe der Ölmischung zum hydrierten Polymergel und Mischen bis zum Erhalt einer einheitlichen Mischung; Entlüften des Films, bis die Luftblasen entfernt sind, Ausbreiten der einheitlichen Mischung auf einem geeigneten Substrat und Trocknen der ausgebreiteten Mischung unter Bildung eines Films.
- Gemäß dem bevorzugten Verfahren zur Herstellung des etherische Öle enthaltenden Films werden die filmbildenden Bestandteile ohne Erwärmen des Wassers hydriert. Das Erwärmen der Bestandteile erhöht die Energiekosten des Herstellungsprozesses. Außerdem führt Erwärmen zum unerwünschten Schwund flüchtiger Bestandteile durch Verdampfen, was sich wegen des Verlusts bei den etherischen Ölen auch auf die keimabtötende Wirkung der Zusammensetzung auswirkt. Weiter wird durch das Mischen der Öle in zwei Schritten der Geschmacksverlust minimiert.
- Ohne sich auf eine bestimmte Theorie festlegen zu wollen wird angenommen, dass die filmbildenden Bestandteile auf Grund eines ionischen Effekts, als das Donnan-Gleichgewicht bekannt, ohne Erwärmen hydriert und gemischt werden können. Die Hydrierung der filmbildenden Stoffe in Gegenwart von Elektrolyten in Lösung senkt wirksam die Viskosität des gebildeten gelförmigen Polymers, wodurch die Effizienz des Hydrierungsvorgangs erhöht wird. Die wasserlöslichen Bestandteile der Formulierung, die vor Zugabe der filmbildenden Bestandteile in der Hydrierlösung gelöst werden, liefern die Elektrolyte. High-Shear-Mischen beschleunigt ebenfalls die Hydrierung, wobei Klumpen des Pulvers zerkleinert werden und so eine größere Oberfläche für den Kontakt mit dem Wasser geschaffen wird. Weiter liefern lokale Wärmeeffekte in den Scherbereichen Energie für die Hydrierung ohne wesentliche Erhöhung der Temperatur der Masse.
- Bevorzugt wird vermieden, Kupfergluconat und Saccharin gleichzeitig der wässrigen Lösung zuzugeben, da sich dann ein Niederschlag bildet. Deshalb werden bevorzugt andere Süßstoffe als Saccharin mi Kupfergluconat kombiniert.
- Beschreibung von Filmzusammensetzungen zur Verabreichung pharmazeutischer Wirkstoffe
- Eine zweite Ausführungsform der Erfindung bezieht sich auf einen schnell löslichen Film, der wenigstens einen physiologisch akzeptablen pharmazeutischen Wirkstoff enthält. Der Begriff "physiologisch akzeptabel" bezieht sich im Rahmen des vorliegenden Dokuments auf Verbindungen, die bei Verabreichung an einen Patienten in annehmbarer Weise vertragen werden, ohne übermäßige unerwünschte Nebenwirkungen zu zeigen. Der Begriff umfasst auch essbare Verbindungen.
- Der Begriff "pharmazeutische Wirkstoffe" bezieht sich im Rahmen des vorliegenden Dokuments auf von Lebensmitteln verschiedene Wirkstoffe, die in und/oder am Körper des Individuums, dem sie verabreicht wurden, strukturelle und/oder funktionelle Veränderungen bewirken. Diese Wirkstoffe sind nicht in besonderer Weise begrenzt; jedoch sollten sie physiologisch akzeptabel und mit dem Film kompatibel sein. Geeignete pharmazeutische Wirkstoffe sind unter anderem die folgenden:
- A. Antimikrobielle Wirkstoffe wie Triclosan, Cetylpyridiumchlorid, Domiphenbromid, quarternäre Ammoniumsalze, Zinkverbindungen, Sanguinarin, Fluoride, Alexidin, Octonidine, EDTA u.ä.,
- B. nichtsteroidale entzündungshemmende Arzneimittel wie Aspirin, Acetaminophen, Ibuprofen, Ketoprofen, Diflunisal, Fenoprofen-Calcium, Naproxen, Tolmetin-Natrium, Indomethacin u.ä.,
- C. Antitussiva wie Benzonatat, Caramiphenedisylat, Menthol, Dextromethorphan-Hydrobromid, Chlophedianol-Hydrochlorid u.ä.,
- D. decongestive Wirkstoffe wie Pseudoephedrin-Hydrochlorid, Phenylepherin, Phenylpropanolamin, Pseudoephedrinsulfat u.ä.,
- E. Antihistaminika wie Brompheniraminmaleat, Chlorpheniraminmaleat, Carbinoxaminmaleat, Clemastinfumarat, Dexchlorpheniraminmaleat, Diphenhydramin-Hydrochlorid, Diphenylpyralin-Hydrochlorid, Azatadinmeleat, Diphenhydramincitrat, Doxylaminsuccinat, Promethazin-Hydrochlorid, Pyrilaminmaleat, Tripelennamincitrat, Triprolidin-Hydrochlorid, Acrivastin, Loratadin, Brompheniramin, Dexbrompheniramin u.ä.,
- F. Expectorantien wie Guaifenesin, Ipecac, Kaliumiodid, Terpinhydrat u.ä.,
- G. Wirkstoffe gegen Diarrhoe wie Loperamid u.ä.,
- H. H2-Antagonisten wie Famotidin, Ranitidin u.ä.,
- I. Protonpumpen-Inhibitoren wie Omeprazol, Lansoprazol u.ä.,
- J. allgemeine nicht selektive ZNS-Depressiva wie aliphatische Alkohole, Barbiturate u.ä.
- K. allgemeine nicht selektive ZNS-Stimulantien wie Coffein, Nicotin, Strychnin, Picrotoxin, Pentylentetrazol u.ä.,
- L. Arzneimittel, die die Funktion des ZNS selektiv modifizieren, wie Phenyhydantoin, Phenobarbital, Primidon, Carbamazepin, Ethosuximin, Methsuximid, Phensuximid, Trimethadion, Diazepam, Benzodiazepine, Phenacemid, Pheneturide, Acetazolamid, Sulthiam, Bromid u.ä.,
- M. Anti-Parkinson-Mittel wie Levodopa, Amantadin u.ä.,
- N. Narkotika und Analgetika wie Morphin, Heroin, Hydromorphon, Metopon, Oxymorphon, Levorphanol, Codein, Hydrocodon, Xycodon, Nalorphin, Naloxon, Naltrexon u.ä.,
- O. Analgetika und Antipyretika wie Salycilate, Phenylbutazon, Indomethacin, Phenacetin u.ä.,
- P. Psychopharmaka wie Chlorpromazin, Methotrimeprazin, Haloperidol, Clozapine, Reserpin, Imipramin, Tranylcypromin, Phenelzin, Lithium u.ä.
- Die Mengen an in den erfindungsgemäßen schnell löslichen Filmen verwendbarem Arzneimittel ist von der zum Verabreichen einer wirksamen Menge an Arzneimittel erforderlichen Dosis abhängig. Beispiele für Dosen bestimmter Medikamente, die pro Streifen schnell löslichen oralen Films verabreicht werden können, sind in Tabelle 1 angeführt.
- Die zur Herstellung der pharmazeutische Wirkstoffe enthaltenden Filme verwendeten Bestandteile ähneln den für die Mundpflegefilme verwendeten. Konkret können die oben beschriebenen Plastifikatoren, Kühlmittel, Tenside, Stabilisatoren, Emulgatoren, Verdickungsmittel, Bindemittel, Filmbildner, Süßstoffe, Geschmacks- und Farbstoffe auch für alle erfindungsgemäßen Filme verwendet werden.
- Die zur Verabreichung eines pharmazeutischen Wirkstoffs dienenden Filme können auch ein Triglycerid enthalten. Beispiele für Triglyceride sind Pflanzenöle wie Maisöl, Sonnenblumenöl, Erdnussöl, Olivenöl, Zimtöl, Sojaöl und deren Mischungen. Ein bevorzugtes Triglycerid ist Olivenöl. Das Triglycerid wird dem Film in Mengen von 0,1 Gew.% bis 12 Gew.%, bevorzugt von 0,5 Gew.% bis 9 Gew.% des Films zugegeben.
- Die pharmazeutische Wirkstoffe enthaltenden Filme können auch einen Konservierungsstoff enthalten. Dieser wird in Mengen von 0,001 Gew.% bis 5 Gew.%, bevorzugt von 0,01 Gew.% bis 1 Gew.% des Films zugegeben. Bevorzugte Konservierungsmittel sind Natriumbenzoat und Kaliumsorbat.
- Die pharmazeutische Wirkstoffe enthaltenden Filme können auch eine Polyethylenoxidverbindung enthalten. Das Molekulargewicht der Polyethylenoxidverbindung liegt zwischen etwa 50 000 und 6 000 000. Eine bevorzugte Polyethylenoxidverbindung ist N-10, zu beziehen bei der Union Carbide Corporation. Die Polyethylenoxidverbindung wird in Mengen von 0,1 Gew.% bis 5 Gew.%, bevorzugt von 0,2 Gew.% bis 4,0 Gew.% des Films zugegeben.
- Die pharmazeutische Wirkstoffe enthaltenden Filme können auch Propylenglycol enthalten. Dieses wird in einer Menge von 1 Gew.% bis 20 Gew.%, bevorzugt von 5 Gew.% bis 15 Gew.% des Films zugegeben.
- Der im Film verwendete Wirkstoff kann beschichtet sein zur Maskierung seines Geschmacks oder um zu verhindern, dass er auf der Zunge oder auf anderen Oberflächen der Mundhöhle ein taubes Gefühl hervorruft. Dem Fachmann sind die verwendbaren Beschichtungen bekannt. Zu ihnen gehören Polymere wie z.B. Eudragit® E, Celluloseprodukte (Cellulosics) wie Ethylcellulose u.ä.
- Eine weitere Möglichkeit zur Maskierung des Geschmacks des Wirkstoffs ist die Verwendung eines Ionenaustauschharzes wie Amberlite RP-69, zu beziehen bei Röhm und Haas, und Dow XYS-40010.00, zu beziehen bei der Dow Chemical Co.
- Beispiele
- Die Erfindung wird im Einzelnen veranschaulicht durch die folgenden Beispiele, wobei die Erfindung aber nicht auf die Beispiele beschränkt ist.
- Herstellungsverfahren I
- Zur Herstellung der Filme der Beispiele 1–13 wurde das folgende Verfahren angewendet.
- A. Die filmbildenden Bestandteile (z.B. Xanthangummi, Johannisbrotkernmehl, Carrageenan und Pullulan) außer Polysorbat 80 und Atmos 300 werden vermischt, in heißem gereinigten Wasser unter Erhalt eines Gels hydriert und über Nacht im Kühlschrank bei einer Temperatur von etwa 4°C unter Erhalt der Zubereitung A gelagert.
- B. Der/die Farbstoff(e), Kupfergluconat und Süßstoff werden in gereinigtes Wasser gegeben und darin aufgelöst unter Erhalt der Zubereitung B.
- C. Die Zubereitung B wird zur Zubereitung A gegeben und gut vermischt unter Erhalt der Zubereitung C.
- D. Der Geschmacksstoff und die Öle (z.B. Kühlmittel, Thymol, Methylsalicylat, Eucalyptol und Menthol) werden vermischt unter Erhalt der Zubereitung D.
- E. Polysorbat 80 und Atmos 300 werden zur Zubereitung D gegeben und gut vermischt unter Erhalt der Zubereitung E.
- F. Zubereitung E wird zur Zubereitung C gegeben und gut vermischt unter Erhalt der Zubereitung F.
- Die Zubereitung F wird bei Raumtemperatur in eine Form gegossen und ausgebreitet unter Erhalt eines Films der gewünschten Stärke. Der Film wird mit Warmluft getrocknet und in der gewünschten Größe geschnitten, verpackt und gelagert.
- Herstellungsverfahren II
- Zur Herstellung der Filme der Beispiele 14–18 wurde das folgende Verfahren angewendet.
- A. Kupfergluconat, Acesulfam K, Aspartam, Glycerin, Sorbit und Farbstoff werden in gereinigten Wasser gelöst unter Erhalt einer wässrigen Mischung;
- B. Pullulan, Xanthangummi, Johannisbrotkernmehl und Carrageenan werden in Pulverform miteinander vermischt unter Erhalt einer Pulvermischung;
- C. die Pulvermischung von Schritt B wird zur wässrigen Mischung von Schritt A gegeben unter Erhalt eines hydrierten gelförmigen Polymers;
- D. das hydrierte Polymer von Schritt C wird langsam (bei etwa 50 bis 100 U/min) bei Raumtemperatur über Nacht gerührt;
- E. Kühlmittel, Thymol und Menthol werden vermischt und im Geschmacksöl gelöst;
- F. Methylsalicylat, Eucalyptol, Polysorbat 80 und Atmos 300 werden der Ölmischung von Schritt E zugegeben;
- G. die Ölmischung von Schritt F wird dem hydrierten Polymergel von Schritt D zugegeben und bis zum Erhalt einer einheitlichen Masse vermischt;
- H. das einheitliche Gemisch von Schritt G wird auf einem geeineten Träger ausgebreitet und
- I. das ausgebreitete Gemisch wird unter Erhalt eines Films getrocknet.
- Beispiel 1
- Gemäß Beispiel 1 wurde ein erfindungsgemäßer Film mit blaugrüner Färbung, Minzegeruch und einem erfrischenden Minzegeschmack erhalten.
- Beispiele 2–4
- Die Filme der Beispiele 2–4 enthalten Sorbit oder Glycerin oder beides. In diesen Beispielen wurden Produkte erhalten, die leicht zerbrachen oder zu feucht waren und/oder an sich selbst anhafteten. Jedoch wurden auch hier Filme erhalten, die sich in der Mundhöhle schnell auflösten und einen erfrischenden Minzegeschmack aufwiesen.
- Beispiele 5–6
- Die Filme der Beispiele 5 und 6 enthielten weder Glycerin noch Sorbit. Die erhaltenen Filme klebten während Herstellung und Verpackung nicht aneinander und waren über einen langen Zeitraum feuchtigkeitsbeständiger.
- Beispiele 7–9
- Die Filme der Beispiele 7–9 wurden hergestellt, um die Auswirkung von Avicel® auf die keimabtötende Wirkung festzustellen. Die Beispiele 7–9 ergaben zwar bezüglich Herstellung und Handhabbarkeit bessere Filme, jedoch wiesen sie im Vergleich zu Filmen ohne Avicel®, wie gemäß Beispiel 8, eine verringerte antimikrobielle Wirkung auf.
- Beispiele 10–15
- In den Beispielen 10–15 wurden die Mengen von Aspartam und Menthol variiert zur Veränderung von Süßegrad und Kühlegrad des Films.
- Beispiel 16
- In Beispiel 16 wurde das Sorbit durch Maltit ersetzt, das weniger feuchtevermittelnde Eigenschaften aufweist. Der erhaltene Film war während Herstellung und langfristiger Lagerung weniger klebrig.
- Beispiel 17
- In Beispiel 17 wurde das Pullulan bei der Herstellung der erfindungsgemäßen Filme durch einen anderen filmbildenden Stoff, nämlich Polyvinylpyrrolidon, ersetzt.
- Beispiel 18
- In Beispiel 18 wurde das Pullulan bei der Herstellung der erfindungsgemäßen Filme teilweise durch einen anderen filmbildenden Stoff, nämlich Konjacmehl, ersetzt.
- Beispiel 19
- In Beispiel 19 wurde ein Film erhalten, der einen die Speichelsekretion anregenden Stoff, nämlich Zitronensäure, enthält.
- Beispiel 20
- Beispiel 20 betrifft die Filmzusammensetzung, die in den oben beschriebenen Versuchen zur antimikrobiellen Wirkung verwendet wurde.
- Die Formulierungen der Beispiele 1–20 sind in Tabelle 2 zusammengefasst. Die Mengen in diesen Beispielen sind als tatsächliches Gewicht in Gramm oder in Gewicht-% angegeben. Mit diesen Formulierungen werden die Lösungen bzw. Gels erhalten, die ausgebreitet und unter Erhalt eines Films getrocknet werden. Die tatsächliche Menge der einzelnen Bestandteile im fertigen, getrockneten Film hängt davon ab, wieviel der relativen Feuchtigkeit beim Trocknen entfernt wird.
- Die folgenden Beispiele betreffen erfindungsgemäße Filme gemäß der zweiten Ausführungsform der Erfindung, worin der sich schnell auflösende Film einen pharmazeutischen Wirkstoff enthält. Die in Tabelle 3 aufgeführten Beispiele 21A bis 21E stellen Formulierungen für sich schnell auflösende, ein Medikament enthaltende orale Filme dar. Die Mengenangaben in Tabelle 3 sind in Milligramm.

- In Tabelle 4 sind weitere erfindungsgemäße Filme aufgeführt. Die Mengenangaben in Tabelle 4 stellen Gewichts-% vor dem Trocknen dar.
- Beispiel 22A diente zur Herstellung von Filmen, die a) 7,5 mg Dextromethorphan-Hydrobromid, b) 2,5 mg Tripolidin, c) 4,0 mg Chlorpheniraminmaleat und d) 12,5 mg Diphenylhydramin-Hydrochlorid enthalten.
- Beispiel 22B diente zur Herstellung eines Films, der 10 mg Dextromethorphan-Hydrobromid enthält.
- Beispiel 22C diente zur Herstellung eines Films, der 10 mg Dextromethorphan-Hydrobromid enthält.
- Beispiel 22D diente zur Herstellung eines Films, der a) 10 mg Phenylepherin-Hydrochlorid, b) 10 mg Phenylepherin-Hydrochlorid und 4 mg Chlorpheniraminmaleat und c) 10 mg Dextromethorphan-Hydrobromid enthält.
- Beispiel 22E diente zur Herstellung eines Films, der 7,5 mg Dextromethorphan-Hydrobromid enthält.
- Beispiel 22F diente zur Herstellung eines Films, der 20 mg beschichtetes Dextromethorphan-Hydrobromid zur Bereitstellung einer Dosis von 7,5 mg enthält.
- Beispiel 22G diente zur Herstellung eines Films, der a) 7,5 mg Dextromethorphan-Hydrobromid, b) 10 mg Phenylepherin-Hydrochlorid und c) 10 mg Phenylepherin-Hydrochlorid und 4 mg Chlorpheniraminmaleat enthält.
- Beispiel 22H diente zur Herstellung eines Films, der 15 mg Dextromethorphan-Hydrobromid enthält.
- Beispiel 22I diente zur Herstellung eines Films, der 15 mg Dextromethorphan-Hydrobromid enthält.
- Verfahren zur Herstellung von pharmazeutische Wirkstoffe enthaltenden Filmen
- Der Film gemäß Beispiel 22A wurde durch folgendes Verfahren hergestellt.
- 1. Natriumbenzoat und Süßstoffe werden dem Wasser zugegeben.
- 2. Johannisbrotkernmehl, Xanthangummi und Carrageenan werden vermischt.
- 3. Die Gummenmischung wird der Mischung aus Schritt 1 zugegeben und bis zur Lösung gerührt.
- 4. Der Wirkstoff wird mit Wasser oder Propylenglycol gemischt. Bei Bedarf wird erwärmt.
- 5. Die übrigen Bestandteile werden der Mischung von Schritt 4 beigegeben oder zu einem separaten Gemisch miteinander vermischt.
- 6. Die Mischungen von Schritt 4 und 5 werden der Mischung von Schritt 3 zugegeben. Es wird ausgebreitet und getrocknet unter Erhalt eines Films, der so zurechtgeschnitten wird, dass sich die gewünschte Dosis ergibt.
- Die Filme der Beispiele 22B bis 22E wurden durch folgendes Verfahren hergestellt.
- 1. Natriumbenzoat wird zu auf 50°C erwärmtem Wasser zugegeben. Es wird bis zur Lösung gerührt.
- 2. Getrennt voneinander werden Peg 1450, Titandioxid und Wirkstoff der Mischung von Schritt 1 zugegeben, und bei jeder Zugabe wird gerührt.
- 3. Johannisbrotkernmehl, Xanthangummi und Carrageenan werden vermischt.
- 4. Die Gummenmischung wird der Mischung von Schritt 2 zugegeben und bis zur Lösung gerührt.
- 5. Die übrigen Bestandteile werden gegebenenfalls unter Erwärmen zugegeben.
- 6. Die Mischungen der Schritte 4 und 5 werden vereinigt. Es wird ausgebreitet und getrocknet unter Erhalt eines Films, der so zurechtgeschnitten wird, dass sich die gewünschte Dosis ergibt.
- Die Filme der Beispiele 22F bis 22I wurden so hergestellt wie die Filme der Beispiele 20B bis 20E, mit dem Unterschied, dass der Wirkstoff unmittelbar vor dem Ausbreiten des Films dispergiert wurde.
Claims (20)
- Konsumierbarer Film, der im Mund der ihn einnehmenden Person haftet und sich darin auflöst, wobei der Film aus einer Schicht besteht, die wenigstens ein wasserlösliches Polymer und eine antimikrobiell wirksame Kombination wenigstens zweier etherischer Öle aus der Gruppe (i) 0,01–4 Gew.% Thymol, (ii) 0,01–4 Gew.% Methylsalicylat, (iii) 0,01–4 Gew.% Eucalyptol und (iv) 0,01–15 Gew.% Menthol enthält.
- Konsumierbarer Film nach Anspruch 1, enthaltend wenigstens drei der genannten etherischen Öle.
- Konsumierbarer Film nach Anspruch 1, enthaltend Thymol, Methylsalicylat, Eucalyptol und Menthol.
- Konsumierbarer Film nach Anspruch 3, weiter enthaltend ein Gluconsäuresalz.
- Konsumierbarer Film nach Anspruch 3, weiter enthaltend Kupfergluconat.
- Konsumierbarer Film nach Anspruch 1, wobei das wasserlösliche Polymer ausgewählt ist aus Pullulan, Hydroxypropylmethylcellulose, Hydroxyethylcellulose, Hydroxypropylcellulose, Polyvinylpyrrolidon, Carboxymethylcellulose, Polyvinylalcohol, Natriumalginat, Polyethylenglycol, Traganth, Guargummi, Akaziengummi, Gummiarabicum, Polyacrylsäure, Methylmethacrylat-Copolymer, Carboxyvinyl-Polymer, Amylose, hoch-amylosehaltiger Stärke, hydroxypropylierter hochamylosehaltiger Stärke, Dextrin, Pectin, Chitin, Chitosan, Levan, Elsinan, Collagen, Gelatine, Zein, Gluten, Sojaproteinauszug, Weizenproteinauszug, Casein und deren Mischungen.
- Konsumierbarer Film nach Anspruch 6, wobei das wasserlösliche Polymer Pullulan darstellt.
- Konsumierbarer Film nach Anspruch 7, enthaltend 40–80 Gew.% Pullulan, 0,01–4 Gew.% Thymol, 0,01–4 Gew.% Methylsalicylat, 0,01–4 Gew.% Eucalyptol und 0,01–15 Gew.% Menthol.
- Konsumierbarer Film nach Anspruch 6, weiter enthaltend: 0,01–5 Gew.% wengistens eines Stabilisators, 0,001–0,1 Gew.% wenigstens eines Farbstoffes, 0,1–8 Gew.% Wasser, 0,1–15 Gew.% wenigstens eines Süßstoffs, 0,1–15 Gew.% wenigstens eines Geschmacksstoffs, 0,1–4 Gew.% wenigstens eines Kühlmittels und 0,1–5 Gew.% wenigstens eines Tensids.
- Konsumierbarer Film nach Anspruch 9, worin der wenigstens eine Stabilisator ausgewählt ist aus Xanthangummi, Sojabohnenkernmehl und Canageenan, und wobei der wenigstens eine Süßstoff ausgewählt ist aus Saccharin, Aspartam und Acesulfam K.
- Konsumierbarer Film nach Anspruch 1, der im wesentlichen nicht an sich selbst anhaftet.
- Konsumierbarer Film nach Anspruch 1, der frei ist von Glycerin und Sorbit.
- Konsumierbarer Film nach Anspruch 1, der frei ist von Feuchthaltemitteln.
- Konsumierbarer Film nach Anspruch 1, worin die etherischen Öle wenigstens 10 Gew.% des Films ausmachen.
- Konsumierbarer Film nach Anspruch 14, wobei die etherische Öle wenigstens 15 Gew.% des Films ausmachen.
- Konsumierbarer Film nach Anspruch 1, weiterhin enthaltend Wasser in einer Menge von 3 Gew.% bis 8 Gew.%.
- Verwendung eines physiologisch kompatiblen Films gemäß den Ansprüchen 1–16 zur Herstellung einer pharmazeutischen Komposition zur Verabreichung und zur Förderung des Verweilens einer wirksamen Menge eines antimikrobiellen Mittels in der Mundhöhle.
- Verwendung nach Anspruch 17, wobei das antimikrobielle Mittel Menthol, Methylsalicylat, Eucalyptol und Thymol enthält.
- Verwendung nach den Ansprüchen 17 und 18, wobei die Menge an Pullulan im Film zwischen 40 Gew.% und 80 Gew.% beträgt.
- Verwendung nach den Ansprüchen 17 bis 19, wobei die Menge an antimikrobiellem Mittel im Film zwischen 5 Gew.% und 12 Gew.% beträgt.
