Die chromatographische Auftrennung erfordert sehr viel Lö¬ sungsmittel, dessen anschliessende Entfernung aufwändig ist. Für die destillative Auftrennung wiederum, ist nach¬ teilig, dass die zu trennenden isomeren THC-Verbindungen sich sowohl in ihren Siedepunkten als auch in ihren PoIa- ritäten sehr ähnlich sind, so dass an die Trennleistung der Geräte sehr hohe Anforderungen gestellt werden müssen. Dies bedingt einen verminderten Materialdurchsatz, wobei auch bei hoher Trennleistung in grosseren Mengen "Misch¬ fraktionen" anfallen, was die Ausbeute deutlich verrin- gert. Auch haben diese THC-Verbindungen vergleichsweise hohe Siedepunkte im Bereich von 1500C bei 0,02 Torr. Für eine Destillation sind deshalb besondere technische Ein¬ richtungen erforderlich. Bedingt durch die hohen Tempera¬ turen findet zudem eine teilweise Zersetzung und/oder Iso- merisierung der THC-Verbindungen statt, welche im Bereich von 18 Gew.-% im Labormassstab liegt. B) Herstellung von kristallinem Δ8-2-Napht. : 22,8 g des unter Abschnitt A) hergestelltem Rohprodukt (0,038 mol Δ8-THC) werden in 300 ml Essigester gelöst und unter Rühren mit 19,2 g (0,10 mol) 2-Naphtoylchlorid sowie 16 ml (0,12 mol) Triethylamin versetzt. Nach zwei Stunden Rühren bei Raumtemperatur (RT) gibt man 300 ml Wasser dazu und wäscht die separierte organische Phase mit 400 ml verdünnter NaHCO3-Lösung und schließlich mit 200 ml Wasser. Die organische Phase wird auf ein Drittel ihres Volumens eingeengt und unter Rühren mit 200 ml Methanol versetzt. Es setzt sofort eine Kristallisation ein. Nach 16 Stunden Rühren bei RT wird der Niederschlag abgesaugt und mit Methanol nachgeschlämmt. Nach der Filtration erhält man (-) -Δ8-6a, 10a-trans-Tetrahydrocannabinol- (2- naphthoyl)ester (15,1 g, 32,2 mmol; 43% bezogen auf Olivetol) als farbloses Kristallpulver.To a stirred mixture of 26.5 g (0.140 mol) of olivetol and 4.0 g (0.02 mol) of p-toluenesulfonic monohydrate in 650 g of toluene at 85 0 C internal temperature 24.2 g (0.154 mol) of trans -p-mentha (2, 8) -dien-1-ol added dropwise. Zu 15,43 g (33 mmol) von in Beispiel 1 hergestelltem (-) -Δ8-6a, 10a-trarJS-Tetrahydrocannabinol- (2-naphthoyl) - ester in 150 ml THF und 25 ml Methanol werden unter Argon 22 ml einer 2 N wässrigen NaOH-Lösung zugegeben. Nach einigen Stunden kräftigem Rühren werden weitere 25 ml 1 N NaOH zugegeben. Nach insgesamt 24 Stunden wird zur Aufarbeitung mit 2,0 g (33 mmol) Eisessig versetzt und die Reaktionsmischung wird eingeengt. Der wässrige Rückstand wird mit 140 ml Wasser aufgenommen und mit 100 ml und 50 ml MTBE (Methyl tert. -butylether) extrahiert, die organische Phase wird mit halbgesättigter NaHCθ3-Lösung (je 100 ml) 4-5 mal extrahiert und zum Schluss mit
gesättigter NaCl-Lösung gewaschen. Trocknen über MgSO4 und Evaporieren des Lösungsmittels ergeben ca. 10,7 g bernsteinfarbenes Öl (GC: > 95% Δ8-THC) .To 15.43 g (33 mmol) of (-) - Δ 8 -6a, 10a-tris-J-tetrahydrocannabinol (2-naphthoyl) ester prepared in Example 1 in 150 ml of THF and 25 ml of methanol are added under argon 22 ml of a 2N aqueous NaOH solution added. 10,7 g des in Beispiel 2 hergestellten (-) -Δ8-6a, 10a- fcrans-Tetrahydrocannabinol werden mit 2,90 g Zinkchlorid und 95 g kaltem 30% HCl in Essigester versetzt und unter kräftigem Rühren auf RT aufgewärmt. Nach 36 Stunden werden 100 ml Essigester und 200 ml Eiswasser hinzugegeben, die organische Phase mit 200 ml Wasser und 7 ml gesättigter Natriumbicarbonat-Lösung sowie 100 ml Wasser und gesättig¬ ter NaCl-Lösung gewaschen. Nach insgesamt einer Stunde wird für 20-25 Minuten auf Rückfluss erhitzt, danach abgekühlt, mit 100 ml MTBE versetzt und mit 100 ml gesättigter Natriumbicarbonat-Lösung und 2 ml Eisessig
ausgeschüttelt. Die organische Phase wird mit 100 ml gesättigter NaCl-Lösung gewaschen, über Na2SO4 getrocknet und eingeengt. Nach 2 Stunden Rühren werden weitere 0,7 ml 1 N NaOH zugegeben. Nach insgesamt 9 Stunden wird mit 20 ml MTBE und mit 20 ml halbgesättig¬ ter NaHCO3-Lösung sowie 0.18 ml Eisessig versetzt. Die
organische Phase wird 4-5 mal mit NaHCθ3-Lösung und einmal mit gesättigter NaCl-Lösung gewaschen, über Na2SO4 getrock¬ net und eingeengt.
